1.Change of serum iron in children with Tourette syndrome

BACKGROUND: The etiology and pathogenesis of Tourette syndrome(TS)is still unacknowledged, and related studies on the relationship between TS and hypoironemia are unavailable. Since hypoironemia has been found existed in children with TS who showed better outcomes after receiving adjuvant ferralium, thereby this study was designed to investigate the relationship between TS and serum iron.OBJECTIVE: To study the relationship between TS and serum iron.SETTING: At a nursing science department of a university-affiliated hospital, pediatric department of a university affiliated hospital and sectional hospital.PARTICIPANTS: Between June 1997 and December 2000, 45 children with TS received treatment at pediatric clinic of Chenghai sectional people' s hospital, Shantou city, who accorded with the diagnostic standards of the second edition of Chinese Psychopathy Category and Diagnostic Standards,those who were confirmed as brain organic diseases by CT and MIR were excluded.METHODS: Serum iron, hemoglobin and the number of red blood cells were determined in 45 TS children and compared with that in 38 controls.MAIN OUTCOME MEASURES: Difference of the level of serum iron and the incidence of hypoironemia between two groups.RESULTS: The level of serum iron was(12.79 ±0.67) μmol/L in experimental group, obviously lower than(19.26 ± 5.38) μmol/L in control.group( P < 0. 005), the incidence of hypoironemia was 42% significantly higher than13% of control group( P < 0. 005).CONCLUSION: Quite a lot of children with TS have hypoironemia that might be one of the pathogenesis factors, the possible mechanism is, as the result of hypoironemia, decreased activity of monoamine oxidase led to abnormal monoamine neurotransmitter.
2.Relationship between therapeutic compliance and complications in children with severeβ-thalassemia

Objective To explore the relationship of therapeutic compliance and complications in children with severeβ-thalassemia.Method One hundred and eight children with severeβ-thalassemia involved in the survey using convenient sampling to investigate the relationships between compliances with blood transfusion and iron removal treatment and complications.Results Of all the patients,66 complied with blood transfusion and the rest 42 did not;73 complied with the therapy and the rest 35 did not. Those complying with the blood transfusion had significant lower rates of developmental retardation,unusual faces,hepatosplenomegaly(P<0.01). Those complying with the therapy had significant lower rates of injuries in liver,heart and endocrinal system(P<0.01).Conclusion Therapeutic compliance in children with severeβ-thalassemia is at lower level so that the incidence of complications in them is higher.
3.The Relationship Between Influenza Peak and Weather in Hong Kong

Objective To discuss the relation between influenza peak, temperature and relative humidity. Methods Statistical analysis on the data of influenza consultation rates, temperature and relative humidity from 1998 to 2002 in Hong Kong area was carried out to propose some relation between influenza peak, temperature and relative humidity. Results It was slightly possible to cause influenza peak when the week-averaged temperature was less than 20 ℃ and largely possible to cause influenza peak when the week-averaged temperature was less than 15 ℃. It was also largely possible to cause influenza peak when the week-averaged temperature was very low and the relative humidity was higher which resulted the lower body-sense temperature compared with the forecasting temperature. Conclusion The prevalence of influenza occurs during four seasons in one year in Hong Kong, but the prevalence peak of influenza mainly appears under the condition of lower temperature(
5.Proliferation and Differentiation of Adult Neural Stem Cell Induced by Traditional Chinese Medicine in Brain

Neural stem cells are a category of cells with self-renewal and differentiation potential. Proliferation and differentiation of neural stem cells have closely relation with growth factor, cell factor, environmental signal, and so on. In recent years, evidence indicates that neural stem cells possessing multipotency naturally occur in adult brain. The found of adult neural stem cells have changed the theory that central nervous system cannot regenerative. Chinese Traditional Medicine can effectively induce adult neural stem cells to proliferate and differentiate, opening up unprecedented direction in the research for the treatment of nerve regeneration, and showing great potency for curing cerebral ischemic injury.
6.Morroniside Inhibits H_2O_2-Induced Oxidative Injury in SH-SY5Y Neuroblastoma Cells

Objective To investigate the effects of Morroniside on H2O2-induced oxidative injury in SH-SY5Y neuroblastoma cells.MethodsSH-SY5Y cells were pre-incubated with Morroniside(1,10,and 100 μmol/L)for 24 h prior to exposure to H2O2(500 μmol/L)for 18 h.The content of reactive oxygen species(ROS),nitric oxide(NO),glutathione(GSH)were determined.ResultsPretreatment the cells with Morroniside(10 and 100 μmol/L)lowered H2O2-induced ROS release by 37%(P<0.01)and 27%(P<0.05)versus H2O2,and NO release in a dose-dependent manner by 31%(P<0.05),53%(P<0.01)versus H2O2.Morroniside(1,10,and 100 μmol/L)increased GSH level in cells treated with H2O2 in comparison with cells exposed only to H2O2,respectively.ConclusionMorroniside shows neuroprotection effect against H2O2-induced oxidation injury in SH-SY5Y cell.
7.Effect of nourishing Yin, strengthening Qi and activating blood decoction on Fas/FasL in salivary glands of NOD mice with Sjogren's syndrome and their mRNA expression.

OBJECTIVETo observe the effect of nourishing Yin, strengthening Qi and activating blood decoction on Fas/FasL in salivary glands of NOD mice with Sjogren's syndrome and their mRNA expression.
METHODThirty-two NOD mice were randomly divided into the model group, the traditional Chinese medicine group (TCM group, orally given 0.4 mL nourishing Yin, strengthening Qi and activating blood decoction as per 100 g x kg(-1) everyday), the hydroxychloroquine group (given 0.4 mL hydroxychloroquine as per 60 mg x kg(-1) everyday), the traditional Chinese medicine and western medicine group (TCM WM group, given nourishing Yin, Strengthening Qi and activating blood decoction 50 g x kg(-1) and hydroxychloroquine 60 mg x kg(-1), 0.4 mL everyday), with eight mice in each group. Eight Balb/C mice were selected as the normal control group (normal group). All of mice were killed after eight weeks, and their submaxillary glands were dissected. The expression levels of Fas/FasL were examined by immunohistochemical method, and the FasL mRNA was detected by RT-PCR.
RESULTThe expression levels of Fas/FasL in salivary glands of the model group were higher than that of other groups (P < 0.05). The expression level of FasL of the normal group was much lower than that in the hydroxychloroquine group (P < 0.05). The relative expression level of Fas mRNA in salivary glands of the model group was higher than that in other groups, but the control group was notably lower than other groups (P < 0.05). The expression level of FasL mRNA in salivary glands of the model group was higher than that in TCM and TCM WM groups (P < 0.05). But the expression level in TCM WM group was notably lower than the hydroxychloroquine group (P < 0.05).
CONCLUSIONThe nourishing Yin, strengthening Qi and activating blood decoction could down-regulate the expression level of Fas/FasL in salivary glands of NOD mice with Sjogren's syndrome and their mRNA expression, and had a better efficacy after being combined with hydroxychloroquine. The nourishing Yin, strengthening Qi and activating blood decoction might treat the Sjogren's Syndrome by reducing apoptosis which is regulated by Fas/FasL

10.Effect of K202A Mutation in the Thermostability of Penicillum expansum Lipase

Lipase gene from Penicillium expansum(lip07) was cloned and over-expressed in Pichia pastoris.a random mutant named ep8,which contained a single amino acid substitution,was obtained by using the lip07 as an error-prone PCR template in previous study.ep8 shows higher thermostability than that of lip07,To further improve the thermostability of the lipase,the Lys of wild-type(lip07) and mutant(ep8) in 202 were substituted by Ala using the Overlap extension PCR technique respectively.The mutant genes(lip07-K202A and ep8-K202A) were subcloned into pAO815,and then transformed into the Pichia pastoris GS115 for extracelluar expression,respectively.15% SDS-PAGE analysis indicated that the molecular mass of PEL-ep8-K202A and PEL-lip07-K202A are both about 28kDa,which is same with the wild-type lipase.The Tm of PEL-ep8-K202A is 41.66℃,2.63℃ higher than that of the wild-type(39.03℃) and 1.21℃ higher than the random mutant(PEL-ep8:40.45℃);the Tm of single mutant(PEL-lip07-K202A) is 37.08℃,2℃ lower than that of the wild-type lipase.
