1.Preparation of nimodipine-loaded PLEG copolymer nanoparticles and quality evaluation

Objective To improve the poor water-solubility of nimodipine,nimodipine-loaded PLEG copoly- met nanopartieles were prepared to avoid the sedimentation of its crystal from water,which could also shun the blood vessel stimulation caused by ethanol,a cosolvent,in the injection.Methods Solvent evaporation method was used to prepare water-dispersing nimodipine-loaded nanoparticles,whose lyophilized powder was further prepared,with PLEG2000 as drug carrier.Particle size,Zeta potential,embedding ratio,drug loading,shape and redispersion ability of the lyophilized powder were determined.Results Concerning the lyophilized nanoparticle powders,the particle sizes were 136nm,the Zeta potentials were -29.90mV,the embedding ratio was 67.9% and the drug loading was 6.17%.Conclusion Prepared nimodipine-loaded nanoparticles solved the problem of poor water-solubility of ni- modipine basically.It also avoided the recrystallization process of the drug and reduced the blood vessel stimulation caused by ethanol in the injection.
2.Diagnosis of diffusion-weighted imaging in cervical lymph nodes lesions

Objective To investigate the diagnosis of diffusion-weighted imaging (DWI) in cervical lymph nodes lesions.Methods Twenty patients with metastatic lymph node,10 patients with tuberculosis lymph node and 10 volunteers were underwent both conventional MRI and DWI,and the ADC values of lymph nodes' solid components were measured.The difference of ADC values between three kinds of lymph nodes were analyzed.Results ADC values of metastatic lymph nodes was (0.842 ± 0.156) × 10-3 mm2/s,ADC values of tuberculosis lymph nodes was (1.249 ± 0.142) × 10-3 mm2/s,and ADC values of normal lymph nodes was (1.291 ± 0.176) × 10-3 mm2/s.There was significant difference between ADC values of metastatic lymph nodes and with the two later kinds of lymph nodes (P< 0.01),and no difference in the two later kinds of lymph nodes (P > 0.05).Conclusion ADC values with features of conventional MR is helpful for differentiate metastatic lymph node,tuberculosis lymph node and normal lymph nodes.
3.Studies on Establishment of Co-expression System and Function of Human Complement Regulatory Proteins MCP and CD59

Recombinant expression vector pcDNA3-MCPCD59DP containing human membrane complement regulatory proteins(hCRPs) MCP and CD59 cDNA was constructed successfully by using two independent promoters.After transfected into NIH3T3 cells with calcium phosphate-DNA precipitate method,NIH3T3 pcDNA3-MCPCD59-DP transfectants were obtained by G418 selection.Extraneous genes integration was identified by PCR.The co-expression of human CD59 and MCP at both mRNA and protein levels was confirmed by using RT-PCR and Western blot analysis.Human MCP and CD59 cDNA were integrated in NIH3T3 pcDNA3-MCPCD59-DP genomic DNA after continuous 30 times passages,indicating that NIH3T3 pcDNA3-MCPCD59-DP were stable cell lines.Human complement-mediated cytolysis assays showed that NIH3T3 cells transfected stably with pcDNA3-CD59,pcDNA3-MCP,and pcDNA3-MCPCD59-DP were protected from C-mediated damage and co-expressed human MCP and CD59 provided more excellent protection against C-mediated attack as compared with either CD59 or MCP expressed alone.The dicistronic vector represents an effective and efficacy strategy to overcome C-mediated damage and has potential therapeutic value for effectively controlling complement activation and finally for preventing hyperacute rejection in clinical gene therapy.
4.Variation in genes related to lipid metabolism and the susceptibility of nonalcoholic fatty liver disease in obese children

Objective To explore the association between variation in genes related to lipid metabolism and the susceptibility of nonalcoholic fatty liver disease (NAFLD). Methods Obese children with fatty liver aged 6~18 years old were included. All of them got ultrasonic testing, routine examination and biochemical detection. In addition, the DNA of peripheral blood was extracted and the 36 target genes related to lipid metabolism were detected by next generation sequencing. Results In 368 obese children who met the inclusion criteria, 183 children (49.7%) were detected to have NAFL . 100 children with NAFLD and 100 children without NAFLD were randomly selected from obese children. The levels of body mass, waistline, alanine aminotransferase (ALT), triacylglycerol (TG), cholesterol, low density lipoprotein (LDL) and apolipoprotein B (ApoB) in NAFLD children were all higher than those in non-NAFLD children, and there were significant differences (P all<0.05). However, the levels of aspartate aminotransferase (AST), alkaline phosphatase (ALP), apolipoprotein A1 (ApoA1), fasting blood glucose, insulin, high density lipoprotein (HDL), free fatty acid (FFA) and insulin resistance index (HOMA-IR) were not significantly different between the two groups (P all>0.05). The levels of bilirubine in the two groups were within normal range. Logistic regression analysis showed that the genes of MTTP rs2306986 (OR=2.70, 95%CI: 1.38~5.27) and MTTP rs3792683 (OR=7.34, 95%CI: 2.04~26.50) that encode microsomal triglyceride transfer protein (MTTP or MTP), and the mutation of rs738409 (OR=2.11, 95%CI:1.31~4.48) in gene PNPLA3 that encode patatin-like phospholipase domain-containing protein 3 are the independent risk factors for the occurrence of the disease. Conclusion Genovariation of MTTP rs2306986, MTTP rs3792683, and PNPLA3 rs738409 may increase susceptibility to NAFLD in children.
5.A comparative analysis on the effect of laparoscopic repair of gastric perforation with open surgery on pulmonary function

Objective To compare the effect of traditional open surgery and laparoscopic repair of gastric perforation on postoperative pulmonary function.Methods Fifty patients were divided into two groups according to therapeutic method.Group A underwent traditional open gastric perforation repair(n=25).Group B underwent laparoscopic repair of gastric perforation(n=25).The pulmonary functions were examined at 1 day,3 days and 7 days postoperatively.Results All patients had different degrees of postoperative pulmonary function decline.After operation,the forced vital capacity(FVC)and forced expiratory volume in 1 second(FEV1)and vital capacity(VC)in group A were significantly lower than those in preoperation(P<0.05).The patients in laparoscopic operation group decreased significantly only on the first day postoperative.The postoperative pulmonary function of patients undergoing open surgery was significantly lower than that of patients undergoing laparoscopic surgery(P<0.05).Conclusion Laparoscopic repair of gastric perforation has less damage to the lung function than traditional open gastric perforation repair.Laparoscopic surgery plays an important role in the recovery of postoperative pulmonary function.

8.Preparation and identification of antibodies against ADMA

Objective To prepare and identify the monoclonal antibody against asymmetric dimethylarginine (ADMA) and to do further research on clinical application .Methods The short immune peptide antigen was synthetized by solid phase technology . Splenocytes from the Balb /c mice immunized by synthetized antigen were fused with myeloma cells SP 2/0 for producing hybrido-ma .Hybridoma cell line secreting antibodies against ADMA was sifted by indirect ELISA and limiting dilution assay .The specificity of monoclonal antibodies against human ADMA were evaluated with Western blot and ELISA .Results Two hybridoma cell lines stably secreting monoclonal antibodies against ADMA were developed and named 22-1-1and 38-1-6 respectively .By applying West-ern blot and ELISA ,the results indicated that all monoclonal antibodies raised could specifically react with ADMA .Conclusion The success in the production of ADMA monoclonal antibody establishes foundation for application in the practice of clinical labora-to ry .
9.Effect of Human Feeders on Growth of Human Embryonic Stem Cells

[Objective] The aim was to choose the best feeder layer by observing the effects of various human feeders supporting human embryonic stem cells (hESCs), and to probe the correlation between the levels of basic fibroblast growth factor (bFGF) secreted by feeders and the growth of the hESCs. [Methods] The primary cells from various tissues were cultured, including foreskin, stromal endometrium, villus, adult fallopian tubal, fetal skin, fetal muscle and mouse embryonic fibroblasts (MEFs). The hESCs were transferred to various feeders, and then the best condition was probed, which was based on the feeder density and the time of mitomycin-C acting on the feeder. Comparing the characteristics of the hESCs, the best feeder was chosen of all kinds of feeders from various tissues that support the hESCs. The level of bFGF secreted by various feeders was detected using ELISA. [Results] All of tested feeders could support the hESCs growth for over 10 passages in the culture, especially the foreskin and the adult fallopian tubal. The density of feeders was related with the morphology and the differentiation rate of the hESCs. According to the characteristics of feeder, the feeder ranking was as follows: foreskin, stromal endometrium, villus, adult fallopian tubal, fetal skin and fetal muscle. Based on the characteristics of the hESCs, the order of feeders was: foreskin, adult fallopian tubal, stromal endometrium, villus, fetal muscle and fetal skin. The levels of bFGF (pg·10~(-5)·mL~(-1)) secreted by various feeders were as follows: adult fallopian tubal (13.23±3.39), foreskin (1.99±0.17), villus (1.40±0.17), fetal muscle (2.02 ±1.59), stromal endometrium (0.38±0.28), and fetal skin (0.29±0.29). [Conclusion] The foreskin and the adult fallopian tubal could support the hESCs better than others though all of them could;do, especially the, foreskin. The bFGF that secreted by the adult fallopian tubal was the highest of all. The correlation was not obvious .between the levels of bFGF secreted by feeders and the growth of the hESCs.
10.Clinical analysis of tracheotomy of severe craniocerbral trauma in 42 cases

Objective To explore the significance, indicatinos and complications of emergency tracheotomy for patients with severe craniocerbral trauma. Methods The clinical data of 42 patients with severe craniocerbral trauma were retrospectively analyzed. Results Of them, 20 were basically cured (48%), 12 had improvement (29%) ,3 were released from the hospital(7 % ), 1 survived(2 % ) in a vegetative state,and 6 died(14%). Conclu- sion Emergent tracheotomy should be performed on patients with severe craniocerbral trauma to resolve respiratory tract obstruction, improve air current, prevent cerebral anoxia, prevent and treat pulmonary infection, increase the therapeutic efficacy.
