1.Histologic change in human cytomegalovirus-infected explants of first trimester human placenta and expression of human cytomegalovirus gene

Objective To observe histologic changes of human cytomegalovirus(hCMV)-infected explants of first trimester human placenta and expression of hCMV gene in the hCMV-infected explants, and investigate the mechanism of intrauterine transmission of hCMV from mother to fetus.Methods The first trimester placenta explants cultures were carried out, and they were infected with hCMV for 10 days. The expression of hCMV immediate early protein(IEP) 72-IEP86 were determined using indirect-immuno fluorescent assay, and in situ hybridization method was used to examine the hCMV late gene (LG)mRNA. For histologic evaluation of morphological changes in villi, transmission electron microscope was used.Results (1) Typical hCMV-induced lesions bearing hCMV IEP72-IEP86 were consistently localized in the trophoblast of covering placenta villi, interstitial cell and vascular endothelia cell 12 hours after infection, and were predominant in cytotrophoblast. (2) Replication of hCMV in placenta explants culture occurred from 12 hours to 24 hours and disappeared since 48 hours after infection with different concentrations of hCMV when examined by in situ hybridization. (3) Tissue integrity and viability of first trimester placenta explants were obtained in culture for 10 days and then explants were infected with different concentrations of hCMV 100 tissue culture infectious doses(TCID_ 50 ),200 TCID_ 50 and 300 TCID_ 50 , the progression of the infection was observed in the tissue that maintained its normal cellular organization under light microscope. But typical inflammation of cellular organization was observed under transmission electron microscope. Conclusions (1) A flash replication of hCMV in placental explants culture occurs; IEP72-IEP86 may be in intrauterine infection of hCMV for a long time. (2) There are pathological ultrastructure changes in hCMV-infected explants.
2.Relationship between Stress and Dehydroepiandrosterone Sulfate in Umbilical Blood of Pre-birth

Objective:To explore the correlation of stress, coping style and the concentration of dehydroepiandrosterone sulfate in umbilical blood between preterm birth and normal birth.Methods:46 with preterm birth and 42 normal birth controls were assessed with Life Event Scale (LES) and Defense Style Questionnaire (DSQ) at 28th-week of pregnancy. The concentrations of DHEA-s in umbilical blood were determined by ELISA for the two groups. 4.Application of magnetic resonance imaging in diagnosis and treatment of pelvic neoplasms

Objective To study the value of magnetic resonance imaging(MRI) in the diagnosis and treatment of pelvic neoplasms. Methods The patients who were suspected of malignant pelvic neoplasms were performed MRI.The surgical approach was determined according to the MRI results.The MRI results were compared with operation findings and pathological reports. Results The MRI results were consistent with operation findings and pathological reports in 22 cases highly suspected of pelvic neoplasms and 15 cases with abnormal endometrial echo.Therapeutic regimen was designed for 5 cases with uterine cervix cancer according to MRI stages,and favourable surgical curative effects were obtained. Conclusion MRI provides a reliable guideline for surgical approach and may improve the outcomes.
5.Application of acoustically evoked stapedius reflex threshold via cochlear implant for maximum comfortable level determination

OBJECTIVE To investigate the feasibility of determining the maximum comfortabl level by using the acoustically evoked stapedius reflex threshold via cochlear implant. METHODS Eleve cases of Nucleus C124 cochlear implant recipients operated in our hospital were selected(8-39 years old, who can decide TIC value by themselves with the postoperative time ranging from device initiation to 1 year after operation),of those four cases were diagnosed as bilateral large vestibular aqueduct syndrome,1 case as common cavity dysplasia,.neural response telemetry of these five patients with inner ear dysplasia were very high or barely measurable.Real-MCL level and acoustically evoked stapedius reflex threshold under activated condition were measured respectively. The stimulating sound was 110dB of white noise.RESULTS The two group values of ASRT and real-MCL were found to be consistant .There is no significant difference statistically (P>0.05). The thresholds of ASR increase with the increasing of real-MCL at the beginning of device initiation. CONCLUSION The acoustically evoked stapedius reflex threshold may reflect (ASRT) the maximum comfortable stimulation level under activated conditions in patients with cochlear implants.it is simple, reliable and may be used to determine the MCL of child cochlear implanted patients.

10.Design, synthesis and cholinesterase inhibitory activity of quinoline-polyamine conjugates.

A series of quinoline-polyamine conjugates (8a-8n) were designed, synthesized and evaluated as inhibitors of cholinesterases (ChEs). Some of these compounds had potent ChEs inhibitory activity with IC50 values at micromolar range. Compound 8n exhibited the strongest inhibition on acetylcholinesterase (AChE) with an IC50 value of 8.78 micromol x L(-1), and compound 8i showed the most potent inhibition on butyrylcholinesterase (BChE) with IC50 value of 1.60 micromol x L(-1) which was slightly better than rivastigmine. The structure-activity relationship revealed that the chain length of polyamine and linker played important roles for inhibitory activity. Molecular modeling studies showed that 8i targeted both the catalytic active site (CAS) and the peripheral anionic site (PAS) of cholinesterases.
