1.Family History, Psychosocial Factors of Parkinson's Disease--a Case-control Study in Linxian County

Objective:To investigate the relationship of family history , psychosocial factors of PD (Parkinson's Disease) in a rural population in north China. Methods: A case control study was conducted in rural population in China. Cases (n=77) have been diagnosed by a two-phase screening by neurologists and interviewers in May, 2000. Controls (n=154), randomly selected from the same cohort, who were matched with patients (control/cases ra-tio=2) for sex, age (?5 years). Information of participants' characteristics came from the interview in 2001. Data was analyzed by using conditional logistic regression by SAS 6.12. Results: Family history (OR=13.54) .negative life events (OR=3.02), interpersonal communication (OR=2.10) were associated with a significantly elevated risk for the onset of PD. Conclusion: These results indicate that family history, psychosocial factors were risk factors of PD.

3.Discrimination method of large log-likelihood study in differential diagnosis of pulmonary diffuse mild micronodule

Objective To analyze HRCT and thin-slice CT scan findings in 150 patients with pulmonary diffuse mild micronodule, and to find the features with the purpose of identifying random micronodule, peri-lymphatic micronodule, and centrilobular micronodule. Methods The useful features in 150 patients with pulmonary diffuse mild micronodule were translated into scores by means of discrimination method of large log-likelihood to identify the micronodular category. Results The accuracy of diagnosis was 94.0% for random micronodule, 76.0% for peri-lymphatic micronodule, and 90.0% for centrilobular micronodule. Conclusion HRCT and thin-slice CT scans were helpful in differential diagnosis of pulmonary diffuse mild micronodule. The discrimination method of large log-likelihood was propitious to diagnosis and differential diagnosis.

Objective To assess the effectiveness and safety of acupuncture and moxibustion for treatment of vertebrobasilar insuffciency (VBI). Methods Randomized controlled trails (RCTs) of VBI treated by acupuncture and moxibustion were collected. 8 RCTs were included and assessed. The data was statistically analyzed by Meta-analysis and Funnel plot analysis. Result 8 RCTs were all trails of low methodological quality. 5.Experimental study of the effect of RNAi to silence STAT3 gene expression on the growth of T24 and 5637 bladder cancer cells

Objective Signal transducers and activators of transcription 3 (STAT3) silenced by RNA interference (RNAi) technique were used to induce the apoptosis and growth inhibition in T24 and 5637 bladder cancer cells. Methods Three recombinant plasmids pGenesil-1-shRNA-STAT3 was constructed and transfected into T24 and 5637 cells. The expression of STAT3 gene was detected by RT-PCR and Western blotting. FCM was used to observe the apoptosis in T24 and 5637 cells. Results pGenesil-1-shRNA-STAT3 was successfully constructed, and transfected into T24 and 5637 cells. RT-PCR and Western blot analysis demonstrated that pGenesil-1-shRNA-STAT3 could significantly inhibit the expression of STAT3 in T24 and 5637 cells; FCM results show that it could suppress the growth of 1'24 and 5637 cells. Conclusion pGeneSiI-1-shRNA-STAT3 could significantly inhibit STAT3 expression, suppress the growth of T24 and 5637 cells.
6.Cloning and expression of human cytomegalovirus UL123 gene exon 2,3 in bacterial two-hybrid bait plasmid

Objective To clone and express HCMV UL123 gene exon 2,3(ie1-exon2,3) in bacterial two-hybrid bait plasmid,and identify its self-activation property.Methods HCMV ie1-exon2,3 carried on pTWIN1/ie1 recombinant was amplified by PCR and cloned into the pBT plasmid,which was transformed into E.coli XL1-Blue MRF' Kan host strain.The positive recombinant was identified by PCR,restriction enzyme digestions and sequencing analysis.The verified plasmid was transformed into bacterial two-hybrid system reporter strain.The soluble fusion protein was analyzed by SDS-PAGE and Western blot.The self activation effect of the recombinant was then tested.Results Bacterial two-hybrid pBT/ie1-exon2,3 bait plasmid was successfully constructed.The corresponding soluble fusion protein rIE1-N_(85)/?C1 was expressed in bacterial two-hybrid system reporter strain,and didn't show self-activation property.Conclusion Bacterial two-hybrid pBT/ie1-exon2,3 bait plasmid without self-activation property was successfully constructed,and it can be used to screen the library of fetal brains.
7.Study on scavenger receptor A from mouse peritoneal macrophages

AIM: To study the role of scavenger receptor A(SR A) in the uptake of oxidized low density lipoprotein(OxLDL) in mouse peritoneal macrophages(MPM). METHODS: Comparing the difference of the uptake of OxLDL in SR A-deficient and wild-type MPM. RESULTS: The results showed that the binding of OxLDL wasn't apparently reduced in SR A-deficient MPM. The association of OxLDL was reduced by 35.8% and degradation of OxLDL was reduced by 42% in SR A-deficient MPM compared with those in wild-type MPM. CONCLUSION: Studies showed that SR A didn't play an important role in the uptake of OxLDL in MPM. Approximately 70% of the uptake of OxLDL in macrophages is attributable to non-SR A receptor.
8.Percutaneous screw fixation of scaphoid fractures aided by computerized imaging processing: preliminary report

Objective To evaluate the preliminary clinical outcome of the screw fixation of minimally displaced scaphoid fracture assisted by computerized imaging reconstruction and preoperative planning system.Methods From March 2013 to Feburary 2014,Five screws on five patients were treated.We calculated and designed the position of scaphoid screw guide wire and ensured the guide wire centrally placed in the scaphoid by using the computerized imaging reconstruction and preoperative planning system software.The appropriate length of the screw was measured too in the software.We also identified the entry and exit points of the guide wire on the custom orthoses splinting the wrist joint.Five patients with scaphoid fractures were treated with percutaneous screw fixation under the guidance of a C-arm fluoroscopy and a guide wire were inserted based on the entry and exit points calculated preoperatively.All of the scaphoid fracture was nondisplaced or less than 1 mm.The modified MAYO wrist scoring system was used to evaluate the pain,function and range of motion for the wrist joint during follow up.Results Total surgical time was 28.8 min (25-39 min)and Fluoroscopy time was 18.4 s (11-23 s).The fracture healing was observed at routine 8 weeks follow up.No complication such as infection or fixation failure was reported.Modified MAYO wrist score was 83.8 (81-88)at average followup at 15 months (6-12 months).Conclusion Computer assisted percutaneous screw fixation is an effective method for the treatment of nondisplaced scaphoid fractures.It leads to significantly reduced guidewire placement time and radiological exposure time while provides accurate placement of the screw.
9.Prostaglandin E2 receptor subtype 3?siRNA reduces the mesangial cell damage induced by TGF ?β1 through inhibiting MAPK pathway in mice

Objective To investigate the effects and mechanisms of prostaglandin E2 receptor subtype 3 (EP3) on transforming growth factor β1 (TGF-β1)-induced mouse mesangial cells damage. Methods Primary mouse mesangial cells were separated and cultured. Three siRNAs were synthesized and transfected into mesangial cells for silencing EP3 by LipofectamineTM 2000 and the best one was chosen. MCs were grouped into: (1)control group; (2)TGF-β1 (10 μg/L) group; (3)NC-siRNA plus TGF-β1 (10 μg/L) group; (4) EP3-siRNA group; (5)EP3-siRNA plus TGF-β1 (10 μg/L). Then the proliferation of MCs was evaluated by CCK-8 assay. The expression of PGE2 and cAMP in cell supernatant were detected by ELISA. The mRNA and protein expression of fibronectin (FN), connective tissue growth factor (CTGF), cyclooxygenase-2 (COX2), membrane-bound prostaglandin E2 synthase 1 (mPGES1) were detected by real - time quantitative PCR and Western blotting. The phosphorylation of p38 MAPK and ERK1/2 was decected by Western blotting. Results Compared with control group, the cell proliferation induced by TGF-β1 was increased (P<0.05), the expression of PGE2 and cAMP were improved, mRNA and protein expression of FN, CTGF, COX2 and mPGES1 were up-regulated (all P<0.05). Compared with TGF-β1 group, the cell proliferation in EP3-siRNA plus TGF-β1 group was reduced, the expression of FN, CTGF, COX2 and mPGES1 mRNA and protein were downregulated (all P<0.05), the phosphorylation of ERK1/2, p38 MAPK were also declined (P<0.05). Conclusion EP3-siRNA may reduce TGF-β1-induced cell damage through upregulating the expression of cAMP, repressing the activity of ERK1/2 and p38 MAPK, inhibiting the expression of COX2 mPGES1 and PGE2 by feedback, then decreased the expression of FN and CTGF.
10.Laparoscopic repair of esophageal hiatal hernia.

This study was to appraise safety and feasibility of laparoscopic approach and investigate the clinical effects of laparoscopic tension-free repair of esophageal hiatal hernia using mesh. From August 2006 to July 2009, 24 patients with esophageal hiatal hernia underwent laparoscopic repair. Twenty-three patients received laparoscopic tension-free repair using mesh, at the same time, Toupet or Dor partial fundoplication was performed. One patient was converted to open surgery. The average operating time was 90 min (70-210 min) and the blood loss was between 10-110 mL. There was no death. The mean postoperative hospital stay was 5 days (3-30 days). During a follow-up period of 12-20 months (mean 15 months), there was no recurrence of the hernia, and no complication with use of mesh. The present study suggested that laparoscopic approach was secure and minimally invasive operation for esophageal hiatal hernia and the use of mesh could reduce recurrence rate.
