1.Comparison and selective preference of preparation methods of platelet-rich gel

BACKGROUND:Platelet-rich plasma gel contains some kinds of growth factors, which play important roles in repair of local trauma and bone defects. Platelet-rich plasma gel is now more and more studied as tissue engineering scaffolds, but there are no unified preparation methods.
OBJECTIVE:To summarize the studies about the preparation methods of platelet-rich plasma gel and the growth factors in platelet-rich plasma gel in recent years, and to compare different methods in order to find which one is better.
METHODS:A online search of PubMed database from January 1980 to January 2013 and China National Knowledge Infrastructure from January 1980 to January 2013 was performed with the key words of“platelet-rich plasma, platelet-rich plasma gel, tissue engineering”in English and Chinese, respectively. The studies about detection and preparation methods of platelet-rich plasma were included and those unrelated to platelet-rich plasma were excluded. Final y, 322 articles were gained and 67 articles were included in result analysis.
RESULTS AND CONCLUSION:Al preparation methods have their advantages. We should select the suitable method according to the goal of study. With the further study of platelet-rich plasma, it wil become more and more important for bone defect repair and tissue engineering research.
2.Protective Role of Anisodamine Against Rabbit Liver Injury by Hemorrhage Shock Complicated with Endotoxemia and the Underlying Mechanism

Objective To study the possible protective mechanisms of Anisodamine against liver injury induced by hemorrhage shock(HS). Methods In this study, we established animal models induced by HS and endotoxin, observed the expressions of NF-?B mRNA and heat shock protein 70(HSP 70) mRNA by In-situ hybridization (ISH) in liver kupffercells(KC) and studied the pathologic changes of the liver tissue by microscope. 4.Connection of Matrix Operation Live Telecast System with Telemedicine Education Web

Objective To explore the telemedicine education by using operation live telecast system.Methods Multi-channel video and audio signals were collected by video camera and transmitter phone and were connected with telemedicine education web after matrix treating and controlling.Results Telecommunication of live telecast was realized.Conclusion Operations can be telecast more widely,and the level of telemedicine education is raised.
5.Study of proliferation of chondrocytes co-cultured with platelet rich gel

Objective Preparing platelet rich gel through two-times centrifugal technique and co-culturing chondrocytes with PRG, then observing the proliferation and gene expression of chondrocytes, in order to provide a favorable way to prepare tissue engineering cartilage. Methods Centrifugating venous blood of rabbit through two-times centrifugal technique to obtain platelet rich plasma( PRP) ,then detecting the concentration of various growth factor in PRP. Admixing PRP with chondrocytes of rabbit and activating them with activator. After co-culti-vation,the proliferation of chondrocytes through MTT method and expression of ACAN,CollagenⅡand SOX-9 through realtime-PCR were ob-served,and compared with common cultured chondrocytes. Results The concentrations of PDGF-AB,TGF-β1,IGF-1 and VEGF in PRG were significantly higher than those in blood(P<0. 05). After co-cultivation, the proliferation rate of chondrocytes and the expression of ACAN,Collagen Ⅱ and SOX-9 were significantly higher than that of common cultured chondrocytes(P<0. 05). Conclusion Co-culturing chondrocytes with PRG is able to promote the proliferation and gene expression of chondrocytes. We considered that it is a excellent method to construct tissue engineering cartilage.
6.Effects of dexamethasone sodium phosphate on the proliferation of in vitro cultured rabbit articular chondrocytes

BACKGROUND: Studies have demonstrated that dexamethasone can reduce type Ⅱ collagen and increase type I collagen in articular cartilage surface matrix, but the action mechanism of glucocorticoid to articular chondrocyte proliferation remains unclear. OBJECTIVE: To investigate the effects of dexamethasone sodium phosphate on rabbit articular chondrocytes cultured in vitro. DESIGN: Comparative observation.MATERIALS: New Zealand rabbits, 1 month old, were used for chondrocyte isolation and culture.METHODS: Rabbit articular chondrocytes cultured in vitro were randomly divided into control and experimental groups. Cells of the control group were cultured in DMEM media without dexamethasone sodium phosphate. Cells of experimental groups were cultured in DMEM media with different concentrations of dexamethasone sodium phosphate (0.02, 0.1, 0.5 g/L), respectively. MAIN OUTCOME MEASURES: Primary and passage chondrocytes were observed. Flow cytometry and immunocytochemistry were used to observe the effect of dexamethasone sodium phosphate on cell proliferation and type Ⅱ collagen synthesis. The ultrastructural changes of cultured chondrocytes were observed by transmission electron microscopy. RESULTS: The attachment and proliferation of experimental group chondrocytes was slower than control group. There average gray scale values of the experimental groups were significantly greater than the control group (P < 0.05). The cellular proportions of S phase and G_2+M phase of the experimental groups decreased but the cellular proportions of G0/G1 period increased. Under transmission electron microscope the amount of rough endoplasmic reticulum and mitochondria of the experimental groups decreased. CONCLUSION: Dexamethasone sodium phosphate inhibited articular chondrocyte proliferation, possibly due to the decrease of type Ⅱcollagen and protein synthesis ability of chondrocyte.
7.Clinical evaluation of percutaneous nephrolithotomy in the treatment of renal stone after repeated extracorporeal shockwave lithotripsy

Objective To evaluate the effects and complications of percutaneous nephrolithotomy (PCNL) in the treatment of renal stone after repeated extracorporeal shockwave lithotripsy (ESWL).Methods Forty-four patients who had a history of repeated ESWL (treatment group) and 50 patients with-out surgical intervention (control group) were submited to PCNL,and clinical data was documented in details and analyzed.Results The time to establish access in treatment group and control group was (11.8 ± 4.1) min and (10.9 ± 2.5) min,respectively,and there was no significant difference (t =1.308,P =0.194).The time to extract stone in both groups was (92.0 ± 13.5) min and (66.6 ± 17.6) min,respectively,and there was significant difference (t =7.776,P =0.000).The operative time in treatment group was (113.9 ± 12.0) min,which was longer than that in control group with (87.6 ± 13.6) min (t =8.354,P =0.000).The clearance in both groups was 81.8% and 94.0%,and there was no significant difference (x2 =3.361,P =0.067).The was no death or other severe complication in both groups.Conclusions The operation time in treatment group was longer than that in control group,and there was no significant difference in clearance and complication rate.Thus it was safe and effctive to perform PCNL in these patients with a history of failed repeated ESWL.
8.Continuous curvilinear capsulorhexis involving zonular area in manual small incision cataract surgery

AIM:To evaluate the efficacy and safety of large sized continuous curvilinear capsulorhexis ( CCC ) involving zonular area in manual small incision cataract surgery ( MSICS) .
METHODS:Totally 1 443 cataract patients ( 1 965 eyes ) underwent MSICS, in which large CCC up to 7-8mm were performed . The related conditions such as success rate of performing CCC, capsule rupture, corneal edema and visual acuity after surgery were recorded and statistically analyzed.
RESULTS:In total of 1 965 eyes, 1 942 eyes ( 98. 83%) had successful CCC, 15 eyes (0. 76%) had a radial tear which could not be saved when performing CCC, 8 eyes (0. 41%) had anterior capsule opened with a cystotome or a scissor due to capsule membrane calcification. No posterior capsule rupture occurred, and intraocular lens was transplanted in all cases. Transient corneal edema was noted in 36 eyes (1. 83%). One day after surgery, 1650 eyes (83. 97%) had visual acuity ≥0. 5, 1867 eyes (95. 01%) ≥0. 3. No unstable intraocular lens was noted in all cases.
CONCLUSION:Large CCC technique for MSICS is safe and reliable, not decreasing stability capsular bag and intraocular lens. It is of high originality despite zonular area is involved so that it is worthy of application in basic medical institutions.
9.Design, synthesis and pharmacological investigation of isoindoline derivatives as 5-HT/NE double reuptake inhibitors.

A series of isoindoline derivatives were designed, synthesized, and evaluated for their double inhibitory activities. All of them were new compounds, and their structures were confirmed by 1H NMR and HR-MS. Preliminary in vitro pharmacological tests showed that all compounds exhibited 5-HT or NE reuptake inhibition activity. Among the tested compounds, compound I-3 exhibited potent inhibitory activity against 5-HT and NE reuptake in vitro, and exhibited potent antidepressant activity in vivo. These compounds designed can be further optimized for finding more potent 5-HT/NE dual reuptake inhibitors and antidepressant candidates as well.

10.Related factors of psychological symptoms in males with traumatic urethral stricture

Objective:To explore the related factors of the psychological symptoms in males with traumatic urethral stricture and to test the mediating effect of social support and coping style on traumatic urethral stricture and psychological symptoms.Methods:Totally 43 male patientswith traumatic urethral stenosis and 1 ∶ 1 matched 43 healthy control subjects were selected as the study samples in the Urology Department of a Third Grade Hospital in Beijing.The Symptom Checklist 90 (SCL-90) was used to evaluate the psychological symptoms of the subjects.The Perceived Social Support Scale (PSSS) and Trait Coping Style Questionnaire (TCSQ) were used to assess the social support and coping styles of the subjects.The non-parametric percentile Bootstrap method was used to test the mediating effect of social support and coping style between traumatic urethral stricture and psychological symptoms.Results:The SCL-90 total scores were higher in patients with traumatic urethral stricture than in healthy controls [(2.9 ± 0.6) vs.(2.4 ± 0.7),P < 0.01].The mediating effect of social support and coping style on psychological symptoms was not significant (P > 0.05).Multiple linear regression analysis showed that the SCL-90 total scores were higher in males with traumatic urethral stricture than in normal controls (β =0.55,95% CI:0.18 -0.92).PSSS family support scores were negatively correlated with SCL-90 scores (β =-0.10,95% CI:-0.14--0.06).Conclusion:It suggests that male traumatic urethral stricture may cause psychological symptoms which may be associated with family support.
