1.Clinical efficacy and safety of glaucoma filtration surgery combined with the application of 5-fluorouracil

Abstract?AlM: To observe the clinical efficacy and safety of glaucoma filtration surgery combined with the application of 5 - fluorouracil for clinical treatment to offer the reference.?METHODS:A total of 91 cases of patients with glaucoma treated in our hospital were randomly divided into three groups, 30 cases of group A were given mitomycin in surgery, 31 cases of group B were given 5-fluorouracil, 30 cases of group C were given cyclosporine. They were given 12 -month-follow-up after operation, the operation success rate were recorded, the incidence of 1 -year-adverse reactions, visual acuity, intraocular pressure, low intraocular pressure, shallow anterior chamber, corneal damage index were recorded.?RESULTS: lntraocular pressure ofgroups A, B, C were respectively (20.5±8.6)mmHg,(15.2±5.5)mmHg,(20.0± 8. 1 ) mmHg, by the analysis of variance, group B was significantly lower than other two groups,P<0. 05;patients of groups A, B, C whose visual acuity increased ratios after 1y were respectively 22. 5%, 38. 1%,and 23. 1% , by the Chi. square test, the increased ratio of group B was higher than other two groups , P<0. 05; The operation success rates after 1y of operation of groups A, B, C were respectively 67. 5%,88. 1%,71. 8%,by the Chi square test, the increased ratio of group B was higher than other two groups, P<0. 05;The general incidence of adverse reaction were respectively 22. 5%,9. 5%,20. 5%,by the Chi square test , the increased ratio of group B was lower than other two groups, P<0. 05.?CONCLUSlON:Glaucoma filtration surgery combined with the application of 5-fluorouracil can improve the operation success rate, decrease the occurrence rate of adverse reaction, and safety can be improved.
2.Effect of high glucose on cultured retinal Müller cell in vitro

Background Retinal Müller cells can offer nutrient and maintain the normal structure of retina.Researches showed that the abnormality of Müiller cells leads to retinal vascular disease.To explore the effect of high glaucoma on retinal Müller cells is of a very important significance for the study on diabetic retinopathy (DR).Objective This study was to investigate the effects of different concentrations of glucose on retinal Müller cells in vitro.Methods Retinal tissue was isolated from 1 10-day-oM clean SD rat.Mtiller cells were cultured by explant culture method and passaged in DMEM containing 20% fetal bovine serum.The third generation of cells were obtained and identified using glial fibrillary acidic protein (GFAP) staning.Then,5.5,30.0 and 40.0 mmol/L glucose were added into the culture medium for 4 days respectively.The proliferation (A570) of Müller cells was detected by MTT,and apoptosis rate of Müller cells was calculated by flow cytometer to evaluate the effects of 5.5,30.0 and 40.0 mmol/L glucose to cell vitality.Results Cultured and passaged cells grew well with the spindle shape.The positive reactive cells were >95% for GFAP.The A570 value of Müller cells was 0.24±0.01,0.21±0.03 and 0.20±0.02 in 5.5,30.0 and 40.0 mmol/L glucose group respectively,showing a significant difference among the three groups(F=6.755,P<0.05).Compared with 5.5 mmol/L glucose group,As70 values were significantly lower in 30.0and 40.0 mmol/L glucose group (q =0.645,0.486,P < 0.05).Apoptosis rates of Miiller cells were (26.40 ±0.25)%,(30.19±0.16)% and (36.23±0.19)% in 5.5,30.0 and 40.0mmol/L glucose groups,with a significant difference among them (F =294.530,P<0.05),and those in 30.0 and 40.0 mmol/L glucose groups were significantly reduced in comparison with 40.0 mmol/L glucose group (q =0.754,0.484,P < 0.05).Conclusions High concentration of glucose inhibits the viability and promote the apoptosis of retinal Müller cells at a concentrationdependent manner.

4.Effect of dexmedetomidine on noise-induced hearing loss in guinea pigs

Objective To evaluate the effect of dexmedetomidine on noise-induced hearing loss in guinea pigs.Methods Twenty-four adult male guinea pigs,aged 3 months,weighing 400-500 g,were randomly divided into 3 groups (n =8 each) using a random number table:dexmedetomdine group (group D),noise-induced hearing loss group (group N) and dexmedetomidine + noise-induced hearing loss group (group DN).A loading dose of dexmedetomidine 5 μg/kg was infused over 5 min,followed by 135 min of infusion at a rate of 10 μg· kg-1 · h-1.The equal volume of normal saline was infused in group N.Groups N and DN were exposed to noise of 4 kHz center frequency and 118-122 dB SPL for 120 min starting from 20 min of administration.Mean arterial pressure (MAP) and cochlear blood flow (COBF) were recorded before administration and every 5 min during drug administration.The changing rate of COBF was calculated.Arterial blood samples were collected for determination of plasma concentration of noradrenaline (NE) by high performance liquid chromatography at 20 and 140 min of administration.Auditory brainstem response (ABR) threshold was recorded before administration and at 1 and 72 h and 10 days after the end of administration.Results Compared with group N,MAP was significantly decreased,the changing rate of COBF was increased at 5-10 min and 30-140 min of administration,ABR threshold was decreased at 1 and 72 h and 10 days after the end of administration,and the plasma concentration of NE was decreased at 140 min of administration in D + N group (P < 0.05).Conclusion Dexmedetomidine can attenuate noise-induced hearing loss in guinea pigs possibly through inhibiting activation of sympathetic nerves and increasing COBF.
5.Effect of propofol on noise-induced hearing loss in guinea pigs

Objective To evaluate the effect of propofol on noise-induced hearing loss in guinea pigs.Methods Forty-eight healthy adult male guinea pigs,aged 3 months,weighing 400-500 g,were randomly divided into 4 groups (n =12 each) using a random number table:control grotup (C group),propofol group (P group),noise-induced hearing loss group (N group),and propofol + noise-induced hearing loss group (P + N group).A loading dose of propofol 5 mg/kg was infused intravenously over 5 min,followed by infusion at 20 mg· kg-1 · h-1 for 115 min in P and P + N groups,while the equal volume of normal saline was given in N group.N and P+ N groups were exposed to the noise of 4 kHz center frequency and 118-122 dB sound pressure level for 120 min.Distortion product otoacoustic emission (DPOAE) was measured before noise exposure (T1) and at 1 h after the end of noise exposure (T2) and the amplitudes were recorded at the frequencies of 2,4,6 and 8 kHz.After the second measurement of DPOAE,all the animals were sacrificed and organs of Corti were harvested for determination of cochlear 8-isoprostaglandin F(2alpha) (8-iso-PGF2α) content (by ELISA assay) and out hair cell (OHC) count.The rate of OHC lesions was calculated.Results The DPOAE amplitude was significantly lower at frequencies of 4,6 and 8 kHz at T2 than at T1 in N and P + N groups (P < 0.05).Compared with C group,the DPOAE amplitude was significantly decreased at frequencies of 4,6 and 8 kHz at T2,while the cochlear 8-isoPGF2α content and rate of OHC lesions were increased in N and P + N groups (P < 0.05).Compared with N group,the DPOAE amplitude was significantly increased at frequencies of 4,6 and 8 kHz at T2,while the cochlear 8-iso-PGF2α content and rate of OHC lesion were decreased in P + N group (P < 0.05).Conclusion Propofol can reduce noise-induced hearing loss in guinea pigs possibility through decreasing oxidative stress response-induced damage to cochlear OHCs.
6.Effect of isoflurane anesthesia on noise-induced hearing loss in guinea pigs

Objective To evaluate the effect of isoflurane anesthesia on noise-induced hearing loss in guinea pigs.Methods Forty-eight healthy adult male guinea pigs,aged 3 months,weighing 400-500 g,were randomly divided into 4 groups (n =12 each) using a random number table:control group (C group),isoflurane group (I group),noise-induced hearing loss group (N group),and isoflurane + noise-induced hearing loss group (I + N group).Isoflurane was inhaled for 140 min at a concentration of 1% in I and I + N groups.N and I + N groups were exposed to the noise of 4 kHz center frequency and 118-122 dB sound pressure level for 120 min starting from 20 min after administration.Mean arterial pressure (MAP) was recorded at 10,40,70,100 and 120 min of exposure to noise and cochlear blood flow (CoBF) was recorded before administration and at 10,40,70,100 and 120 min of exposure to noise.Auditory brainstem response (ABR) threshold was recorded before administration and at 1 h,72 h,and 10 days after the end of exposure to noise.Arterial blood samples were obtained and the plasma noradrenaline (NE) concentration was detected by HPLC before exposure to noise and immediately after the end of exposure to noise.Results Compared with group C,MAP and the change rate of CoBF were significantly decreased,and the plasma NE concentration was increased immediately after the end of exposure to noise in I group,and MAP was increased,the change rate of CoBF was decreased,and the plasma NE concentration immediately after the end of exposure,and ABR threshold after the end of exposure were increased in N and I + N groups.Compared with N group,MAP was significantly decreased,the change rate of CoBF was increased,the plasma NE concentration immediately after the end of exposure,and ABR threshold at 1 and 72 h after the end of exposure were increased,and no significant was found in ABR threshold at 10 days after the end of exposure in I + N group.Conclusion Isoflurane anesthesia exerts temporary but not permanent protective effects against noise-induced hearing loss in guinea pigs and partial inhibition of activation of sympathetic nerve and increased CoBF may be involved in the mechanism.
8.Study on the Improvement Effects of Different Polar Parts of Sanye Chongcha on Model Rats with Ulcer-ative Colitis

OBJECTIVE:To explore the improvement effect of different polar parts of Sanye Chongcha on models rat with ul-cerative colitis(UC). METHODS:The total extract was obtained from Sanye Chongcha by extraction with 80% ethanol,and dis-persed in water,from which the extracts of corresponding parts were obtained after extraction successively with trichloromethane and ethyl acetate and drying. 60 rats were randomized into normal group,model group,positive group(sulfasalazine enteric-coated tablets,312.5 mg/kg),trichloromethane extraction part group(70 mg/kg),ethyl acetate extraction part group(35 mg/kg)and wa-ter extraction part group(320 mg/kg). The rats except for normal group received 2,4,6-trinitrobenzenesulfonic acid solution(TN-BS)to establish UC model. 24 h later,the rats in the drug administration groups were given corresponding drugs ig,while those in normal group and model group were given normal saline ig,once a day,for 14 consecutive days. For the rats,the pathological change of the colon tissue was observed,and the colon mucosa damage index(CMDI)was scored;the activity of superoxide dis-mutase(SOD)and the content of malondialdehyde(MDA)in serum were determined. RESULTS:Compared to normal group,the rats in model group had obvious UC-like lesion,higher CMDI,lower activity of SOD and higher content of MDA in serum(P<0.01). Compared to model group,the rats in the drug administration groups had the lesion which became less serious or improved and had lower CMDI score,among which those of the ethyl acetate extraction part group and the positive group demonstrated a sta-tistically significant difference(P<0.01);and the rats in the drug administration groups except for trichloromethane extraction part group had higher activity of SOD and lower content of MDA in serum,among which the activity of SOD of the ethyl acetate ex-traction part group and the activity of SOD and content of MDA of the positive group demonstrated a statistically significant differ-ence(P<0.05 or P<0.01). CONCLUSIONS:The ethyl acetate extraction part of Sanye Chongcha can reduce colonic lipid peroxi-dation and thus has obvious improvement effect on the rats with UC caused by TNBS.
9.The X-ray features of breast ductal carcinoma in situ and its small invasive foci and correlation between mammographic features and prognostic biologic factors

Objective To retrospectively evaluate the mammographic features of breast ductal carcinoma in situ (DCIS)and DCIS with small invasive foci,and to analyze the correlation between the mammographic findings and the prognostic biologic factors.Methods The mammographic examination was performed in 95 consecutive women with breast DCIS(n = 50)and DCIS with invasive foci(n = 45 ).The prognostic biologic factors including progesterone receptor(PR),C-erbB-2,and p53 were evaluated in 62 of 95 cases.Categorical data were expressed as percentages and analyzed by using the X~2 test,and furthermore the odds ratio was measured.Results(1)Only one abnormality was seen on mammography in 62 patients. Combined two abnormalities on mammography were seen in 26 patients.Mammograms were normal in 7 patients.(2)Calcifications with or without other abnormality were noted in 62 cases.Of them,73% (n =45)had higher probability of malignancy calcifications and the others were intermediate concern calcifications.Clustered calcifications(36 lesions)was the most common distribution,which usually accompanied by another abnormality.And then were segmental(18 lesions)distributed pattern.As far as the shape of mass (n = 22)was concerned,the oval shaped lesion(13 cases)was the most common,and the margin of the mass appeared as ill-defined in 15 eases,microlobulated in 1,circumscribed in 4,and obscured in 2,respectively.Isodensity mass had a higher frequency in this group(12/22,55%).Other non-calcification findings included architecture distortion(7 cases),local asymmetry (15 cases),global asymmetry (5 cases),and solitary dilated duct (3 cases),and most of them accompanied with other signs. (3)For expression profile of the biological factors,significant differences were found among malignant calcification group,intermediate concern calcification group,and non-calcification group. The odds of PR positive for the lesions noted as non-calcification were 11.00 times higher (X~2 =8.571 ,P=0.003 ;95% CI, 1.998—60.572)than the lesions noted as intermediate concern calcifications,and 8.80 times higher (X~2 = 9.748,P=0.002 ;95% CI,2.024—38.253)than the lesions noted as malignant calcifications.The odds of C-erbB-2 positive for the lesions showed as malignant calcifications were 12.35 times higher (X~2=7.353, P=0.007 ;95% CI,1.447—105.443)than the lesions showed as non-calcification,and 5.74 times higher (X~2=4.977,P = 0.026;95% CI,1.110—29.645)than the lesions showed as intermediate concern calcifications.Conclusion The mammographic features of DCIS and DCIS with small invasive foci were characteristic.Mammographic findings could be a prognostic markers,which could provide a possibility for making a treatment plan.
