2.Inhibitory effects of calmodulin antagonist EBB on cultured fibroblast

Inhibitory effects of the O - ( 1 - ethoxvl - butyl ) berbarmine (EBB) on cultured fibroblast was studied by observating calmodulin (CaM ) content of cultured fibroblast with ELISA and DNA content at each phase of cell cycle with flow - cvtometerv. The CaM con-tent in the test group . compared to control . decreased markedly and DNA content increased significantly in the G0+Gi phase but reduced in the S phase. These results suggested that inhibitory mechanism of EBB on fibroblast proliferation may be closely related to CaM decrease in cells.

Objective To analysis the DNA sequence of HEV ORF3 cDNA and express in HepG2 cell .Methods Recombinant vector carrying HEV ORF3 cDNA was constructed and identified by DNA sequencing .Expression of recombinant vector in HepG2 cell was determined by immunofluorescence technique .Results The recombinant vector was constructed and expressed in HepG2 cell successfully .Conclusion The research laid experimental foundation for exploring the biological function of ORF3 protein in the pathogenesis of viral hepatitis E .

5.Effect of anticoagulation combined with hypolipidemic in treatment of diabetic atherosclerosis

Objective To investigate the effect of aspirin combined with atorvastatin in the treatment of diabetic atherosclerosis.Methods 120 patients with diabetic atherosclerosis form August 2014 to February 2016 were selected,and randomly divided into treatment group and control group,60 cases in each group.Control group treated with aspirin enteric-coated tablets,the treatment group treated with aspirin enteric-coated tablets combined with atorvastatin,comparing the two groups before and after treatment of fasting blood glucose,blood lipid related index,carotid intima-media thickness and plaque thickness,and the adverse reactions.Results After treatment, the levels of fasting blood glucose and fasting insulin in treatment group were significantly lower than control group(P<0.05),the carotid intima-media thickness and plaque thickness in treatment group was significantly lower than control group(P<0.05).The levels of TG,TC,LDL-C and HDL-C in treatment group were significantly improved(P <0.05).Conclusion Aspirin enteric coated tablets combined with atorvastatin efficacy significantly in the treatment of diabetic atherosclerosis.
6.Effects of soy bean isoflavone on the expression of VEGF and NOS subtype mRNA in ovaries during perimenopause period in rat

Objective To investigate the effects of soy bean isoflavone(SI)on the expression of VEGF,nitric oxide synthase(NOS)subtype,endothelial NOS(eNOS)and inducible NOS(iNOS)mRNA in ovaries during perimenopause period in rats.Methods A rat model of perimenopause was reproduced by enforced ageing.42 female Wistar rats of 12 months old were randomly divided into five groups:aging model group,low,middle,high doses of SI groups,and nilestriol(NI)group.10 female rats of 3 months old served as control group.The animals in low(50mg/kg),middle(158mg/kg)and high(500mg/kg)dosage of SI groups,and in NI group(0.158mg/kg)were gavaged with corresponding drugs for 8 weeks.The expressions of VEGF and NOS mRNA in ovaries were detected by RT-PCR.The total NOS activity in serum and ovaries was detected by chromatometry.Results Compared with control group,the expressions of VEGF mRNA in ovaries of perimenopause rats increased significantly(P

Objective To compare the changes of serum levels of heme oxygenase‐1(HO‐1) ,C‐reactive protein(CRP) and Chitinase‐3‐1ike protein 1(YKL‐40) in patients with rectal cancer after laparoscopic‐assisted radical operation versus open radical surgery .Methods According the integration and elimination standard ,60 cases of patients with rectal cancer were selected and di‐vided into laparoscopic group (30 cases) and open group (30 cases) ,the concentrations of HO‐1 ,YKL‐40 and CRP in peripheral blood of patients were detected on the first and third postoperative day and preoperative day(P<0 .05) ,and were compared between the two groups .Results The serum levels of HO‐1 ,YKL‐40 and CRP in the laparoscopic group were significantly lower than those in the open group on the first and third postoperative day(P<0 .05) .The serum levels of HO‐1 ,YKL‐40 and CRP were significant‐ly increased after surgery ,and serum levels of HO‐1 and CRP on third postoperative day were significantly increased compared with that on the first postoperative day in the two groups after surgery ,while serum level of YKL‐40 on the third postoperative day was significantly lower than that on the first postoperative day in the two groups after surgery .Conclusion The stress reaction is com‐paratively mild in patients with rectal cancer after laparoscopic assisted radical operation ,and the postoperative period may be shor‐ter in patients treated with laparoscopic assisted radical operation than those treated with open radical surgery .
8.The effect of high-level of glucose on ghrelin expression in human umbilical vein endothelial cells

Objective To investigate the influence of high-level of glucose on the expression of ghrelin in human umbilical vein endothelial cells (HUVECs). Methods After 12 h glucose free culturing, the effects of different concentrations of glucose and different incubation times on expressions of ghrelin were observed in HUVECs. ( 1) The cells were treated with 0, 5, 10, 15, 20 mmol/L glucose for 2 h, then ghrelin mRNA expression levels were detected by RT-PCR, and the protein levels of ghrelin were detected by ELISA. (2) The cells were treated with 10 mmol/L glucose for 0, 0.5, 1, 2, 6, 12 hours, ghrelin mRNA and protein levels were detected respectively. Results (1) The expression levels of ghrelin mRNA and protein decreased along with increased glucose concentrations, which showed no obvious changes when the glucose was above 15 mmol/L. (2) The expression levels of ghrelin mRNA and protein decreased with the prolonged incubation time. But more than 6 h culturing time showed no further effect on reducing the expression level of ghrelin. Conclusion High levels of glucose can inhibit the expression level of ghrelin, which may be one of the mechanisms of atherosclerosis.
