1.Roles of regulator of G-protein signaling 5 in tumors

Regulator of G-protein signaling 5 (RGS5) belongs to RGS family,which can negatively regulate the conduction of this signaling pathway.RGS5 mainly expresses in vascular pericyte,and is closely related to the occurrence,development and maturation of the blood vessels.Loss of RGS5 results in pericyte maturation,tumor vascular normalization,and these changes can improve the curative effect combined with chemotherapy and immunotherapy,indicating that RGS5 may become a new target of anti-tumor treatment.In addition,RGS5 involves in tumor metastasis and apoptosis,which can improve antineoplastic effect by inducing tumor cells apoptosis.
2.CT quantitative diagnosis in fatty liver: a clinical study

Objective To establish the CT criteria of quantitative diagnosis for liver steatosis by means of studying the CT features of fatty liver cases proven histologically. Methods Twenty-eight cases of fatty liver were underwent non-enhanced CT scan, and the attenuation of liver parenchyma was measured. To differentiate the degree of fatty liver, the mean CT value and the relative density of hepatic vessels were observed. The quantitative diagnosis was made according to the CT number threshold and the criteria of relative density of hepatic vessels, respectively. Results Among the 28 cases, there were 17 cases of mild steatosis with mean CT number of 46 HU (32-65 HU), 7 cases of middle degree fatty liver with mean CT number of 28 HU (15-38 HU), and 4 cases of sever fatty liver with mean CT number of 0.2 HU (-7-11 HU). For the relative density of hepatic vessels, 16 of the 17 cases of mild fatty liver had a appearance of hepatic vessels immersion and 1 mild case had reverse hepatic vessels display, 6 of 7 middle degree cases had reverse hepatic vessels display with 1 case having the appearance of hepatic vessels immersion, and all the 4 case of sever steatosis had the appearance of reverse hepatic vessels display with sharp contrast between vessels and the liver parenchyma. 3.The Influence of Neoadjuvant Chemotherapy Combined with Psychological and Ethical Intervention on Mood and Life Quality of Patients with Cancer

Objective:To understand the influence of neoadjuvant chemotherapy combined with psychological and ethical intervention on mood and life quality of patients with cancer. Methods: The 120 patients with locally advanced breast cancer were randomly divided into control group ( neoadjuvant chemotherapy group) and interven-tion group ( neoadjuvant chemotherapy combined with psychological ethics intervention group) , Chemotherapy and after chemotherapy respectively adopt Zung Self -Rating Depression Scale ( SDS ) , Zung Self -Rating Anxiety Scale (SAS) and Quality of Life Core Scale (QLQ-C30) as an investigative tool, mood and quality of life of the patients were assessed. The patients' mood and life quality were investigated. Results: Emotional state intervention group patients more significantly than control group patients improved (P<0. 01) and improve life quality. Con-clusions:Neoadjuvant chemotherapy for localized breast cancer patients with psychological ethics intervention can effectively reduce the patient's anxiety and depression, and improve the life quality of patients.
5.Application of anatomic segmental hepatectomy in treatment of left hepatolithus

Objective To investigate the application value of anatomic segmental hepatectomy in treatment of left hepatolithus.Methods The 80 patients with left hepatolithuswere divided into the control group (n =40)and the observation group (n =40)in accordance with dif-ferent surgical methods,and they were given traditional surgery and precise pedicle anatomic segmental hepatectomy respectively.Operation index levels,postoperative complications and stress reaction levels before and after surgery of the two groups were compared.Results The blood loss,operative time,the time of drainage tube remove,time of analgesic drug use,and postoperative hospital stay of the observation group were significantly less than the control group,and the difference was statistically significant (P <0.05).The rate of postoperative com-plications was 17.50% in the control group,while it was 15.00% in the observation group,which showed no significant difference between the two groups (P >0.05).During the surgery,epinephrine,cortisol,IL-6 and CRP levels of patients decreased after a slight increase, while NK cell levels of patients decreased firstly and then increased.There were statistically significant differences in trems of the above inde-xes immediately after surgery to 24 hours after surgery (P <0.05),and the rangeability of the observation group was less than the control group (P <0.05).Until 48 hours after surgery the above indexes recovered to the preoperative level.Followed up for 3 to 24 months,the calculi recurrence rate and abdominal ache recurrence rate of the observation group were lower than the control group,and the difference was statistically significant(P <0.05).Conclusion Precise pedicle anatomic segmental hepatectomy applied in the treatment of left hepatolithus showed makedly curative effect,and it has little effect on the stress response of patients.
6.Expression and purification of soluble recombinant Hexastatin in E.coli

purpose To construct the expression vector of Hexastatin gene,to express and to purify the recombinant protein for further activity research.Methods The human Hexastatin gene was isolated by RT-PCR from EC9706 cells total RNA and cloned into pMDl8-T for sequencing.Then the Hexastatin gene was subcloned into pMAL-c4x expression vector and induced to express by IPTG.The recombinant fusion protein was purified with Amylose Resin Heads.Results RT-PCR product was about 687 bp and its sequence was the same as that of Hexastatin reported.The recombinant protein was expressed in E.coli BL21 with high level and the soluble protein accounted for 24.8% of the total bacterial protein,The purification of recombinant protein purified with Amylose Resin Heads reached more than 90%.Conclusion The cloning,expression and purification of human Hexastatin have laid a foundation for its anti-angiogenesis therapy for tumor.
7.Comparison of myocardial injures between asphyxia and ventricular fibrillation models of cardiac arrests

Objective To compare the difference in cardiac injuries between asphyxia and ventricular fibrillation modes in different periods after cardiac arrest (CA).Methods The model was established in Cardiopulmonary Resuscitation Lab,Sun Yat-sen University.A total of 35 male SD rats were used to produce the asphyxia or ventricular fibrillation (VF) cardiac arrest models randomly.Both of the two modes were induced 8 minutes cardiac arrest.The myocardial HE stains,mitochondrial respiratory control ratio (RCR),and echocardiography were observed at 4 h,24 h and 72 h after ROSC (restoration of spontaneous circulation).The results were expressed as (-x ± s),t test was performed to compare between two groups,and one way analysis of variance was used to compare multiple groups.P < 0.05 was considered as significant difference.Results HE stains showed damages were more serious in the VF mode than in asphyxia mode at 4 h,and both of them had a disorderly-arranged myocardium at 72 h.RCR in VF mode became worse at 4 h,and RCR resumed at 24 h in both modes without significant difference compared with the sham operated rats.The echocardiography showed VF mode had a lower left ventricular ejection fraction (LVEF) than asphyxia mode at 4 h (29.68% vs.42.16%,P =0.03),and there was no difference in LVEF between VF mode and the sham operated rats at 24 h,however no difference in LVEF between the asphyxia and sham operated rats at 72 h.Both of them had a thicker left ventricular anterior wall than the sham operated rats at 72 h (2.41 mm vs.1.72 mm,P=0.013; 2.61 mmvs.1.72 mm,P=0.007),and there was no significant difference between them.Conclusions The ventricular fibrillation mode has a more severe injuries in early period,but it recovers sooner than asphyxia one.Both of two groups get compensatory left ventricular hypertrophy in later period of ROSC.
8.Expression of Bmi-1 and p16 gene in transitional cell carcinoma of bladder

Objective To detect the expression of Bmi-1 and p16 gene in transitional cell carcinoma of bladder(TCC) tissue and explore its clinical significance.Methods The expression of Bmi-1 and p16 gene were detected by real-time quantitative polymerase chain reaction in 61 cases of TCC tissue and 12 cases of normal bladder tissue.Results The expression of Bmi-1 gene in TCC tissue was significantly higher than that in normal bladder tissue (0.242 ± 0.129 vs.0.031 ± 0.011),and the expression of p16 gene was significantly lower than that in normal bladder tissue (0.059 ± 0.021 vs.0.165 ± 0.029),there was significant difference (P < 0.05).The expression of Bmi-1 and p16 gene were highly correlated with pathological grades,clinical stages and tumor recurrence (P < 0.05 or < 0.01).But there were not correlated with age and gender (P > 0.05).There was a negative correlation between the expression of Bmi-1 gene and p16 gene in TCC tissue(rs =-0.714,P< 0.05).Conclusions Bmi-1 gene high expression and p16 gene low expression may be involved in the occurrence and development process of TCC.Bmi-1 may decrease the expression of p 16 gene in some ways,and then lead to the occurrence and development of TCC.
9.Management of urinary tract obstruction in patients with kidney transplantation

Objective To investigate the treatment of urinary tract obstruction after kidney transplantation, and to introduce the novel ‘antegrade percutaneous urinary tract throughout guidance technique’ with guide wire in selected patients. Methods 43 cases of renal transplantation patients due to urinary tract obstruction (ureteral stenosis and hydronephrosis in 24 cases, urinary calculi with or not ureteral stenosis in 19 cases) received endourological treatment. Retrograde pathway was tried firstly, if failed, antegrade pathway was adopted with‘antegrade percutaneous urinary tract throughout guidance technique’, 18 Fr percutaneous tract was established if necessary, endoscopy was needed in antegrade or combine with retrograde pathway. Results Of the 43 patients, 9 (20.9 %) patients were managed directly through the retrograde ureteroscopy, 28 (65.1 %) patients were managed with anterograde percutaneous technique in 18 Fr tract, 6 (14.0 %) patients were treated with ‘antegrade percutaneous urinary tract throughout guidance technique’. Operation time was (72.0 ± 16.0) (45 ~ 95) minutes. Postoperative stone clearance rate of 89.5 % (17/19). Of 32 cases with ureteral stricture, 6 (6/32, 18.8 %) cases were cured after one time of dilation and JJ stent indwelling for 2 months, 18 (56.3%) cases with stable hydronephrosis after 2-3 times of dilation and JJ stent indwelling, 5 cases (15.6 %) needed long-term repeated stent indwelling for drainage, 3 (9.4%) cases required nephrostomy tube drainage. Conclusion Minimally invasive treatment of urinary tract obstruction after renal transplantation is effective. Antegrade percutaneous urinary tract throughout guidance technique provided a powerful guarantee for retrograde operation and avoided the bleeding risk following percutaneous renal surgery, when combined with flexible ureteroscopy, urinary calculi in patients with kidney transplantation can be effectively managed with little trauma.
10.Clinical significance of detection of CD_4~+CD_(25)~(high)CD_(127)~(low) regulatory T cells in peripheral blood of NHL patients

Objective To investigate the change of CD_4~+CD_(25)~(high)CD_(127)~(low)> regulatory T cells (Trcg cells) sub-group level in peripheral blood of non-Hodgkin lymphama (NHL) patients, and to explore its clinical significance. Methods Treg cells levels in peripheral blood of lymphoma patients and normal were detected by flow cytometry, followed by statistical analysis. Results In the 65 cases of NHL patients, Treg cells in peripheral blood were (6.72±1.38) %, higher than that in the normal control group (5.65±0.68) % (P <0.05). Percentage of Treg cells are significantly different between clinical stages and normal: [P <0.05, normal control group (5.65±0.88) %, Ⅰ -Ⅱ period (6.08±1.18) %, Ⅲ-Ⅳ period (6.95±0.85) %]. The percentage of Treg cells are also different among pathological types of patients and normal [P <0.05, normal control group (5.65± 0.68) %. The percentage of Treg diffuse large B-cell lymphoma (5.83±0.95) % and other subtypes of lymphoma (7.83±1.76) %]were observed. It is not sure that Treg cells percentage among patients with different levels of lactate dehydrogenase and normal are significantly different. [P >0.05, normal control group (5.65±0.68) %, patients with normal LDH group (6.97±1.20) %, patients with lactate dehydrogenase (6.54±1.02) %]. Conclusion Treg cells induced by tumor and could inhibit the immune cells, Treg cells percentage in peripheral blood of tumor patients is higher than that the normal control group, and increased with the clinical staging, so the percentage of Treg cells may serve as a clinical indicator to evaluate tumor load.
