3. Screening for clear cell renal cell carcinoma metastasis-associated methylations by genome subtractive hybridization

Objective: To screen for methylated biomarkers for the early diagnosis and prediction of clear cell renal cell ancer (ccRCC) metastasis. Methods: Highly and lowly metastatic ccRCC cell lines,which were established using fresh surgical specimens in our laboratory,were used in this study. Genomic DNA was extracted after pathological identification. Methylated genomic fragments were enriched by 3 cycles of Not I genomic DNA subtractive hybridization and were linked to vectors,which was then used for transformation, followed by α selection. Forty positive clones were randomly selected for DNA sequencing. Bioinformatics analysis was used to identify methylation of CpG islands and to predict the function of unknown genes. Results: DNA sequencing revealed 27 independent clones with different methylations between the highly and lowly metastatic ccRCC. Five of the 27 clones contained CpG islands,and 2 of the 5 fragments contained CpG islands in the promoter regions of genes MYADM and LOC646024. MYADM was associated with maturation of hematopoietic cells and regeneration of stem cells. LOC646024 shared 85% homology with UL16 binding protein 1; the latter was related to tumor killing function of NK cells. Conclusion: Novel methylated sequences have been discovered from Chinese ccRCC cells with different potentials for metastasis. Methylation of 2 candidate genes,MYADM and LOC646024,is indicated to be involved in ccRCC metastasis. Our findings are valuable for the biomarker exploration to predict metastasis as well as molecular epidemiological research.
5.Clinical research of canalicular intubation combined external dacryocystorhinostomy for chronic dacryocystitis and upper lacrimal duct stenosis

AlM: To evaluate the clinical efficacy of canalicular intubation combined with external dacryocystorhinostomy ( ext-DCR ) for treatment of chronic dacryocystitis and upper lacrimal duct stenosis.
METHODS:Thirty-three patients (33 eyes) with chronic dacryocystitis and upper lacrimal duct stenosis who underwent canalicular intubation combined ext-DCR were retrospective analyzed. The silicon tube was indwelt for 6mo. All cases were re-examined 1wk;1, 3, 6, 9mo post-operation, flow of tears, pus excretion and lacrimal duct clearance were observed.
RESULTS: Clear lacrimal duct ratio was 100% in all cases during 1wk~6mo post-operation; Silicon tube was removed 6mo post-operation, 3mo after tube removal, rechecking reported 32 cases with clear lacrimal duct (97%) and 1 case with resistant duct (3%). Two cases ( 6%) with minor lacerations, no other complications were observed.
CONCLUSlON: Canalicular intubation combined ext-DCR is an effective treatment for chronic dacryocystitis and upper lacrimal duct stenosis.
7.Nasal cavity volume and nasopharyngeal cavity volume in adults measured by acoustic rhinometry

Objective:To investigate the range of normal value of total nasal cavity volume(NV)and total na-sopharyngeal cavity volume (NPV)of healthy adults. Method: Six hundred and fifty-nine healthy adults andeighty-two adults of chronic rhinitis were measured with acoustic rhinometry. Result:The range of NV was29. 922~37. 481 cm3 and NPV was 29. 369~44. 159 cm3. Comparing healthy adults with adults of chronic rhini-tis,there was a significant difference in NV. Conclusion:Acoustic rhinometry suited for objective assessment ofthe nasal airways in adults. It was demonstrated that these data could provide available information for the studyof nasal physiology and pathophysiology,as well as for the diagnosis and judgement of therapeutic effectiveness ofnasal diseases.
9.Summarization of the pathogen and therapy of secondary glaucoma after vitrectomy

Secondary glaucoma is a kind of complications after vitrectomy, its etiologies are various and complex. Ineffective therapies might cause irreversible damage on optic nerves and visual field defect, even the loss in visual function. Nowadays, this project has been paid great attention by various researches both in China and abroad. Both the pathogens and therapies of secondary glaucoma after vitrectomy are analyzed as follows.
10.Analysis of CT features of pulmonary sarcoidosis

Objective To investigate the CT features of pulmonary sarcoidosis.Methods Ninety patients with histologically proved pulmonary sarcoidosis were retrospectively studied by using CT scans and clinical recording.Results The main CT findings of pulmonary sarcoidosis were nodules which were seen in 69 cases(76.7%),and the nodules mostly distributed around the bronchovascular bundle(n=37, 41.1%).Other abnormalities included consolidation(n=31,34.4%),ground-grass(n=39,43.3 %), thickening of bronchovascular bundle(n=30,33.3%),interlobular septal lines(n=58,64.4%), fibrosis(n=17,18.9%)including bronchial distortion(n=8,8.9%),linear shadow(n=5,5.6%), and honeycombing shadow(n=4,4.4%),air-trapping(n=3,5.3%),bronchial straitness(n=8, 8.9%),pleural thickening(n=42,46.7%),and hilar and mediastinal adenopathy(n=76,84.4%). Two or more abnormal findings co-existed in 83 cases.The pulmonary lesions co-existed with hilar and mediastinal adenopathy in 76 cases.The nodules(n=25),consolidation(n=9),ground-grass(n=11), thickening of bronehovascular bundle(n=10)were improved after therapy.Ten cases of the interlobular septal(10/22),0 of bronchial distortion(0/4),1 case of diffuse linear(1/3),and 0 case of honeycombing(0/2)were improved.Conclusion CT manifestations of pulmonary sarcoidosis are varied, but has some specific radiographic features.A correct diagnosis can be made.combined with hilar and mediastinal adenopathy.
