1.Indocyanine green angiography in ocular contusion

Purpose To observe the changes of choroidal circulation and the retinal lesions caused by ocular contusion with indocyanine green angiography (ICGA). Methods ICGA examination was performed on 30 cases (30 eyes) of various traumatic condition in conjunction with fundus fluorescein angiography (FFA). Results FFA of 19 cases (63.3%) showed the hypofluorence in quadrant or whole disc in accordance with the area of delayed filling of choroid. Twenty six cases (86.2%) showed defected choroidel perfusion in ICGA,among them 16 cases showed localized delayed perfusion, in which the shortest perfusion time was 1 min 50 s and the longest time was 5 min.43 s,and 10 patients showed localized perfusion defect,and reversed filling time of retinochoroid vessels in 6 patients. Five cases (16.6%) had delayed filling time in both choroidal and central retinal vessels. Damage of retinal pigment epithelium was found in the areas of choroidal abnormal perfusion. Conclusion ICGA combined with simultaneously FFA, is valuable in evaluating blunt injury of the ocular fundus and beneficial to its diagnosis and treatment.

Objective To optimize methods of culturing smooth muscle progenitor cells(SPCs) from mononuclear cells(MNCs) of peripheral blood. Methods Human MNCs isolated from buffy coat were seeded on M-199 with bovine pituitary extraction.On the eighth day outgrowth cells were stimulated with platelet-derived growth factor-BB(PDGF-BB).Fifteen days later,immunofluorescence,Western blot or RT-PCR was used to analyzed the expression of smooth muscle cell specific ?-actin(?-SMA),smooth muscle myosin heavy chain(SM MHC),Calponin,CD34,Tie-2 and Flk-1,and fluorescence activated cell sorter was employed to examine ?-SMA positive cells ratio. The coronavirus disease 2019 (COVID-19) is an acute infectious disease caused by the new severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection, which has led to serious worldwide economic burden. Due to the continuous emergence of variants, vaccines and monoclonal antibodies are only partial effective against infections caused by distinct strains of SARS-CoV-2. Therefore, it is still of great importance to call for the development of broad-spectrum and effective small molecule drugs to combat both current and future outbreaks triggered by SARS-CoV-2. Cathepsin L (CatL) cleaves the spike glycoprotein (S) of SARS-CoV-2, playing an indispensable role in enhancing virus entry into host cells. Therefore CatL is one of the ideal targets for the development of pan-coronavirus inhibitor-based drugs. In this study, a CatL enzyme inhibitor screening model was established based on fluorescein labeled substrate. Two CatL inhibitors IMB 6290 and IMB 8014 with low cytotoxicity were obtained through high-throughput screening, the half inhibition concentrations (IC50) of which were 11.53 ± 0.68 and 1.56 ± 1.10 μmol·L-1, respectively. SDS-PAGE and cell-cell fusion experiments confirmed that the compounds inhibited the hydrolysis of S protein by CatL in a concentration-dependent manner. Surface plasmon resonance (SPR) detection showed that both compounds exhibited moderate binding affinity with CatL. Molecular docking revealed the binding mode between the compound and the CatL active pocket. The pseudovirus experiment further confirmed the inhibitory effects of IMB 8014 on the S protein mediated entry process.
5.Advances of chemical constituents and pharmacological activities of Myristica genus.

The genus Myristica (Myristicaceae) consists of 120 species, which were distributed in South Asia, from west Polynesia, Oceania, eastern India to the Philippines. Phytochemical studies showed that 164 compounds including a majority of lignans, along with phenglpropanoids, flavonoids and phenolics, have been isolated from this genus, which exhibited anti-microbial, anti-inflammatory, anticancer, hyperglycemic and hepatic protective activities. This article summarizes research progress of the chemical compositions and their pharmacological activities from this genus, which could provide reference for the in-depth development and utilization of the Myristica plants.

6.Clinical study on mild moxibustion for knee osteoarthritis

Objective:To observe the clinical effect of mild moxibustion for knee osteoarthritis (KOA) and to explore the role of mild moxibustion in relieving pain,reducing stiffness and improving joint dysfunction in patients with KOA.Methods:Eighty patients with KOA were randomly allocated into either a moxibustion group or a medication group by the random number table,with 40 cases in each group.The moxibustion group used mild moxibustion at Neixiyan (EX-LE 5),Dubi (ST 35),Xuehai (SP 10) and Liangqiu (ST 34),30 min each time,3 times a week;the medication group was given celecoxib capsule (celebrex),0.2 g each time,once a day.Both groups were treated for 4 weeks.The visual analog scale (VAS) and Western Ontario and McMaster Universities osteoarthritis index (WOMAC)scores were evaluated before and after treatment.The efficacy of the two groups was compared after treatment.Results:After treatment,the overall efficacy of the moxibustion group was significantly different from that of the medication group (P<0.05).The VAS and WOMAC scores of the two groups were lower than those before treatment (both P<0.01).The changes in the VAS and WOMAC scores after treatment in the moxibustion group were significantly different from those in the medication group (both P<0.05).After treatment,in single item of WOMAC,the changes in pain and joint dysfunction in the moxibustion group were more statistically significant than those in the medication group (both P<0.05).Conclusion:Mild moxibustion and oral celebrex can reduce the VAS and WOMAC scores of patients with KOA.Mild moxibustion is superior to oral celebrex in relieving pain and improving joint function.
7.Respiratory Function Tests of 102 Cases of Healthy Children with Impulse Oscillometry

Objective To study the respiratory function tests reference values of healthy children in China with impulse oscillometry(IOS).Methods The respiratory function tests reference values of 102 children were measured by using masterscreen IOS and analysed it.Results Impedance and airway resistance reference values decreased as age and body height increased step by step,at the same time capacitance decreased.The difference was significant while the predicted values compared to the measured data.Conclusion We shall use respiratory function tests reference values which are established by us,and the difference is significant compared with the foreign predicted(values.)

Objective To study the relationship between serum lipid level with embryo quality in vitro fertilization cycle.Methods A total of 603 pairs of infertile couples who were treated with in vitro fertilization and embryo transfer technique (IVF-ET) in our hospital from October 2015 to June 2016 were collected.All subjects underwent long-term ovarian hyperstimulation,blood lipid and the embryo quality were measured and evaluated.Results Triglyceride (TG),total cholesterol (TC) and body mass index(BMI) were significantly negatively correlated with fertilization rate,2PN fertilization rate,cleavage rate and embryo rate (P<0.05).Density lipoprotein (LDL) and non-high density lipoprotein (NHDL) and lipoprotein (a) [Lp(a)] were negatively correlated with fertilization rate,2PN fertilization rate,high density lipoprotein.The cholesterol (HDL) were positively correlated with fertilization rate,2PN fertilization rate,cleavage rate and embryo rate(P>0.05).Conclusion There is a linear correlation between blood lipid level and AMI and embryo quality.Serum lipids and AMI can be used to evaluate the quality of embryo and provide suggestion for the quality control of IVF-ET.
10.Effect of trimetazidine combined with atorvastatin on cardiac function andlevel of hs-CRP, IL-6, and Fib in patients with coronary heart disease

Objective To investigate the effect of trimetazidine combined with atorvastatin on cardiac function and the level of hs-CRP,IL-6,and Fib in patients with coronary heart disease.Methods Patients (120 cases) with coronary heart disease in the first people's Hospital of Xianyang City from January 2014 to January 2016 were selected and randomly divided into two groups.The observation group was treated with trimetazidine combined with atorvastatin,the control group was treated with trimetazidine,the curative effect,left ventricular ejection fraction,left ventricular diastolic end systolic diameter,left ventricular posterior wall thickness and serum hs-CRP,IL-6,and Fib levels were compared.Results The effective rate of observation group was 91.67% (55/60),significantly higher than the control group 76.67% (46/60) (P < 0.05);The left ventricular ejection fraction,left ventricular end-diastolic diameter,and left ventricular posterior wall thickness of two groups were significantly improved (P < 0.05),and the improvement of the observation group was significantly better than that of control group (P < 0.05);The serum levels of hs-CRP,IL-6,and Fib were significantly lower (P < 0.05),and the observation group was more obvious (P < 0.05);The incidence of adverse reactions of two groups had no significant difference.Conclusion Trimetazidine combined with atorvastatin have high curative effect on coronary heart disease,and can improve cardiac function,significantly reduce the levels of hs-CRP,IL-6,and Fib,worth clinical promotion.
