1.Modification of GABAA receptor subunit mRNA content in audiogenic seizure rat cortex and hippocampus following tolerance to flurazepam

Objective: To investigate the modification of GABAA receptor subunits in audiogenic seizure rat cortex and hippocampus when rendered tolerant to flurazepam. Methods: Rats were administrated with flurazepam for two weeks, which resulted in tolerance to flurazepam, in the absence of behavioral signs of withdrawal. GABAA receptor subunits α1, α3, α5, γ2L and γ2S were assayed using quantitative competitive RT-PCR in rat frontparietal motor(FrPaM) and hippocampus. Results: In FrPaM, the contents of mRNA encoding for α1, α3, γ2L and γ2S were all significantly decreased (24%,17%,35% and 45% respectively),whereas that of α5 was significantly increased (33%) compared with the control. In hippocampus, α1, γ2L, and γ2S mRNA contents were significantly decreased (33%, 35%and 27% respectively). Conclusion: The accomodated change in GABAA receptor subunits α1, α3, α5,γ2L and γ2S in FrPaM and hippocampus may be associated with the mechanism of flurazepam tolerance in audiogenic seizure rats.
2.Modification of brain GABA_A receptor subunit ?1,?3,?5,?2L and ?2S expression in audiogenic seizure rat following tolerance to clobazam

AIM To investigate the modification of GABA A receptor subunits in audiogenic seizure rat cortex and hippocampus when rendered tolerant to clobazam. METHODS Rats were administrated with clobazam for two weeks, resulting in tolerance to clobazam. GABA A receptor subunit ?1, ?3, ?5, ?2L and ?2S were assayed using quantitative competitive RT PCR in rat FrPaM and hippocampus. RESULTS In FrPaM,the content of mRNA encoding for ?1,?2L and ?2S were all significantly decreased (27%,43% and 37% respectively),whereas that of ?5 was significantly increased (73%) compared with the control. In hippocampus, ?1, ?2L, and ?2S mRNA content was significantly decreased (28%, 32% and 31% respectively). CONCLUSION The accomodated change in GABA A receptor subunit ?1, ?5,?2L and ?2S in FrPaM and hippocampus may be associated with the mechanism for tolerance to clobazam in audiogenic seizure rat.
3.Effect of rosiglitazone combined with all-trans retinoic acid on anti-angiogenesis of transplanted gastric cancer in nude mice

Objective To investigate the effect of rosiglitazone (ROS),a peroxisome poliferator- activated receptor (PPAR)?ligand,combined with all-trans retinoic acid (ATRA) on anti-angiogenesis of transplanted gastric cancer in nude mice,and to explore the mechanism of anti-angiogenesis prelimina- rily.Methods The model of xenograft tumor in nude mice were established by inoculating human gastric cancer cells line MGC803 (lower differentiated) into the back of nude mice subcutaneously.The cancer- bearing nude mice were divided randomly into 5 groups:group 1 (n=6) with no treatment;ROS treat- ment (group 2,n=6),ROS combined ATRA treatment including:low dose treatment (group 3,n= 6),moderate dose treatment (group 4,n=6) and high dose treatment (group 5,n=6).After treated for forty days,the volume change of tumor and tumor inhibition rates were observed.The expression of CD34,vascular endothelial growth factor (VEGF) in grafts were detected by immunohistochemical and calculated the difference of MVD.The mRNA expression levels of VEGF,HIF-l?were detected by RT- PCR assay accordingly.Results①The volume of tumor was significantly decreased in ROS treatment group compared with group 1 (P<0.01).The tumor inhibition rates of group 2 were similar to group 3 (P>0.05).With the increasing of the dose of ROS the tumor inhibition rates were increased.They were dose-dependent in specified dose-range.②ROS could inhibit angiogenesis of xenograft tumor and depress expression of mRNA of VEGF and HIF-l?.When ROS combined with ATRA,the increasing of dose of ROS,inhibiting angiogenesis of tumor and depressing expression of mRNA of VEGF and HIF-l?were found (P<0.05).Conclusion ROS (25 mg?kg?2 d~(-1)) can inhibit the growth of tumor,and ROS combined with ATRA can further inhibit the growth of tumor,which may be through the path of PTEN by inhibiting the angiogenesis of tumor
4.Comparison of the occurrence rates of cervical nerve entrapment at cervical intervertebral foramen and spinal nervous sulcus

BACKGROUND: Nerve root could be directly or indirectly entrapped due to lateral backward prolapse of nucleus pulposus of intervertebral disc, as well as parenchymal inflammatory tumefaction of the soft tissues in nerve root tube. But the factors and their severity related to cervical nerve entrapment at cervical intervertebral foramen and spinal nervous sulcus still need further studies.OBJECTIVE: To investigate the relationship between the occurrence rate of cervical nerve entrapment at cervical intervertebral foramen and spinal nervous sulcus and the severity of cervical syndrome.DESIGN: A single sample study.SETTING: Department of Anatomy, Department of Geriatrics of the Affiliated Hospital, and Department of Teaching Affairs, Chengde Medical College.PARTICIPANTS: The research was conducted in the Department of Anatomy, Chengde Medical College. Totally 120 sides of 60 cadavers(28 males and 32 females) were provided by the Department of Anatomy.METHODS: The outside diameters of cervical intervertebral foramen and spinal nerve root, as well as the width of external opening of spinal nerve sulcus and transverse diameter width of the anterior branch of intra-spinal nerve were measured, and the data were dealt with statistically.MAIN OUTCOME MEASURES: The ratio of transversal diameter of the forepart of cervical nerves to the width of spinal nerve sulcus; the ratio of the peripheral diameter of cervical nerve roots to the size of intervertebral foramen; the occurrence rate where the ratio was ≥ 1.RESULTS: The ratio between cervical nerve root and intervertebral foramen was significantly larger than that between spinal nerve root and spinal nerve sulcus( t = 2.66, P < 0.01 ). The occurrence rate of cervical nerve entrapment at the intervertebral foramen(24.6% ) was higher than that in spinal nerve sulcus(6.3% ), and the difference had a considerable statistical significance(x2 =6.95, P < 0.01).CONCLUSION: It is suggested that the spinal nerve may be entrapped in the cervical intervertebral foramen and the spinal nervous sulcus, and the spinal nerve lesions in the cervical intervertebral foramen was more serious than in the spinal nervous sulcus. Therefore, in clinic, when diagnosing and treating cervical spondylosis, while nerve entrapment at the intervertebral foramen is fully considered, the possibility of nerve entrapment at the spinal nervous sulcus should not be neglected.
5.Effect of preoperative transcatheter arterial chemoembolization on Inhibitor of apoptosis protein-Survivin and Survivin mRNA of hepatocellular carcinoma.

Objective To study the effect of transcatheter arterial chemoembolizafion (TACE) on inhibitor apoptosis protein-Survivin, Survivin mRNA and proliferating cell nuclear antigen (PCNA). Methods Group A of hepatocelluar carcinoma with preoperative TACE (n = 26), group B of hepatocelluar carcinoma without any interven-tional therapy (n = 30) ,group C of Normal liver (n = 16) were divided. The expression of Survivin mRNA and the expression of Survivin and PCNA were detected. Result ①The positive Survivin mRNA expression in group A and the grey difference were lowere than that in group B (P < 0.05) ; ②The positive Survivin expression and the grey difference in group A were lower than that in group B (P < 0.05) ; ③The proliferating index (PI) of carcinoma tis-sure in group B was higher in group A and group C (P < 0.05 for each). Conclusion There is a high positive ex-pression of Survivin mRNA and Survivin, which may be decreased by TACE and then the proliferation of hepatocellu-lar carcinoma cell is surpressed.
6.Correlation analysis of postoperative prophylactic antibiotics and surgical site infection rates after breast cancer surgery

Objective To explore the relationship between the postoperative prophylactic antibiotics and surgical site infection rates after breast cancer surgery.Methods Retrospective review of patients with breast surgical procedures between March 2007 to March 2016 were included,267 cases only preoperative antibiotic prophylaxis as control group,and 117 cases of preoperative and postoperative prophylactic antibiotics as research group.The surgical site infection( SSI) rates were compared.Results There was no statistically significant association between research groups and control group of SSI rates for both a 30-day postoperative period and beyond.Further,There was also no statistically significant association between research groups with drains and control group with drains of SSI rates for both a 30-day postoperative period and beyond.Conclusion There is no significant relationship between postoperative prophylactic antibiotics and surgical site infection rates after breast cancer surgery.
7.Effect of Membrane-coating on Dissolution and Stability of Sirolimus Dropping Pills

Objective:To study the effect of membrane-coating on dissolution and stability of sirolimus ( SRL) dropping pills to prove the effect and rationality of the coating process. Methods:Opadry was used as the coating material for SRL dropping pills. Com-pared with those of uncoated SRL dropping pills, the dissolution and stability of membrane-coating SRL dropping pills were studied in vitro. Results:Compared with that of uncoated SRL dropping pills, the drug release amount of membrane-coating SRL dropping pills was lower (P<0. 05), however, the stable release rate showed no significant difference (P>0. 05). After the membrane-coating, the stability of SRL dropping pills was notable enhanced under high humidity (75% ± 5%) and strong light (4500lx ± 500lx) conditions, however, the stability showed no improved under high temperature(40℃ ± 2℃) condition. Conclusion: The membrane-coating can enhance the stability of SRL dropping pills without significant effect on drug release in vitro.

Objective To describe the burden of caregivers of children with asthma,and to provide reference for the development of family care support program.Methods Qualitative research was used to carry out semi-structured interviews with 13 caregivers of children with asthma in the pediatric outpatient of the First Affiliated Hospital of Guangzhou Medical University.The data collected were analyzed with Colaizzi 7-step analysis method.Results The carcgiver burden was classified into 4 themes,including time and energy exhaustion,heavy economic burden,insufficient knowledge of care and negative emotions.Conclusions The burden of caregivers of children with asthma has a relatively great impact on the daily work and life,even the physical and mental health.Care workers should be concerned about the status of caregiver burden and provide targeted interventions,which could help caregiver adapt to their roles,maintain and promote the health of children patients and their families.
10.Correlation of quantitative examination of urinary sediment bacteria and urine culture

Objective To evaluate quantitative examination of urinary sediment bacteria as a basic feasibility of screening indicators for urinary tract infection .Methods 191 outpatients and inpatient specimens were gathered firstly to implement a urine culture ,and then the rest of the urine were used for sediment bacteria quantitative testing .Meanwhile ,bacterial culture was conducted as the standard .According to the results of bacterial culture ,receiver operating characteristic(ROC) was drawn ,the threshold values of leukocyte and bacteria counts for diagnosis of urinary tract infection were found out and its sensitivity ,specificity ,positive / negative predictive value ,false positive/false negative rate and accuracy were calculated .Results The positive rate of urine culture was 39 .7% ,and the most common pathogen was Escherichia coli .The threshold value of bacteria and leukocyte counts for diagnosis of urinary tract infection was 1 024 .5/μL and 135 .8/μL respectively .When combined leukocyte and bacteria counts for urinary tract infection ,the optimum sensitivity was 62 .5% ,specificity was 98 .1% ,positive predictive value was 95 .7% ,negative predictive value was 79 .6% ,false positive rate was 1 .9% ,false negative rate was 37 .5% ,and accuracy was 83 .8% .Conclusion With UF‐1000i urinary sediment analyzer ,the combined determination of leukocyte and bacteria counts can remove the great mass of negative results ,Especially the results of bacterial culture positive predictability is higher ,but still can not replace of quantitative bacterial culture .
