1.Role of AP-1 Activation in Regulation of Angiotensinogen Gene Expression Induced by AngⅡ

Angiotensin Ⅱ(AngⅡ)can induce the expression of its precursor,angiotensinogen,in vascular smooth muscle cells(VSMC),which is related with increased activating protein-1(AP-1)binding to its cis-element located in the angiotensinogen gene promoter.In the present study,cycloheximide(CHX)was used as an inhibitor to interrupt c-Jun,the role of AP-1 in AngⅡ-induced its precursor gene activation was investigated by DNA-protein interaction and immunoblotting.The results showed that the level of c-Jun,the component of transcription factor AP-1,was significantly increased in the nucleus of VSMC after AngⅡ treatment.The majority of c-Jun was found in the nucleus but hardly detected in the cytoplasm by immunocytochemistry staining.Immunoprecipitation assays confirmed that AngⅡ could induce serine phosphorylation of c-Jun.EMSA results indicated that the level of phosphorylated of c-Jun had a positive correlation with AP-1 binding activity to cis-acting element of angiotensinogen gene and transcription activation of angiotensinogen.CHX inhibited AngⅡ-induced binding activity of AP-1 by reducing the phosphorylation of c-Jun,though it did not affect the expression of c-Jun.These findings suggest that the AP-1 phosphorylation induced by AngⅡ is one of the important mechanisms whereby AngⅡ regulates its precursor gene expression in feedback manner.It is found that CHX is an inhibitor to phosphorylation of c-Jun.
2.Application of hysteroscopy in assisted reproduction

Objective To explore the application of hysteroscopy in assisted reproduction. Methods A retrospective analysis was made on results of routine hysteroscopy in 150 cases of infertile women scheduled for assisted reproduction.Results Among the 150 cases,abnormal hysteroscopic findings were identified in 47 cases(31%),including endometrial polyps in 18 cases(12%),endometrial hyperplasia in 17 cases(11%),uterine deformity in 7 cases(5%),uterine adhesion and stenosis in 3 cases(2%),endometrial tuberculosis in 1 case(0.7%),and submucous myoma or adenomyoma in 1 case(0.7%).To think of hysteroscopic findings as the standard,the sensitivity,specificity,positive predicted rate,negative predicted rate,and accuracy were 66%(31/47),98%(101/103),94%(31/33),86%(101/117),and 88%(132/150) for transvaginal ultrasonography,respectively,and were 26%(12/47),100%(95/95),100%(12/12),73%(95/130),and 75%(107/142) for uterophotography,respectively.Conclusions Hysteroscopy is a simple and reliable way to explore the uterine cavity.It may be selected as a routine examination before assisted reproduction.

5.Quality Evaluation of Anticoagulant Therapy in Patients Who Taking Warfarin in Our Hospital

OBJECTIVE:To evaluate the quality of anticoagulant therapy in patients who took warfarin and provide data sup-porting for strengthening the management of these patients. METHODS:A retrospective analysis of related clinical data of 214 pa-tients who meet the inclusion criteria was performed. RESULTS:The average time for patients took warfarin was (321.64 ± 189.50)d,average times for tested INR was(12.01±7.03)times in clinic;when anticoagulant therapy,INR<2.0 accounted for 51.96%,2.0≤INR≤3.0 accounted for 39.13%,and INR>3.0 accounted for 8.91%;patients with target INR≤2.0 accounted for 45.33%,target INR=2.5 accounted for 38.32%,and target INR≥3.0 accounted for 0.93%;the average TTR was (50.80 ± 22.32)%;and there was no statistical significance in the TTR of different ages and diseases(P>0.05). CONCLUSIONS:The anti-coagulant therapy in some patients who took warfarin shows poor quality,it needs strengthening the quality management to make it safe and effective.
6.Comparison of the clinical effect of phacoemulsification and middle incision extracapsular cataract extraction

To compare the clinical effect of phacoemulsification and middle incision extracapsular cataract extraction ( Ml-ECCE) . METHODS: One hundred and eighty - five eyes of phacoemulsification (137 cases) and 185 eyes of 139 cases for Ml-ECCE from January 2011 to May 2013 were involved in this study. And the ratio posterior capsular rupture during surgery, visual acuity, corneal edema, corneal astigmatism and intraocular pressure post operation were followed up.RESULTS: On 1d after surgery, uncorrected visual acuity in the group of Ml-ECCE was better than that of phacoemulsification group, while from 3d; 1 and 3mo after surgery, no significant difference was found from the above two groups. On 1d postoperation, corneal edema ratio in phacoemulsification group ( 45 eyes ) was higher than that in Ml-ECCE group(20 eyes) ( X2=11. 665, P=0. 0006 ) . No significant difference was found for the ratio of posterior capsule rupture during surgery in these two surgical technique groups(X2=0. 094,P=0. 759). On 1wk;1 and 3mo after surgery, significant difference was found for the average of surgical induced corneal astigmatism between two groups (u=6. 661, 6. 880, 4. 187, P = 0. 00, respectively ). During following up, no significant difference was found for the intraocular pressure between two groups ( u=1. 858, 0. 963, 0. 471, 1. 349, 1. 388; P= 0. 063, 0. 335, 0. 638, 0. 177, 0. 165). lntraocular pressure on 1d postoperation in phacoemulsification and Ml-ECCE groups was higher than before operation ( u = 19. 86, 19. 39, P = 0. 00, respectively). And on 1wk; 1 and 3mo postoperation, intraocular pressure in the operated eyes in both groups was lower than before operation for 2~3mmHg.CONCLUSlON: Although phacoemulsification and Ml-ECCE could both get good visual rehabilitation, with similar visual outcome, no significant effect for intraocular pressure, and no severe complications, the latter one owns the advantage that easier maneuver, quicker recovery, and cheaper instruments needed, which is suitable for the hard nuclei cataract in local hospitals. Ml-ECCE is a safe, effective and easy manipulation for local hospital for large batch of cataract surgeries.
7.Detection of microvesicle miRNA expression in ALL subtypes and analysis of their functional roles.

Microvesicles (MVs) are the heterogeneous mixtures of vesicles. MVs released by leukemia cells constitute an important part of the leukemia microenvironment. MVs might act as important reservoirs of microRNAs (miRNAs). It is worth evaluating whether MVs possess some unique miRNA contents that are valuable in understanding the pathogenesis. In this study, we investigated the miRNA expression patterns of Nalm-6-derived MVs, Jurkat-derived MVs and normal cell-derived MVs using miRNA microarrays. The potential target genes regulated by differentially expressed miRNAs were also predicted and analyzed. Results demonstrated that 182 miRNAs and 166 miRNAs were differentially expressed in Nalm-6-MVs and Jurkat-MVs, respectively. Many oncogenes, tumor suppressors and signal pathway genes were targeted by these aberrantly expressed miRNAs, which might contribute to the development of B-ALL or T-ALL. Our findings expanded the potential diagnostic markers of ALL and provided useful information for ALL pathogenesis.
9.Fluorescence enhancement of flavoxate hydrochloride in alkali solution and its application in pharmaceutical analysis.

Fluorescence enhancement reaction of flavoxate hydrochloride (FX) in strong alkali solution was studied, the mechanism of the reaction was investigated, and a novel fluorimetric method for analysis of FX in drug sample was established. FX has no intrinsic fluorescence, but it can slowly produce fluorescence in strong alkali solution. Heating can promote the fluorescence enhancement reaction. In 3D fluorescence spectra of the decomposition product of FX, two fluorescence peaks, located respectively at excitation wavelengths λex/ emission wavelength λem =223/410 nm, and 302/410 nm, were observed. Using quinine sulfate as a reference, fluorescence quantum yield of the decomposition product was measured to be 0.50. The structural characteriza- tion and spectral analysis of the decomposition product reveal that ester bond hydrolysis reaction of FX is firstly occurred during heating process, forming 3-methylflavone-8-carboxylic acid (MFA), then a cleavage reaction of the γ-pyrone ring of MFA occurred, producing α, β-unsaturated ketone. This product includes adjacent hydroxyl benzoic acid group in its molecule, which can form intramolecular hydrogen bond under alkaline condition, so that increase the conjugate degree and enhance the rigidity of the molecule, and thereby cause fluorescence enhancement. Based on this fluorescence enhancement reaction, a fluorimetric method was proposed for the determination of FX. A linear calibration curve covered the concentration range 0.020 3-0.487 µg · mL. The regression equation was I(F) = 23.9 + 5357.3 c, with correlation coefficient r = 0.999 7 (n = 8), detection limit D = 1.1 ng · mL(-1). The method was applied to the analysis of FX tablets, with a spiked recovery rate of 100.2%. The reliability of the method was verified by a UV-spectrophotometric method.

10. Study on genetic relationship of Swertia mileensis and its related species based on infrared spectroscopy combined with chemometrics

Objective: Genetic reationships betwen Swertia mileensis and its relatives have been researched using Fourier transform infrared (FTIR) spectroscopy combined with chemometrics methods in order to provide a theoretical basis for the development and utilization of medicinal plant resources of genus Swertia. Methods: Infrared spectrum information of Swertia mileensis, Swertia cincta, Halenia elliptica, Swertiaion nervosa, Swertia punicea, and Swertia binchuanennsis was collected and used in this study. Original infrared spectra data were pretreated by these methods including automatic baseline correction, automatic smoothing, ordinate normalization, multiplicative scatter correction and second derivative, and analyzed by principal component analysis (PCA), partial least squares discriminant analysis (PLS-DA) and hierarchical cluster analysis (HCA). Results: Absorption area of relationship between Swertia mileensis and its relatives ranged from 900-400 cm-1, 1 310-900 cm-1, 1 500-1 310 cm-1, 1 800-1 500 cm-1, 2 800-3 000 cm-1, and 3 000-3 500 cm-1. Absorption peaks of the second derivative of fingerprint region in 400 to 1 000 cm-1 were distinct, and the absorption peaks as well as peak numbers, intensities and patterns among species were quite different. Analysis of preprocessed IR data showed that PCA analysis of six Swertia species was superior to PLS-DA analysis. The results of HCA analysis showed that Swertia mileensis was closely related to Swertia cincta and Swertia nervosa. Conclusion: FTIR spectroscopy combined with chemometrics method could discriminate different species of genus Swertia and display the closely genetic relationship of Swertia mileensis and its relatives, furthermore, this research would provide a fast and effective method for studying genetic relationship of genus Swertia.
