1.Transvaginal ultrasound guided methotrexate injection for unruptured early tubal pregnancy

2 000 mIU/ml,combined use of mifepristone(100 mg,twice daily,for 3 days) was employed. Results The treatment succeeded in 28 patients,with a success rate of 87.5%.Acute lower abdominal pain occurred in 4 patients and laparoscopic salpingectomies were carried out.The serum ?-hCG levels decreased by 23%~35% at 3 days after injection,by 38%~62% at 1 week,and to normal levels(≤6.1 mIU/ml) at 2~4 weeks.The mass completed subsided at 30~55 days after injection. Conclusions Transvaginal ultrasound guided methotrexate injection for the management of unruptured early tubal pregnancy is simple,safe, and effective.Further investigations should be carried out in respect of the recovery of tubal anatomy and physiological functions.
3. Preparation of PEGylated graphene oxide nanoparticles loaded with oridonin and its inhibitory effects on human colon cancer

Objective To prepare PEGylated graphene oxide nanoparticles (GO-PEG-NPs) loaded with oridonin (ORI) and investigate its inhibitory effect on human colon cancer. Methods 4-armed PEG was grafted onto GO via an amidation process and GO-PEG was characterized by IR and TGA. Then, ORI, a widely used cancer chemotherapy drug, was absorbed onto GO-PEG via blending. The encapsulation efficiency and drug loading ratio were measured by UV. The cytotoxicity of ORI-GO-PEG, GO-PEG, and free ORI on SW620 and HT29 human colon cancer cells were evaluated using MTT assay. In vivo anti-tumor activity of ORI-GO-PEG-NPs were evaluated in mice bearing SW620 tumor. Results IR and TGA data indicated that 4-armed PEG was successfully coupled with GO and UV data showed that the encapsulation efficiency and drug loading ratio were 95.81% and 48.92%, respectively. Moreover, ORI-GO-PEG-NPs showed good stability in physiological condition. The results of cytotoxicity test indicated that compared to free ORI, the ORI-GO-PEG-NPs exhibited higher cytotoxicity in SW620 and HT29 cells. Meanwhile, it was confirmed that ORI-GO-PEG- NPs could significantly inhibit the growth of SW620 tumor in vivo compared with ORI. Conclusion The obtained ORI-GO-PEG-NPs displayed excellent drug-loading capacity and better tumor inhibitory effect. These results provided the experimental basis for development of anticancer drug delivery system.
4.Changes of Interleukin-6 in Serum and Urine in Children with Primary Nephrotic Syndrome and Their Clinical Significance

Objective To explore the clinical significance of the changes of interleukin-6(IL-6) contents in serum and urine in children with primary nephrotic syndrome(PNS).Methods The contents of IL-6 in serum and urine in 38 cases of PNS were exa-(mined) by ELISA before and after treatment with prednisone 2 mg/(kg?d) for 8 weeks.Results Before the treatment with prednisone,the contents of IL-6 in serum in steroid-responsive group and steroid-resistant group were significantly higher than those in the normal controls(P0.05).In the steroid-responsive group and steroid-resistant group,the levels of IL-6 in urine were significantly higher than those in the normal controls(P0.05).The contents of IL-6 in steroid-resistant group in serum and urine had significant difference in the comparison with the normal controls(P

Objective To explore the effect of preemptive anti-cytomegalovirus therapy on prognosis of patients with acquired immunodeficiency syndrome (AIDS) and cytomegalovirus (CMV) viraemia.Methods A non-random prospective study of male hospitalized patients were carried out who were newly diagnosed as AIDS during July 1,2011 to June 30,2013 in the First Affiliated Hospital of China Medical University.There were 69 patients who were divided into treatment group (57 cases) and control group (12cases).The duration of anti-cytomegalovirus therapy was a week after plasma CMV-DNA becoming undetectable.Baseline age,CMV-DNA,human immunodeficiency virus (HIV)-RNA,CD4+T cells,the rate of pneumocystis carinii pneumonia (PCP) and tuberculosis were evaluated between two groups.After 12-month follow-up,CD4 + T cells,the rate of rehospitalization,CMV retinitis,and mortality were evaluated.Results There were no difference in CMV-DNA,HIV-RNA,CD4 + T cells,the rate of PCP,and tuberculosis(P > 0.05).At the end of 12 months,there were higher CD4 +T cells and lower rehospitalization rate in treatment group than control group(t =-3.850,P < 0.05;x2 =6.078,P < 0.05).CMV retinitis was not found.The mortality was not different between two groups(P >0.05).Conclusions Anticytomegalovirus therapy was beneficial to the increment of CD4 +T cells and decrement of rehospitalization,and prevention from CMV retinitis.
7.Study on cytokines and dendritic cells involving immunologic inhibition of rat high-risk corneal transplantation induced by SEB

objective To explore the role of cytokines and dendritic cells (DCs) in rat high-risk corneal allograft survival pro- longed by superantigen Staphylococcal Enterotoxin B (SEB) and to compare the different effects between SEB and glucocorticoid. Design Experimental study.Participants Fisher 344 and Lewis rats.Methods The Fisher 344 and Lewis inbred rats were used for high-risk penetrating keratoplasty model.All of the Lewis rat recipients were divided into three groups by blinded fashion.GroupⅠand GroupⅡrats were injected intraperitoneally with 0.2ml saline buffer or SEB (75?g/ml) respectively at 4-day intervals on three occasions before transplantation.GroupⅢrats were injected subconjunctivally with 0.1ml dexamethasone (1mg/ml) daily from the first day after surgery for 2 weeks.The allograft survival was examined under slit-lamp.The concentration of interleukin IL-1?,IL-2,IL-4,IL-5,IL-6, IL-10,TNF-?in rat aqueous humor and peripheral blood were measured by liquichip and the cytokine and CD11c,CD80,MHC-Ⅱex- pression in corneal grafts were examined by immunohistochemestry staining.Main Octcome Measures Mean survival time of the allo- grafts,the level of cytokines in aqueous humor,peripheral blood and corneal grafts.Results Compared with group control,the grafts mean survival time was delayed about 3.8d in group SEB (P=0.00) and 7.1d in group dexamethasone(P=0.01).But there was no signifi- cant difference between group SEB and dexamethone (P=0.26).Liquichip test showed that the level of IL-1?in aqueous humor was re- duced and IL-4,IL-6 and IFN-?,ascended.Only IFN-?,and IL-6 could be found in peripheral blood,and the changing shift of them was similar to that in aqueous humor.The immunohistochemistry staining showed that the expression of IL-2 in rat corneal grafts was signif- icantly decreased but IL-4,IL-6 and IL-10 elevated.The expression of DCs in group SEB was similar to that in group control,which was elevated after keratoplasty,but the phenotype of DCs was not the same.There were predominantly mature DCs in group control while immature DCs in group SEB.Conclusions The immunological inhibiting effect of SEB is same to glucocorticoid,but the mecha- nism is different.SEB can modulate immune response,which might induce immune inhibition via the local production of cytokines and effect on DCs maturation involving corneal graft rejection prevention.
8.Evaluation of Bioavailability and Bioequivalence of Rabeprazole Sodium Enteric-coated Pellets Capsules in Humans

Objective: To evaluate the bioavallability and bioequivalence of rabeprazole sodium enteric-coated pellets capsules. Methods:A randomized crossover design was performed in 32 healthy male volunteers. A single oral dose of 20 mg rabeprazole sodium enteric-coated pellets capsules ( test preparation) or enteric-coated shell capsules ( the reference capsules) was administrated under fed conditions. The wash period was 7 days. The blood samples were collected at different time points. The concentration of rabeprazole in plasma was determined by an LC-MS/MS method. The pharmacokinetic parameters were calculated by DAS 3. 0 software and the bio-equivalence was evaluated. Results:The maln pharmacokinetic parameters of the two formulations were shown as follows:T1/2 of (2. 20 ± 0. 83)h and(1. 951 ± 0. 515)h,Tmax of (3. 88 ± 1. 11)h and(4. 64 ± 1. 504)h,Cmax of (401. 06 ± 170. 75)ng·ml-1 and(394. 63 ± 215.64)ng·ml-1,AUC0→t of (918.42 ±427.39)ng·h·ml-1 and (994.49 ±520.73)ng·h·ml-1, and AUC0→∞ of(937.30 ± 445.13)ng·h·ml-1 and(1 011.69 ±534.77)ng·h·ml-1. The analysis showed that the maln pharmacokinetic parameters of the two formulations had no significant differences(P>0. 05) except for Tmax(P<0. 05). The relative bioavallability of rabeprazole sodium enteric-coated pellets capsules was (99. 80 ± 7. 20) %. Conclusion:Compared with the reference capsules, rabeprazole sodium enter-ic-coated pellets capsules show the property of higher dispersion degree, milder influence from food, more rapid release and absorption. The enteric-coated pellets capsules and the reference capsules are bioequivalence.
