1.EFFECTS OF EXTRACT FROM LAPEMIS HARDWICKII ON RED BLOOD CELLS (RBCs) AND HEMOGLOBIN (Hb) IN ANEMIA MICE.

Effects of the extracts from Lapemis hardwickii on RBCs count and Hb content were studied in the both of mice model of which was made by acetylphenyhydrozine (APH) for hemolytic anemia and of by Chinese rubarb for Xue Xu (debility due to blood deficiency). The results showed that counts of red blood cell (RBC) and content of hemoglobin (Hb) of experimental group in hemolytic anemia model mice were higher significatly than that of control gooup in hemoly ticanemia mice. The content of Hb was increased in Xue Xu mice model,but no marked effect on the counts of RBCs. The study showed that the extract could have a better profective effect on red blood cells of hemolytic anemia mice.
2.Value of 18F-FDG PET/CT in evaluation of curative effect and progression-free survival on lymphoma

Objective To evaluate the value of 18F-fluorodeoxyglucose (FDG) PET/CT in evaluation of curative effect and progression-free survival (PFS) for lymphoma patients.Methods The data of 85 lymphoma patients were retrospectively analyzed.Before and after 4-8 cycles standardized chemotherapy,the patients were evaluated with 18F-FDG PET/CT.The two-year PFS rate was compared.The value of 18F-FDG PET/CT in evaluation of curative effect and PFS for lymphoma patients received chemoradiotherapy was analyzed.Results The non Hodgkin lymphoma (NHL) was the common type,and the common pathogenic sites were head and neck lymph nodes,mediastinum and retroperitoneum.The majority of patients were accompanied with spleen enlargement and local lesions with high metabolism.The complete remission (CR) rate of Hodgkin lymphoma (HL)patients in PET/CT negative group was significantly higher than that in positive group (86.4% vs.42.9%,P =0.038).The two-year PFS rates in PET/CT positive group and negative group were 42.9% and 81.8%,and the difference was statistically significant (x2 =7.70,P =0.006).Thirty-five NHL patients achieved CR,13 achieved partial remission (PR),and 8 achieved stable disease (SD) or disease progression (PD).The two-year PFS rates in the CR group,PR group,SD or PD group were 89.7%,65.3% and 19.4% respectively,and the difference was statistically significant (x2 =12.41,P =0.002).PET/CT imaging had a strong PFS predictive effect on T cell lymphoma (TCL) patients (x2 =13.85,P =0.001) and diffuse large B cell lymphoma (DLBCL)patients (x2=13.51,P =0.001),and had no predictive effect on follicular lymphoma (FL) patients (x2 =4.63,P =0.099).Conclusion 18 F-FDG PET/CT can evaluate the curative effect of lymphoma effectively and early predict prognosis,and has great guiding significance in choosing therapeutic schedule.
3.Effect of safflor yellew on cultured vascular endothelic cell

AIM To study toxicity of Safflor Yellew (SY) for vascular endothelic cell (VEC) and whether SY has antagonistic effect on platelet activating factor (PAF). METHODS Use light microscopical observation and crystal violet colorimetric method to study morphological and bioactivity change of HVEC and EVC 304 after incubated with PAF, SY, SY+PAF, Ginkolide,and Ginkolide+PAF. RESULTS Cultured with PAF 30 minutes, HVEC had histopathological changes: from fusiform, polygond squamous cell to ellipsoid, circular cell , not binding tightly, but separated by wide gap. As cultured with SY+PAF, cell's morphological changes just as same as PAF's group. And its bioactivity had tendency of decline. Either HVEC was incubated with SY (0.571 g?L -1 ) for 30 minutes or EVC 304 cultured with SY(1 143 g?L -1 ) for 4 hours, their morphology and bioactivity had changes. CONCLUSION SY do not inhibit PAF's injury effect. And high concentration SY has toxicity to cultured endothelic cell.
4.Expression in E.Coli and Antigenicity of HGV NS5 Protein

A 880 bp cDNA localized to the putative NS5 region of HGV genome was expressed in E.coli BL21(DE3). The cDNA fragment was inserted into a plasmid pGEX-5X-1,at the downstream of the DNA sequence encoding Schistosoma japonicum glutathione S- transferase(GST),in the same reading frame with the gene of GST. A 60KD GST-NS53 fusion protein was expressed at 37℃ in a form of inclusion bodies amounting to 30 percent of total host protein whereas at 20℃ mainly in a form of soluble protein . The fusion protein was extracted and purified to homologue. The purified GST-NS53 fusion protein could be specifically recognized with either the sera from the patient infected by HGV or the antisera directed against GST.
5.Efficacy observation of percutaneous vertebroplasty combined with zoledronic acid in treatment of spinal metastatic carcinoma

Objective To observe the efficacy of percutaneous vertebroplasty (PVP) combined with zoledronic acid in treatment of spinal metastatic carcinoma.Methods Fifty cases of spinal metastatic carcinoma were divided into control group and treatment group by random digits table method with 25 cases each.The control group was given zoledronic acid 4 mg,intervals of 28 d intravenous drip 1 time.The treatment group was given PVP besides zoledronic acid.The visual analog scale score,activity ability score and analgesic usage score after treatment for 3 months were observed and compared.The quality of life was evaluated and the occurrence of relapse of fracture was also recorded.Results After treatment for 3 months,the visual analog scale score,activity ability score and analgesic usage score were (2.00 ± 0.17),(2.12 ± 1.38),(1.47 ± 0.10) scores in treatment group,and (7.84 ± 0.18),(3.21 ± 0.13),(3.30 ± 0.09) scores in control group,and there were significant differences between two groups (P < 0.01 or < 0.05).The occurrence of relapse of fracture was 2 cases (8%,2/25) in treatment group and 9 cases (36%,9/25) in control group,and there was significant difference between two groups (P < 0.05).Conclusions PVP combined with zoledronic acid in treatment of spinal metastatic carcinoma has many advantages such as small incision,short surgery time and rapid relief from pain.It can obviously improve the quality of life,and it is worthy of spreading in the clinic.
7.Modification of mPEGylated novel bifunctional fusion protein GAD and its propenty

In order to solve the difficucties of renaturation and immunogenicity of new bifunctional fusion protein GAD,inclusion bodies of GAD were modified by PEG-maleimide.Conformational changes of the modified GAD were compared by circular dichroism and tryptophan fluorescence spectroscopy.The biological activity was verified by oral glucose tolerance test and lipid scavenging.The results showed that PEG-maleimide completed the specific-point modification of GAD,and improved its refolding efficiency.The secondary and tertiary structures of mPEGylated GAD were consistent with that of GAD.PEG-GAD has significant hypoglycemic and lipid-lowering effects (P <0.001) with longer half life in vivo and lower immunogenicity (P <0.01).This study provides effective strategies for the development of strongly hydrophobic peptide drugs.
8.Peripheral blood stem cell transplantation in the treatment of dilated cardiomyopathy: A 3-month follow-up in 42 cases

AIM: Recent studies showed that stem cells could replace injured cardiomyocyte and increase the number of functional cardiomyocytes. Researching the pertinent literature in China Journal Full-text Database (CJFD) published between 2005 and 2008 indicated that the researches on stem cell transplantation in the treatment of primary dilated cardiomyopathy were few. This study investigated the therapeutic efficacy and security of peripheral blood stem cell transplantation in coronary artery in treatment of dilated cardiomyopathy and its effects on left ventricular function. METHODS: Forty-two patients with dilated cardiomyopathy between December 2006 and September 2007 were enrolled at the Department of Cardiology of First People's Hospital of Yunnan, including twenty-eight males and fourteen females, averagely aged (56?3) years. Inclusive criteria: patients with less than 65 years, left ventricular enlargement, and left ventricular ejection fraction (LVEF) ≤ 45%, and without coronary artery disease after coronary arteriongraphy. Informed consents were obtained from patients. Patients were divided into stem cell transplantation group (n=15) and control group (n=27) on the basis of whether being treated by stem cell transplantation. Patients in the stem cell transplantation group were consecutively administered granulocyte colony-stimulating factor (G-CSF) by hypodermic injection to stimulate bone marrow stem cells themselves based on conventional treatment for five days. Peripheral blood stem cell suspension was disassociated on the 6th day, and the collected suspension was injected into left anterior descending branch over the wire saccule tube for autologous peripheral blood stem cell transplantation. Patients in the control group were administered by conventional treatment of dilated cardiomyopathy. Security and adverse reaction were observed during the mobilization, collection and returning injection of peripheral blood stem cell by coronary artery. Morphous, cardiac function and motion index of left ventricle wall were evaluated using ultrasoundcardiogram before and 3 months after transplantation. Survival rate and incidence rate of heart incidents were compared. 9. Effect of APS-II-2 on intra-amniotic lipopolysaccharide-induced alveolarization arrest in bronchopulmonary dysplasia model rats

Objective: To investigate the role of APS-II -2 (a kind of plant-derived natural drug) on amelioration of chorioamnionitis-induced alveolarization arrest and the underlying mechanism. Methods: Bronchopulmonary dysplasia (BPD) model was constructed by intra-amniotic injection of lipopolysaccharide (LPS) in SD rats (E16.5). The SD rats were randomly divided into control group (Saline group), LPS model group (LPS+Saline group) and APS-II -2 administration group (LPS+APS-II-2 group), Then neonatal rats in LPS+APS-II-2 group were given an intraperitoneal injection with APS-II-2 (50 mg/ kg) for 3 consecutive days after birth, whereas rats in LPS+Saline group and Saline group were administrated with an equal amount of normal saline. To examine pathologic change of pulmonary in neonatal rats, hematoxylin-eosin (H-E) staining was performed at postnatal day1 and 3. Then bone marrowderived macrophages (BMDMs) from SD rats were detected by the technology of RNA-sequence to research the immunomodulation of APS-II -2. Results: APS-II -2 administration group had drastically higher terminal air spaces (P=0.033 at postnatal day1) and secondary septa counts at postnatal day1 and 3, respectively (P=0.002, P=0.026) than LPS-induced model group, while mean linear intercept was the opposite situation at postnatal day1 and 3, respectively (P=0.006, P=0.004). The detection of RNA-sequence indicated that APS-II -2 suppressed the expression of inflammatory cytokines such as Tlr3, Tlr7 and Tlr8 in BMDMs. Meanwhile, it also promoted some pleiotropic cytokines with anti-inflammatory effects such as Alox15 and Cd74. Conclusion: Administration of APS-II -2 could improve the pathology of BPD, thereby supporting the ethnopharmacological uses of the plant. This effect may be directly caused by modulatory effects of APS-II -2 on inflammation.

This article was to elucidate that the needling depth is closely related to the meridian qi, disease location, disease nature and needled area based on the records of needling depth in Nei Jing (Canon of Internal Medicine). Moreover, different depths will produce different therapeutic efficacies;meanwhile, improper depth may lead to grave consequences.
