2.Determination and Consistency Analysis of Particle Size Distribution of Budesonide Nasal Spray

OBJECTIVE:To establish the method for the determination of particle size distribution of Budesonide nasal spray, and to analyze the consistency of particle size distribution of spray samples. METHODS:Water was used as dispersant for mixing and dispersing(1800 r/min). The particle sizes [d(0.1),d(0.5),d(0.9)] corresponded to accumulative particle size of 10%,50%and 90%were used as characteristic value. The distribution of granularity was determined by laser scattering method. The consisten-cy of particle size distribution of samples from 2 manufacturers (A,B) were analyzed among different batches or same batch of same manufacturer by SAS 9.3 statistical software. RESULTS:The mean values of d(0.1),d(0.5) and d(0.9) were 3.96 μm, 29.58 μm and 67.10 μm in manufacturer A. The mean values of d(0.1),d(0.5)and d(0.9)were 2.00 μm,7.53 μm and 28.51 μm in manufacturer B. By analysis,there was great difference in particle size of samples from 2 manufacturers. The particle size of the samples from manufacturer A were larger than that of manufacturer B. The consistency among different batches from manufacturer B was better,and the consistency among same batch were all good from 2 manufacturers. CONCLUSIONS:The established meth-od is suitable for particle size distribution of Budesonide nasal spray and the consistency analysis of particle size distribution.
3.Allergic rhinitis treated with acupuncture and the triple-strong stimulation therapy at dazhui (GV 14): a randomized controlled trial.

OBJECTIVETo evaluate the impacts on the short-term efficacy and the long-term prevention of recurrence of allergic rhinitis treated with the triple-strong stimulation at Dazhui (GV 14) so as to provide the convenient and long-term effective therapy of acupuncture and moxibustion for allergic rhinitis.
METHODSOne hundred and twenty cases of allergic rhinitis were randomized into an acupuncture group, an acupuncture + medication group and a triple-strong stimulation group, 40 cases in each one. In the acupuncture group, acupuncture was applied at Dazhui (GV 14), Fengchi (GB 20), Baihui (GV 20), Yintang (GV 29) and the others, stimulating with reinforcing manipulation for the deficiency and reducing manipulation for the excess, once every day. In the acupuncture + medication group, on the basis of acupuncture therapy, claritin (loratadine tablets) was supplemented for oral administration, 10 mg, once every two days, continuously for 30 days. In the triple-strong stimulation group, on the basis of acupuncture therapy, the strong needling, strong cupping and strong moxibustion were applied at Dazhui (GV 14). This combined therapy was given once every day in the first 3 days and once every two days afterwards. The 10 day treatment made one session, at the interval of 3 days between the sessions and totally 3 sessions were required in the three groups. Separately, before treatment, after treatment and in 6 months after treatment, the changes of symptom and physical sign score and value of single item symptom including nasal itching, nasal blockage, sneezing and rhinorrhea were observed in the patients of the three groups. And the long-term clinical efficacy was compared among the three groups.
RESULTSThe symptom and physical sign score and the value of single item symptom were all reduced in the three groups after treatment and in 6 months after treatment (P < 0.001, P < 0.01, P < 0.05). The results in the triple-strong stimulation group were superior to the other two groups (all P < 0.05). In the triple-strong stimulation group, the total effective rate was 92.5% (36/40) in the follow-up of 6 months after treatment, which was better than 60.5% (23/38) in the acupuncture group and 69.2% (27/39) in the acupuncture + medication group (both P < 0.01).
CONCLUSIONThe combined therapy of acupuncture and the triple-strong stimulation at Dazhui (GV 14) achieves the reliable and effective result in the clinical treatment of allergic rhinitis and displays the good role on the prevention from long-term recurrence.

4.A preliminary study of serum-deprivation-induced release of S100A13 and flbroblast growth factor 1 from thyroid cancer TT cell

Objective To investigate the effect of serum-deprivation on the changes of [ Ca2+ ] i and the protein release of S100A13 and fibroblast growth factor 1 ( FGF-1 ) in thyroid cancer TT cell,and to reveal the role of Ca2+in the protein release of S100Al3 and FGF-1.Methods The protein expressions of FGF-1 and S100A13 in TT cells under serum-deprivation were detected by Western blot.The released FGF-1 protein from TT cells in the supernatant fluid was detected by ELISA.Realtime dynamic examinations on the change of 1 h [ Ca2+ ] i in TT cells under serum-deprivation were detected by confocal laser scanning microscopy.Then,the effect of EGTA( 2.5 mmol/ L),BAPTA-AM (2.5 μmol/L)on distributions of the fluorescence of S100AI 3 and FGF-1 in TT cells under serumdeprivation for6 h were detected by indirect immunofluorescence.Results The expressions of FGF-1 and S100A13 in TT cells after serum-deprivation for4 h and 6 h were reduced( P<0.05 or P<0.01 ),but the released FGF-1 protein from TT cells in the supernatant fluid was elevated ( P<0.05 or P<0.01).Confocal laser scanning of Ca2+ imaging indicated that [ Ca2+ ] i of serum-deprivation TT cells maintained the relative stabilization within 23 win,but the rapid rise of [ Ca2+ ] i achieved peak value 1.6 μmol/L after 30 min,and remained stable for about 17 win,and thereafter 40 win slowly dropped to a low level From 40 win to 60 win the [ Ca2+ ] i was about 0.3-0.6 μ mol/L.The average [ Ca2+ ] i was higher than that in normal group,EGTA group,and BAPTA-AM group within 1 h.The protein expressions of S100A13 and FGF-1 did not drop obviously in EGTA group and BAPTA-AM group.Conlusion The release of S100A13 and FGF-1 from TT cell under serum-deprivation is possibly related with the change of [ Ca2+ ]i.Both Ca2+-chelating agents EGTA and BA PTA-AM are able to inhibit the rise of [ Ca2+ ] i and release of S100A 13 and FGF-1 from TT cells under serum-deprivation.
5.Effects of small hairpin RNA-mediated S100A13 gene inhibition on the release of fibroblast growth factor-1 in human thyroid cancer cells

Objective To investigate whether the release of fibroblast growth factor-1 ( FGF-1 ) was changed after inhibition of S100A13 gene (small hairpin RNA, shRNA)and serum-deprivation in human thyroid cancer cells (TT cells ). Methods The S100A13-shRNA pENTRTM/U6 entry vector was transfected into TT cells. The expression of S100A13 mRNA and protein was detected by immunoflurescence, real-time RT-PCR, and Western blot. Then TT cells were treated with S100A13 gene inhibition and serum-deprivation. The changes in release of FGF-1 were detected by indirect immunoflurescence, RT-PCR, and ELISA. Results S100A13 shRNA transfected TT cells (S100A13 RNAi cells)had a reduction of S100A13 gene and protein expression by 80%.Indirect immunofluorescence indicated FGF-1 was mostly localized in the cytoplasm and nucleus of TT cells in primary culture. When serum-deprivation stress was given to TT cells, FGF-1 in cytoplasm almost disappeared in the cells at 6 h. RT-PCR indicated that when serum-deprivation stress was given to TT cells the mRNA of FGF-1 was reduced. ELISA showed that with inhibition of S100A13, the release of FGF-1 was reduced (P<0.05).Conclusion S100A13-shRNA pENTRTM/U6 entry vector transfected TT cells may inhibit the expression of S100A13 and reduce the release of FGF-1.

7.Study on Medication Rules of Treatment of Stroke in Gu Jin Ming Yi Lin Zheng Jin Jian Zhong Feng Juan

Objective To analyze the medication rules of the treatment of stroke in Gu Jin Ming Yi Lin Zheng Jin Jian Zhong Feng Juan; To provide references for the clinical treatment. Methods Prescriptions with confirmed efficacy of famous TCM doctors in the history in Gu Jin Ming Yi Lin Zheng Jin Jian Zhong Feng Juan was searched. Excel2003 was used to establish database to analyze medication frequency. SPSS17.0 statistical software was used to conduct cluster analysis, and tree view was used to show results. Results Totally 112 prescriptions for the treatment of stroke of 36 famous TCM doctors were included in the study, including 204 kinds of Chinese materia medica and 1169 times of medication frequency. The high-frequency medicines (>10 times) were Paeoniae Radix Alba, Achyranthis Bidentatae Radix, Angelicae Sinensis Radix, Pinelliae Rhizoma Praeparatum, and Poria. The high-frequency medicine categories were tonifying deficiency medicine (22.58%), pacifying liver and wind medicine (12.31%), activating blood and dispelling stasis medicine (11.89%), clearing heat medicine (11.46%) and dissipating phlegm, cough and asthma medicine (8.72%). Cluster analysis showed that high-frequency medicine (>10 times) could be clustered as 6 categories. Conclusion Gu Jin Ming Yi Lin Zheng Jin Jian Zhong Feng Juan focuses on tonifying deficiency medicine, accompanied with pacifying liver and wind medicine, activating blood and dispelling stasis medicine, clearing heat medicine and dissipating phlegm, cough and asthma medicine, which can be used to guide clinic.
8.An experimental study on dexamethasone combined with puerarin in treatment of acute paraquat-poisoned rats

ObjectiveTo explore the therapeutic effect and mechanism of dexamethasone combined with puerarin for treatment of paraquat (PQ) poisoned rats.Methods Thirty Sprague Dawley (SD) rats were divided by random number table into 5 groups: control, model, dexamethasone, puerarin and combined groups, 6 rats in each group. The PQ poisoned rat model was established by intraperitoneal injection of PQ 25 mg/kg (1 mL), while in the rat of the control group, the same volume of normal saline was injected intraperitoneally. After 2 hours, in the rat of dexamethasone group, 20 mg/kg dexamethasone in 1.5 mL was injected intraperitoneally, and in the rat of puerarin group, 100 mg/kg puerarin in 1.5 mL was injected intraperitoneally. In the rat of combined group, 20 mg/kg dexamethasone combined with 100 mg/kg puerarin in a total amount of 1.5 mL was injected intraperitoneally. In the control group and model group, 1.5 mL of normal saline was injected intraperitoneally. The above mentioned treatments were repeated once more 26 hours later. In 26-50 hours after modeling, urine was collected, and urine N-acetyl-beta-D-amino glycosidase enzymes (NAG) was tested; at the 50th, abdominal aortic blood was collected to test oxygen partial pressure (PaO2). The lung index, kidney index, and the levels of malonaldehyde (MDA), superoxide dismutase (SOD) and myeloperoxidase (MPO) were measured in the left lung and kidney tissue homogenates separately; the right lung and kidney were
harvested to observe pathological changes under light microscope. Another 30 SD rats were treated the same as above measures but no sacrifice to observe 30-day survival rate in each group.Results The lung index, kidney index, NAG in urine, MDA and MPO levels in lung and kidney tissue homogenates in model group were significantly higher than those in the control group [lung index: 9.80±1.83 vs. 4.97±1.14, kidney index: 9.40±1.32 vs. 7.01±0.32, NAG (U·kg-1·day-1): 1.93±0.18 vs. 0.41±0.03, MDA of lung (nmol/mg): 1.04±0.15 vs. 0.28±0.10, MDA of kidney (nmol/mg): 1.39±0.16 vs. 0.66±0.13, MPO of lung (U/g): 1.14±0.08 vs. 0.81±0.06, MPO of kidney (U/g): 0.88±0.08 vs. 0.52±0.12]; while PaO2 [mmHg (1 mmHg = 0.133 kPa): 59.83±4.40 vs. 97.00±2.83] and SOD (U/mg): lung was 27.38±3.48 vs. 86.88±5.88; kidney was 24.18±3.74 vs. 57.86±6.14) were obviously lower than those in control group (allP < 0.05). After drug treatment was given, lung index (7.21±1.87), urine NAG (1.01±0.21) and MDA (lung was 0.49±0.09, kidney was 0.85±0.08), MPO (lung was 0.97±0.07, kidney was 0.68±0.10) in the puerarin group were significantly lower than those in model group, while PaO2 (82.17±5.38), SOD (lung was 68.99±6.51, kidney was 37.90±3.62) were remarkably higher than those in model group (allP < 0.05). The improvement in the indexes related to the kidney injury was not obvious in dexamethasone group and in the dexamethasone combined with puerarin group, while the lung and kidney pathological changes were lesser in extent in each of the above two treatment groups than those in model group. The 30-day survival rate in model group was significantly lower than that in control group (0% vs. 100.0%);the 30-day survival rates in control group, dexamethasone group, puerarin group and combined group were remarkably higher than those in model group (100.0%, 16.7%, 50.0% and 33.3% vs. 0%, allP < 0.05). But there were no statistical significant differences in the survival rates among those treatment groups (P > 0.05).Conclusions Dexamethasone can improve the prognosis of rats with acute paraquat intoxication, it can provide lung protection markedly, but cannot provide significant protective effect on kidney; puerarin has therapeutic effect on rats with acute PQ poisoning, it can provide not only lung protection but also kidney protection. The effect of treatment with dexamethasone combined with puerarin is not superior to that by using dexamethasone or puerarin alone.

Increased recognition of human parvovirus B19,as a significant human pathogen has resulted in intensive researches to understand the pathogenesis of B19 infection,to elucidate the nature of Th1-mediated cellular immune response,to improve diagnostic strategy that is deployed to detect B19 infection and blood-product contamination,and to lay a foundation that should contribute to the development of an effective vaccine to prevent B19 infection.In this review,the biologic characteristics and the pathogenesis of human parvovirus B19,and B19-related manifestations as well as laboratory diagnostic methods for B19 infection were comprehensively discussed.
