1.Study on methylation of the p73 gene in acute lymphoblastic leukemia and its clinical significances

Objective To investigate the effects of methylation of p73 gene on pathogenesis of acute lymphoblastic leukemia.Methods The restriction endonuclease enzyme digestion combined with PCR was used to detect p73 gene and methylation in 42 patients with acute lymphoblastic leukemia.Results Methylation of p73 gene was found in 28.6%(12/42).Patients without methylation of p73 gene had a higher complete remission rate of first chemotherapy and a longer survival time than those with methylation of p73 gene.Conclusion Methylation of p73 gene probably plays a role in the pathogenesis of acute lymphoblastic leukemia and may have clinical significance in predicting prognosis of ALL.

3.Clinical value of hepatic vein drainage territory evaluation before LDLT with right lobe graft

Objective To investigate the relationship between diameter and drainage territory of tributaries of the hepatic vein and the clinical value of hepatic venous drainage territory evaluation before LDLT.Methods MSCT covering abdomen was performed on 68 donors.The images were transported and reconstructed in GE Advantage Windows 4.2 workplace and IQQA-liver CT images readout and analytical system.The tributaries of hepatic vein were marked,and the diameter and drainage territory were measured.Functional hepatic volume (FHV),and effective hepatic volume (EfHV) of donors and recipients according to the hepatic venous reconstruction results during the operation were calculated.The corresponding graft-to-recipient body weight ratio of recipients was calculated.Results In the tributaries of 220 hepatic veins studied,the coefficient correlation of diameter and drainage territory was 0.752 (P≤0.01),but there was significant difference in the drainage territory of the veins with same diameter.By using Chi-square analysis,venous diameter and drainage territory are classified and statistical frequency by 5 mm and 50 cm3 respectively,and there was statistically significant difference between them (X2 =61.97,P<0.01).The mean value of drainage territory of RHV (RHVV) was 510.80 ± 168.33 cm3,and that of RFHV (drainage territory with reconstructed IRHV added) was 577.26± 156.72 cm3 respectively.The mean value of drainage territory of LHV (LHVV) was 292.70 ± 76.61 cm3,and that of LFHV (drainage territory with Ⅳ segment tributaries of MHV added) was 551.26 ± 111.82 cm3 respectively.In 25 grafts without MHV,the mean value of drainage territory of EfHV was 405.52-1038.43 cm3.In 43 donors donating grafts with MHV,the mean value of drainage territory of EfHV was 175.35-575.35 cm3.Conclusion There is significant difference in drainage territory of same diameter veins.Bigger diameter doesn't always mean bigger drainage territory.Hepatic vein drainage territory evaluation before LDLT has important clinical significance to formulate surgery scheme.

Objective:To investigate the immune depression to donor-origin cells induced by the Mixed Chimera after sublethal whole body irradiation.Methods:Recipients in the experiment were Wistar and SD rat,and Wistar rats were selected as the donor.Donor and recipient rats were divided randomly into three groups.Recipient were conditioned with sublethal whole body irradiation (WBI).Group A was infused with bone marrow cells (BMC) of Wistar rats;group B infused with bone marrow mesenchymal stem cells (BM-MSCs)of Wistar rats;and group C with normal saline.Then they were administered cytoxan(CTX) by intraperitoneal injection.The mechanisms for immune depression were explored by performing mixed lymphocyte reaction (MLR).Results:The results showed that donor lymphoid chimeras could be found in the immune depression SD rats and chimerac cells in group A was more than in group B by FCM assay (P
5.Gastrointestinal stromal tumors:imaging features with intraoperative and clinicopathological correlation

Objective To analyze imaging of gastrointestinal stromal tumors(GIST),and to compare their imaging features with operational and pathological findings.Methods Clinical,imaging,and pathological data of 20 patients with GIST were collected.Results Imaging findings were endophytic or exophytic tumors with heterogeneous density or signal intensity,corresponding to hemorrhage,necrosis,and cystic changes.Imaging was correct for the location of the lesion in 11 of 16 primary GIST and 4 cases of relapsed tumors.Preoperational CT did not detect mesenteric,peritoneal,and omental metastasis in 5 cases. Hepatic metastases detected at CT (3 cases )were identified by operational findings.Conclusions GIST has some imaging features.CT is a useful tool in detecting and characterizating of lesions rather than detecting mesenteric,peritoneal,and omental metastasis.
6.Serum levels of antibody against enterovirus 71 in healthy children at Shanghai in 2011

Objective To test the levels of enterovirus 71 (EV71) antibody among children of different ages in Shanghai in 2011,and to investigate the relationship between antibody levels and virus infection.Methods EV71 antibody was detected by microneutralization assay from the serum specimens of healthy children of different ages collected during July to August,2011.The results were analyzed by t test for quantitative data with normal distribution,and by x2 test for count data.Results The positive rate of EV71 antibody among the 93 serum specimens was 58.1% (54/93).The geometric mean titer (GMT) of EV71-specific neutralizing antibody was 1 ∶ 14.48.The positive rate of EV71 antibody in infants less than 6 months old was 87.5% (21/24),and the GMT was 1∶29.56.In children aged 2 to 3 years,the positive rate of EV71 antibody decreased to 3.7% (1/27),and GMT decreased to 1∶4.21,which were both statistically significantly lower than those less than 6 months old (x2 =36.37,t=7.58; both P<0.01).The positive rate of EV71 antibody increased to 83.3% (20/24) in children aged 5 to 6 years,with GMT reaching 1∶21.74.Whereas in children aged 7 to 8 years,the positive rate was 66.7% (12/18) and GMT was 1∶20.76,without statistically significant difference compared with those aged 5 to 6 years (x2 =1.58,t=0.597; both P>0.05).No statistically significant difference was found between boys and girls in positive rate of EV71 antibody [62.7 % (32/51) vs 52.4 % (22/42),x2 =1.02,P>0.05] or GMT (1 ∶ 16.23 vs 1 ∶ 12.61,t=0.881,P>0.05).Conclusions Children aged 2 to 3 years were at higher risk for EV71 infection,with EV71 antibody level significantly lower than other age groups.
7.Case-control study on spinal leveraging manipulation and medicine for the treatment of degenerative scoliosis.

OBJECTIVETo compare clinical effects of spinal leveraging manipulation and medicine for the treatment of degenerative scoliosis in pain and function.
METHODSFrom July 2010 to June 2013, 38 patients with degenerative scoliosis were randomly divided into spinal leveraging manipulation group and medicine group by coin tossing. In manipulation group, there were 9 males and 11 females aged from 58 to 74 years old with an average of (66.63±7.73), the courses of diseases ranged from 3 to 8 months with an average of (5.65±2.58), spinal leveraging manipulation(following meridian to straighten tendon,relieving spasm, osteopathy and massage, clearing and activating the channels and collaterals) were performed for 30 min, once a day, 4 days for a period treatment, totally 9 courses. In medicine group, there were 8 males and 10 females aged from 57 to 70 years old with an average of (63.51±6.61) the courses of diseases ranged from 3 to 5 months with an average of (4.82±1.43), celecoxib with eperisone hydrochloride were orally taken, 4 days for a period treatment, totally 9 courses. VAS score, Cobb angle and ODI score were measured.
RESULTSAfter treatment, VAS score in manipulation group was (5.38±0.99), (6.36±1.31) in medicine group,and had significant meaning (t=2.618, P<0.05); there was significant differences in Cobb angle between manipulation group (16.51±4.89)° and medicine group (19.85±5.03) °(t=2.074,P<0.05); and had obviously meaning in ODI score between manipulation group (20.20±2.93) and medicine group (26.01±3.11) (t=5.592, P<0.05).
CONCLUSIONSpinal leveraging manipulation for degenerative scoliosis could regulate muscle balance on both side of spine, correct coronal imbalances in spine, recover normal sequence of spine, reduce and remove opperssion and stimulation of nerve root, relieve pain in leg and waist and further improve quality of life.

8.Rare parotid gland tumors: enhanced computed tomography and pathological correlation.

OBJECTIVETo investigate the correlation between enhanced computed tomography (CT) findings and pathological results of rare parotid gland tumors, and improve diagnosis accuracy.
METHODSThe enhanced CT manifestations of 22 cases with pathologically documented rare parotid gland tumors, which included 6 cases of basal cell tumor, 5 cases of myoepithelioma, 4 cases of vascular invasion, 3 cases of lymphatic cyst, 3 cases of lipoma, and 1 case of chondrosarcoma, were retrospectively analyzed. The location, size, shape, density, and relationship with surrounding structure were evaluated on CT images.
RESULTSThe enhanced CT showed that basal cell tumors occurred in the superficial lobe of the parotid gland, with clear boundary, within the cystic lesion. The lesions were moderate to obviously enhanced, which may be accompanied by enlarged lymph nodes. Myoepithelial tumors were located in the superficial lobe of the parotid gland, with a small cystic prone and microcalcification within a few cases. The lesions were moderate to obviously enhanced. Hemangiomas of soft tissue mass prominent in the parotid gland surface were mild to significantly enhanced. Larger lesions may occupy the entire parotid gland, with uneven density and visible vein stone. The CT density values of the lymphatic cyst were usually higher. Chondrosarcoma mainly manifested cystic mass at the calcification edge. Lipoma with fat density mass exhibited clear boundary without enhancement. Fiber separation could be observed in the lesion.
CONCLUSIONCT can reflect the pathological features of rare parotid gland tumors by demonstrating their corresponding imaging features. Enhanced CT is the most effective means of imaging to identify the nature of rare tumor of the parotid gland lesions.

9.Cropping system and research strategies in Panax ginseng.

Panax ginseng is the king of herbs and plays important roles in the traditional Chinese medicine industry. In this paper, we summarized the development of ginseng cultivation in China and other main countries, analyzed the effects of ecological factors of soil and climate on ginseng distribution, and investigated the characteristic of main cultivation patterns (conversion of forest to cultivate ginseng soils, cultivated ginseng in the farmland and wild nursery). Aimed at the serious issues in the cultivation, research strategies have been provided to guarantee the sustainable development of the ginseng industry. The patterns of cultivated ginseng in the farmland should be strive to develop; pollution-free cultivation and studies of continuous cropping obstacles should be carried out; ginseng varieties suited to ecological environment of farmland should be bred using modern biotechnology.

10.EFFECTS OF 1?,25(OH)_2D_3 ON THE EXPRESSION OF RECEPTOR ACTIVATOR OF NF-?B LIGAND AND OSTEOPROTEGERIN IN RAT OSTEOBLASTS IN VITRO

Objective To investigate the effect of 1?,25-dihy-droxyvitamin D3 (1?,25(OH)2D3) on the expression of receptor activator of NF-?B ligand (RANKL),osteoprotegerin (OPG) and RANKL mRNA,OPG mRNA,of rat osteobalsts (OB). Method The expression of RANKL and OPG was detected by the method of Immunohistochemistry and ELISA. RANKL mRNA and OPG mRNA were determined through FQ-PCR. Results Compared with the control group and the group with 1 nmol/L 1?,25(OH)2D3,10 and 100 nmol/L 1?,25(OH)2D3 can significantly induce the expression of RANKL and RANKL mRNA. 1,10 nmol/L 1?,25(OH)2D3 can stimulate the expression of OPG and OPG mRNA significantly,while 100 nmol/L 1?,25(OH)2D3 can inhibit the expression of OPG mRNA significantly. There were no difference in the rate of RANKL/OPG and RANKL mRNA/OPG mRNA between the control group and the group with 1 nmol/L 1?,25(OH)2D3,however the rate of RANKL/OPG and RANKL mRNA/OPG mRNA in the group with 10,100 nmol/L were higher than the control group and the group with 1 nmol/L 1?,25(OH)2D3. Conclusion Lower dosage of 1?,25(OH)2D3 (1 nmol/L) had no significant effect on bone turnover,but higher dosage of 1?,25(OH)2D3 (10,100 nmol/L) can enhance bone turnover through facilitating the formation and activity of osteoclasts via enhancing RANKL mRNA/OPG mRNA and RANKL/OPG..
