1.Effects of Green Tea Polyphenols on LDLR Function and Potential Mechanism in HepG2 Cells

[Objective] To investigate the effects of the bioactive constituents of green tea polyphenols epigallocatechin gallate (EGCG) on low density lipoprotein receptor (LDLR) function of HepG2 cells.[Methods] The optimal concentration and cell proliferation of HepG2 cells were determined by CCK8 assay,and Western blotting was used to determine LDLR and PCSK9 protein levels,respectively,and LDL uptake in HepG2 cells was detected by fluorescence microscope.[Results] EGCG elevated LDLR protein expression,reduced PCSK9 protein expression and promoted LDL uptake in HepG2 cells.[Conclusion] EGCG may increase LDLR abundance by down-regulating PCSK9 protein and attenuating LDLR protein degradation,which providing a new approach for lipid lowering therapy.
2.Magnetic resonance spectroscopy in prefrontal white matter of bipolar Ⅱ disorder and its relation with executive function

Objective To explore the magnetic resonance spectroscopy characteristics of prefrontal white matter in bipolar Ⅱ disorder and its relation with executive function.Methods Thirty bipolar Ⅱ disorder patients and twenty healthy controls were evaluated with Multi-Voxel proton magnetic resonance spectroscopy (1H-MRS)scans on prefrontal white matter to assess the N-acetyl-aspartate (NAA),Choline (Cho),Creatine (Cr) and myoinositol (MI),and then the ratios of NAA/Cr,Cho/Cr,MI/Cr,NAA/Cho and NAA/Cho + Cr were calculated.All subjects were assessed for executive function using the Wisconsin Card Sorting Test (WCST).Results NAA/Cr in right prefrontal white matter(1.43 ± 0.26) and NAA/Cr,NAA/Cho,NAA/Cho + Cr in left prefrontal white matter (separatelyl.40 ± 0.29,1.13 (1.53,0.24),0.62 ± 0.12) were lower than healthy controls (separately 1.58 ±0.18,1.59,0.23,1.30 (0.53 ± 0.29),0.71 ± 0.08) (P < 0.05).The correct trials and the number of categories of WCST in Bipolar Ⅱ disorder (separately 26.97 ± 8.97,3(6,3)) were less than in healthy controls(separately 36.35±4.85,5 (3,2)),and perseverative errors and random errors were more in patients (separately 12.77 ±5.73,7 (21,5)) than healthy controls (separately 7.35 ± 3.01,4 (13,2)).NAA/Cho and NAA/Cho + Cr in left prefrontal white matter were positively correlated with correct trials,number of categories,and negatively related with perseverative errors (P<0.05).Conclusions Bilateral prefrontal white matter fiber damage occurs to bipolar Ⅱ disorder.And left prefrontal white matter fiber damage in bipolar Ⅱ disorder may lead to executive impairment.
3.Comparative Study on Electroacupuncture Versus Warm Needling for Low Back Pain

Objective To observe and compare the clinical efficacies between electroacupuncture and warm needling in treating low back pain.Method Seventy-eight eligible low back pain patients were randomized into group A of 28 cases, group B of 26 cases, and group C of 24 cases. Group A was intervened by electroacupuncture, group B was by warm needling, and group C was by medication. The short-form McGill Pain Questionnaire, Japanese Orthopaedic Association Scores (JOA), and Oswestry Disability Index were observed before and after treatment, and the therapeutic efficacies were compared.Result In group A, the McGill item scores [Sensory Pain Rating Index (S-PRI), Affective Pain Rating Index (A-PRI)] respectively after 1-week and 2-week treatment as well as in the 1-month and 3-month follow-up were significantly different from that before treatment (P<0.01,P<0.05). In group B and C, the McGill item scores after 2-week treatment and in the 1-month and 3-month follow-up were significantly different from that before treatment in the same group (P<0.01,P<0.05). The JOA and Oswestry scores were significantly changed respectively after 1-week and 2-week treatment and in the 1-month and 3-month follow-up in the three groups compared with that before treatment (P<0.05,P<0.01). After 1-week and 2-week treatment and in the 1-month and 3-month follow-up, the JOA and Oswestry scores in group A were significantly different from that in group C (P<0.05,P<0.01). In the 1-month and 3-month follow-up, the JOA scores in group B were significantly different from that in group C (P<0.05). The total effective rate was 85.7% in group A and 73.1% in group B, both significantly higher than 58.3% in group C (P<0.05). Conclusion Electroacupuncture and warm needling both can produce a significant efficacy in treating low back pain, but warm needling acts comparatively slowly and is less safe.

OBJECTIVETo evaluate the RT-PCR-ELISA method applied for testing live attenuated hepatitis A vaccine titer.
METHODSA solid phase hybridization-enzyme colorimetric detection method was used for detecting specific nucleic acid. Primer labeled with biotin was used to amplify viral gene fragment, then the product was quickly hybridized with the specific probe covalently coupled on DNA-binding microplate wells. Finally, peroxidase-labeled streptavidin was used in colorimetric detection. The results were judged by reading A value. Eleven batches of live attenuated hepatitis A vaccine titer were tested by this method. The results were compared with that of routine cell culture method (CCID50).
RESULTSThe sensitivity was similar to routine cell culture method (P>0.05). This method was convenient, fast and specific.
CONCLUSIONCCID50 method may be replaced by the RT-PCR-ELISA method in evaluating the titer of live attenuated hepatitis A vaccine.

Objective To explore the magnetic resonance spectroscopy characteristics of prefrontal white matter and lenticular nucleus in patients with bipolar Ⅱ disorders.Methods Thirty patients with bipolar Ⅱ disorders and 20 healthy controls,collected in our hospital from September 2012 to April 2013,were evaluated with Multi-Voxel 1H-MRS scans on prefrontal white matter and lenticular nucleus to assess the levels of N-acetyl aspartate (NAA),choline compound (Cho),creatine (Cr) and myo-inositol (mI),and then,the ratios of NAA/Cr,Cho/Cr,mI/Cr,NAA/Cho and NAA/Cho+Cr were calculated.Results Levels ofNAA,NAA/Cr,Cho and mI in the right prefrontal white matter were significantly lower in patient group than those in the normal controls (P<0.05); NAA level,and ratios of NAA/Cr,NAA/Cho,NAA/Cho+Cr in the left prefrontal white matter were obviously lower in patient group than those in the normal controls (P<0.05); NAA and Cho in right lenticular nucleus were lower as compared with those in the healthy control subjects (P<0.05).Conclusion Prefrontal white matter fiber damage and glial cell disfunction occur in patients with bipolar Ⅱ disorder; besides,neuronal loss and disfunction in the right lenticular nucleus exist.

Objective To observe the effectiveness of different administration time and positions of Kaiselu for patients with acute myocardial infarction On prevention of their constipation.Methods 68 acute myocardial infarction patients without three days puzzles stool were randomly divided into two groups f 34 case8 in each group).The patients in the observation group received Kaiselu 40~50 ml without the awareness of defecation,by the anal insertion 7~10 cm,while the control group received Kaiselu 40~50 ml with the awareness of defecation, by the anal insertion 3~4 cm. Results The defecation effect in the observation group Wag significantly higher than the control group(P<0.01).Conclusions It shows better defecation effect for patients with acute myocardial infarction received Kaiselu without the awareness of defecation by the anal insertion 7~10 cm.

BACKGROUND:
It remains unclear whether the outcomes of ST-elevation myocardial infarction (STEMI) patients treated with primary percutaneous coronary intervention (PPCI) during off-hours are as favorable as those treated during on-hours, especially those with a first medical contact-to-device (FMC-to-device) time within 90 min. We aimed to determine whether off-hours admission impacted late outcomes in patients undergoing PPCI and with an FMC-to-device time ≤90 min.
METHODS:
This multicenter retrospective study included 670 STEMI patients who underwent successful PPCI and had an FMC-to-device time ≤90 min from 19 chest pain centers in Beijing from January 2018 to December 2018. Patients were divided into on-hours group and off-hours group based on their arrival time. Baseline characteristics, clinical data, and key time intervals during treatment were collected from the Quality Control & Improvement Center of Cardiovascular Intervention of Beijing by the "Heart and Brain Green Channel" app.
RESULTS:
Overall, the median age of the patients was 58.8 years and 19.9% (133/670) were female. Of these, 296 (44.2%) patients underwent PPCI during on-hours and 374 (55.8%) patients underwent PPCI during off-hours. Compared with the on-hours group, the off-hours group had a longer FMC-to-device time and fewer patients with FMC-to-device time ≤60 min (P < 0.05). During the mean follow-up period of 24 months, a total of 64 (9.6%) participants experienced a major adverse cardiovascular event (MACE), with 28 (9.1%) in the on-hours group and 36 (9.6%) in the off-hours group (P > 0.05). According to the Cox regression analyses, off-hours admission was not a predictor of 2-year MACEs (P = 0.788). Similarly, the Kaplan-Meier curves showed that the risks of a MACE, all-cause death, reinfarction, and target vessel revascularization were not significantly different between the two groups (P > 0.05).
CONCLUSIONS
This real-world, multicenter retrospective study demonstrated that for STEMI patients who underwent PPCI within 90 min, off-hours admission was safe, with no difference in the risk of 2-year MACEs compared with those with on-hours admission.

OBJECTIVETo study the bioactive constituents from tubers of Bolbostemma paniculatum.
METHODCompounds were isolated by extraction and partition as well as several-chromatographic techniques guided with Pyricularia oryzae bioassay method. Their structures were determined on the basis of spectral analysis and chemical evidence.
RESULTBisdesmoside (I) was isolated as active compound causing morphological abnormality of Pyncularia oryzae mycelia and elucidated as 3-0-alpha-L-arabinopyranosyl (1-->2)-beta-D-glucopyranosyl-bayogenin-28-O-beta-D-xylopyranosyl (1-->3)-alpha-L-rhamnopyranosyl (1-->2)-alpha-L-arabinopyranoside.
CONCLUSIONI is a new natural product and exhibited significant cytotoxicity against cancer cell lines K-562 and BEL-7402, but no hemolytic activity to rabbit erythrocytes.

10.Clinical application of three methods for total ear reconstruction.

OBJECTIVETo investigate the indication and results of three methods for total ear reconstruction.
METHODS960 cases of total ear defect were treated with skin expansion and autogenous rib cartilage framework (n = 786), or skin expansion and Medpor framework (n = 150), or ear prosthesis (n = 24). The indication and results of the three methods were analysed.
RESULTSGood cosmetic results were achieved with all three methods. Autogenous rib cartilage framework was suitable for patients under 30 years old with unaffected skin at mastoid region. Medpor framework was good for grown-up, especially over 30 years old, or with mild infection at mastoid region. Patients with severe skin injury at mastoid region or unwilling to accept surgery should choose ear prosthesis.
CONCLUSIONSSatisfactory results can be achieved with suitable methods for total ear reconstruction.
