1.Impacts on the curative effect of peripheral facial paralysis treated with acupuncture and moxibustion at different times.

OBJECTIVETo explore the optimal intervention time of acupuncture and moxibustion in the treatment of peripheral facial paralysis.
METHODSTwo hundred and four cases of peripheral facial paralysis were collected in clinic for retrospective analysis. According to the time between the disease onset and the intervention of acupuncture and moxibustion, 3 groups (groups A, B and C) were divided. In group A, the treatment was given in 1 to 3 days after onset; in group B, the treatment was given in 4 to 7 days after onset; in group C the treatment was given in 8 to 10 days after onset. The treatment with acupuncture and moxibustion was applied according to the regular stages of disease in the three groups. At the acute stage, the less points and shallow puncture therapy combined with distal acupoints at the limbs were adopted on the affected side. At the stable stage, the more points and shallow puncture therapy were used on the affected side. At the recovery stage, the deep puncture or penetrating punctures was applied on the affected side. The curative effects was given once every day and the treatment of 10 days made one session. The curative effects were analyzed statistically at the end of three sessions of treatments separately.
RESULTSAt the end of the 1st session of treatment, the total effective rate was 58.8% (40/68) in group A, 69.7% (53/76) in group B and 46.6% (28/60) in group C. At the end of the 2nd session of treatment, the total effective rate was 85.3% (58/68), 90.8% (69/76) and 71.6% (43/60) in group A, B and C separately. At the end of 3rd session of treatment, the total effective rates were 89.7% (61/68), 97.4% (74/76) and 83.3% (50/60) in the three groups separately. The differences were significant statistically at the same session of treatment among three groups (all P<0.05), in which, the results in group B was the best, followed by group A and C in sequence.
CONCLUSIONthe best intervention time of acupuncture and moxibustion is in 4 to 7 days after onset of facial paralysis.

3. Rapid identification of classical herbal formulae Baoyinjian's chemical substance based by HPLC-Q/TOF-MS

Objective: To analyze the chemical constituent cluster of classical herbal formulae Baoyinjian systemically by HPLC-Q/TOF-MS. Methods: The seperation was performed on Diamonsil C18 (250 mm × 4.6 mm, 5 μm) column with gradient elution with acetonitrile-0.1% formic acid. The column temperature was 30 ℃, the flow rate was 1 mL/min, the injection volume was 10 μL, and the mass spectrometry condition was X500R QTOF mass spectrometer, electrospray ion source, positive and negative mode scanning. Results: A total of 52 chemical constituents were identified by reference confirmation, literature comparison, and high mass spectrometry data analysis. The chemical constituent cluster was composed of 17 flavonoids, six phenolics, 12 iridoid glycosides, eight alkaloids, one phenethyl alcohol glycosides, four monoterpene glycoside, two triterpenes and two other compound. Conclusion: This study can identify various chemical constituents of Baoyinjian systematically, accurately, and rapidly, which provides a basis for the determination of the quality attributes of Baoyinjian.
4. Practice and progress on bionic technology in identification on five tastes of Chinese materia medica

Four properties and five tastes are the core content of medicinal theory of Chinese materia medica (CMM), and it is important for modern study of CMM to elucidate the scientific connotation of five tastes based on the application of bionic technology. With the development of bionic technology, electronic nose and electronic tongue have become the objective method to determine the taste and smell in recent years. Based on the taste and odour connotation of five tastes of CMM, the working principle, research method, and identification of flavor substances in CMM of electronic nose and electronic tongue were summarized, and the progress on original variety, production place, growth period, storage life and processing technology of CMM were reviewed in this article. Combined with the practice of our research group on the characterization of medicinal properties, the basic research model of the definition and characterization of the five tastes of effective materials basis was proposed, which would provide a reference method for the medicinal research on five tastes of CMM.
5.Biological characteristics of human amniotic fluid-derived embryonic mesenchymal stem cells isolated and cultured using the two-step method

BACKGROUND:In vitro isolation and purity technique of stem cells mostly depends on the identification of cell surface marker,such as monoclonal antibody adherent spreading method,flow cell sorting method and immunomagnetic beads sorting method,but the operation was complicated and the price was high.OBJECTIVE:To observe the biological characteristics of human amniotic fluid-derived embryonic mesenchymal stem cells,which were isolated and cultured using the two-step method.DESIGN,TIME AND SETTING:The opening study was conducted at the Stem Cell Research Room of Xinjiang Medical University from March 2008 to March 2009.MATERIALS:Totally 10 amniotic fluid specimens were obtained from pregnant women who underwent prenatal diagnosis following 16-22 weeks of gestation or voluntarily induced abortion.With ultrasonic guidance,amniocentesis was performed to collect 20-40 mL amniotic fluid.METHODS:Human amniotic fluid-derived embryonic mesenchymal stem cells were isolated and cultured using the two-step method.Amniotic fluid was first centrifuged and incubated till spindle-shape cells were seen,with the presence of flbroblast-tike cell colonies.Supematant was moved to a new 25 cm~2 culture flask for further culture till spindle-shape fibroblast-like mesenchymal stem cell colonies.When 70% confluence,cells were digested,and incubated in α-MEM,supplemented with basic fibroblast growth factor,served as the first passage.MAIN OUTCOME MEASURES:Morphological changes in human amniotic fluid-derived embryonic mesenchymal stem cells of primary culture and subculture were measured.Karyotype,cycle,growth curve and colony formation ability of human amniotic fluid-derived embryonic mesenchymal stem cells were measured.Surface antigen and cytokine were examined using flow cytometry,immunofluorescence and RT-PCR.RESULTS:Human amniotic fluid-derived embryonic mesenchymal stem cells were successfully isolated and subcultured.During metaphase,primarily cultured amniotic fluid cells presented scattered spindle cells and flbroblast-like mesenchymal stem cell colonies every 7 days.Passaged cells completely adhered in 12 hours.Following 1 or 2 days of latent period,cells proliferated rapidly.About 90% confluence was observed following 6 or 7 days of culture.Cell arranged regularly,showing whirlpool-shape,radiated shape.Cells were spindle-shape,with unclear boundary.Chromosome karyotype of human amniotic fluid-derived embryonic mesenchymal stem cells was normal diploid.Growth curve showed "S" shape,but the two-step method reached a peak at (6.1±0.5) days,which was significantly rapid compared with the one-step method (7.2±0.6) days (P=0.035).Flow cytometry analyses showed that P3 cells at S phase took up (14±2.3)% using the two-step method,which was more than the one-step method (9.0±1.4)% (P=0.031).Low-density human amniotic fluid-derived embryonic mesenchymal stem cells were incubated for 7 days prior to cells formed scattered cell colonies.However,colony forming efficiency using the two-step method (15.0±2.3)% were significantly more than the one-step method (10.0±1.8)% (P=0.021).Flow cytometry results showed that human amniotic fluid-derived embryonic mesenchymal stem cells expressed CD44,CD29 and CD105,but were negatively for CD45,CD34,HLA-DR.Immunofluorescence suggested that Oct-4-positive cells were observed in amniotic fluid.However,the proportion of Oct-4-positive cells using two-step method (1.2±0.3)% was significantly greater than the one-step method (0.9±0.2)% (P=0.041).RT-PCR suggested that human amniotic fluid-derived embryonic mesenchymal stem cells obtained using the two methods expressed Oct-4.CONCLUSION:Human multipotent mesenchymal stem cells are present in human amniotic fluid.The two-step culture protocol could be a kind of high performance and simple protocol which may not interfere with the normal prenatal diagnosis procedure.
6.The Normal Anatomy and CT Study of HDL

Objective To study the normal anatomy and pathologic CT findings of HDL and compare the HDL dimension of two conditions.Methods 160 consecutive CT scans of the normal upper abdomen were reviewed and 19 cases of cadavers were used.Results There was no statistics differences between the cadavers and normal subjects on HDL.Conclusion The real dimension of HDL can be replaced by the measurement on CT image,CT is a accurate means of measuring HDL in vival of human being.
7.Study on the Expression of Suppressor of Cytokine Signaling -1 ( SOCS1 ) in the Liver of Septic Mice

Objective To investigate the change of the content of suppressor of cytokine signaling (SOCS-1) in the liver of septic mice and its working mechanism.Methods Adopted Cecalligation and puncture (CLP) to create models of sepsis and divided randomly adult male BALB/c mice into 8 groups,including normal controlled group,sham-operated group,and the killed groups 2 hours,6 hours,12 hours,24 hours and 48 hours after operation.After extracting the RNA and protein from the liver tissue of the mouse groups,reverse transcription polymerase chain reaction (RT-PCR) was adopted to determine the relative content of SOCS-1 mRNA in the tissue,Western blot was adopted to determine the relative content of protein and the SPSS statistics software was adopted to calculate the correlation.Then observed the pathological change of liver tissues,and detected SOCS-1 protein expression by immunohistochemistry.Results After CLP suergery,the expression of SOCS-1 on gene degree in the liver and the expression of SOCS1 on protein degree in the liver increased rapidly at the 6th hour ( P < 0.05 ),with the former reaching peak ( P < 0.05 ) at the 24th hour and the latter remaining high all the time.There were pathological changes such as fatty degeneration and necrosis in the septic liver tissue,hepatic SOCS-1 protein expression could be detected by immunohistochemistry.Conclusions CLP induced sepsis could lead to the increase of the expression of SOCS1 in the liver.
8.Analysis of Interleukin-1 Recepetor Antagonist Gene Polymorphisms of Children with Febrile Seizwres

Objective To evaluate the relationship between children interleukin-1 receptor antagonist gene(IL-1RN) polymorphisms and susceptibility to febrile seizures(FS).Methods Matching case-control study was carried out,blood samples from 52 patients with FS and 53 healthy children were collected.Genomic DNA was extracted and examined by polymerase chain reaction(PCR) amplification for judging IL-1RN genetype.Results Only Ⅰ/Ⅰ,Ⅰ/Ⅱ,Ⅱ/Ⅳ genetypes of IL-1RN were found in the surveyed Chinese population of Han nationality in Guangdong Province.No significant differences of these genotypes were observed in patients and controls.But the allele frequencise of Ⅰ,Ⅱand Ⅳ in patients were 95.2%,3.8% and 1.0%,respectively,while these frequencise in controls were 84.9%,10.4% and 4.7%,respectively.The allele frequency of IL-1RNⅠin patients was obviously higher than that in controls(P

OBJECTIVETo investigate the effects and underlying mechanism of notoginsenoside R1 on amyloid-β (1-42) (Aβ(1-42)) induced mitochondrial apoptotic death in SH-SY5Y cells.
METHODCell viability was assayed by MTT, apoptotic rates were analyzed with PI/Annexin V flow cytometry, Bax and Bcl-2 expression were detected with Western blotting, enzymatic activity of caspase-3, caspase-8 and caspase-9 were measured by ELISA assay.
RESULTThe 6.25-100 nmol x L(-1) of notoginsenoside R1 attenuate Aβ(1-42) induced apoptotic death of SH-SY5Y in dose dependent manner. The ratio of Bcl-2/Bax was elevated in SH-SY5Y with notoginsenoside R1 treatment. Caspase-3 and caspase-9 were activated with notoginsenoside R1 treatment while caspase-8 was not affected.
CONCLUSIONNotoginsenoside R1 could protect SH-SY5Y cells from Aβ(1-42) induced apoptosis via mitochondria related apoptotic pathway.

Sailonggu, a traditional Chinese medicine is whole skeleton of Myospalax baileyi, which is a kind of animal of rodent from Qinghai-Tibet Plateau of China. Osteon Myospalacem Baileyiis the first category medicinal materials of China Food and Drug Administration. For better quality control, a method of the morphological identification of Osteon Myospalacem Baileyi was established by means of studying characteristics of the animal skeleton, it's microscopic characteristics of powder, and literatures comparison. The characteristics of Osteon Myospalacem Baileyi were observed and recorded in detail and marked by number, which could be used for identifying crude drug of Osteon Myospalacem Baileyi efficiently.
