Objective To study the morphology changes and the expression of phosphatidyl serine(PS) in apheresis platelets(Plts) during storage,also to analyze their relationship,in order to provide laboratorial evidence for the in vitro storage period of plts.Methods The morphological changes of plts during 8-day storage were detected by May-Grunwald-Giemsa dyeing method,and the expression of PS on platelet membrane was tested by flow cytometry(FCM).Results The morphological lesions appeared on the fourth day of storage,because the modified morphological score(MMS) of 4-day group was significantly lower than that of fresh plts(t=2.341,P

Colorectal cancer is currently the third most common cancer worldwide,and there are still half of the patients undergoing recurrence and metastasis after surgical treatment,so it is necessary for colorectal cancer patients to receive radiation therapy routinely.Due to the side effects brought by radiotherapy,it is of great importance to solve how to minimize the radiation dose in radiation therapy and improve radiation sensitivity.In recent years,people discovered that microRNAs can not only be involved in the origins of colorectal cancer and progress,but also play a increasingly important role in cancer radiosensitivity.MicroRNAs can regulate tumor radiosensitivity by influencing tumor microenvironment and function on target genes.DNA damage response caused by radiation includes the activation of ATM,histone modification and chromatin remodeling,cell cycle arrest,damage repair and apoptosis.microRNAs can regulate tumor radiosensitivity through above processes.This review focuses on the mechanism of microRNAs in affecting DNA damage repair and prospects the future of microRNAs in influencing the sensitivity of cancer radiotherapy in clinical application.

OBJECTIVETo investigate the short-term clinical outcome of unicompartmental knee arthroplasty for the treatment of spontaneous osteonecrosis of the knee.
METHODSFrom September 2013 to April 2014,5 patients with spontaneous osteonecrosis of the knee underwent unicompartmental knee arthroplasty, included 3 males and 2 females, aged from 65 to 80 years old with an average of 74 years. The courses of disease was from 1 to 6 years with the mean of 3 years. According to the radiographic staging criteria of Koshino, 1 case was stage II, 2 cases were stage III, 2 cases were stage IV. Clinical effects were assessed by VAS score, HSS score, and knee range of motion, tibiofemoral angle before and after operation.
RESULTSAll the patients were followed up from 6 to 7 months with an average of 6.4 months. All incisions obtained primary healing, and there were no complications such as infection, thrombosis, fracture of lower limbs. All 5 patients' pain relieved and their knee function improved significantly after operation, but knee range of motion had no obviously improved. Postoperative HSS scores, VAS scores, tibiofemoral angle were significantly improved than that of preoperative.
CONCLUSIONThe short-term effect of unicompartmental knee arthroplasty in treating spontaneous osteonecrosis of the knee is satisfactory.

6.Neuroprotective Effect of Recombinant Human Erythyropoietin Theraphy for Neonatal Hypoxic-Ischemic Encephalopathy

Objective To evaluate the neuroprotective effect and safety of neonatal hypoxic-ischemic encephalopathy(HIE)treated with recombinant human erythropoietin(rhEPO).Methods Fifty-three neonates with HIE were randomly divided into rhEPO treated group(n=29) with the dosage of 300 U/(kg?time),three times a week for 2 weeks and control group(n=24)without rhEPO.All supportive measures were same between 2 groups.Neurological scoring was evaluated at d3,d5 and d7 Neonatal behavioral neurological assessment(NBNA) was evaluated at d7,d14 and d28.The neurodevelopment quote was evaluated at age of 3 and 6 months.Blood pressure,liver and renal function,blood electrolytes and blood hemoglobin,platelet and reticular red blood cell count were monitored before and after treatment in all infants.Results The neurological scoring between two groups had no difference at d3.The significant difference was found at d7(P0.05).Conclusions Teraphy with rhEPO on neonatal HIE infants can promote neurological recovery,and there is no serious side effect with rhEPO treatment.
7.Study on genetic characteristics of VP1 region of Enterovirus 71 strains isolated from Shanghai, China in 2010

Objective To study the gene characteristics of VP1 region of Enterovirus 71 (EV71) strains isolated from clinical specimens of children with hand-foot-and-mouth disease(HFMD) in Shanghai in 2010.MethodsEighteen EV71 isolates were selected from different periods of year 2010,including strains isolated from fatal cases and non-fatal cases.Complete VP1 gene (891nucleotides) of the eighteen EV71 isolates were amplified and sequenced,and then compared with that of genotype A,B,C reference EV71 strains in GeneBank by homogeneity and phylogenetic tree analyses.ResultsThe nucleotide homogeneities between these 18 Shanghai strains and the representative isolates of genotype A and B were 81.5 % -82.6 % and 83.4 %- 84.2 %,respectively,while the amino acid homogeneities were 94.3 %- 95.0% and 96.6% -97.0%,respectively.The nucleotide and amino acid homogeneities between the 18 Shanghai strains and the representative isolates of genotype C were 87.4%- 99.2% and 98.7% -100.0%,respectively.Of note,the nucleotide and amino acid homogeneities between Shanghai strains and Fuyang EV71strains (representative strain of C4 subtype) appeared to be 97.8%- 99.2% and 99.3%- 100.0%,respectively.The eighteen EV71 Shanghai strains were classified as genotype C,subgenogroup C4 in the phylogenetic tree.There was no remarkable difference in VP1 gene between the strains isolated from fatal cases and non-fatal cases.ConclusionThe EV71 strains isolated from Shanghai belong to subgenogroup C4.
8.Survivability of hand foot mouth disease virus in tap water

Objective To evaluate the survivability of hand foot mouth disease(HFMD)virus,in tap water for daily use.Methods HFMD viruses were isolated from cases of HFMD in Shanghai and Zhejiang from in 2008.Six isolated strains (five subtype of enterovirus 71 and one coxackie virus)were selected in this study.These viruses were mixed with chloride 1.0 mg/L tap-water and then inoculated into Vero cells.The cytopathic effect (CPE)was checked everyday in order to survey the survivability of each virus strain.The decline of virus survivability was analyzed by scatter diagram.Results These six strains of HMFD virus could survive longer than one month in tap water with initial chloride concentration of 1.0 mg/L and still had celluar infectivity.The survivabilities were varied between viruses isolated from different HFMD cases.Conclusions The survivabilities of enterovirus 71 and coxackie virus stains are quite strong in water.Therefore,the transmission route of water-borne pathogens should be monitored in regions using tap water during HFMD epidemic period.
9.Variations of influenza A (H1N1) viruses in Shanghai area in the winter of 2010

Objective To understand the genetic and antigenic variations of influenza A (H1N1) isolates in Shanghai area in winter of 2010.Methods A total of 137 throat swabs were collected from patients with influenza-like illness in the sentinel hospital in Shanghai area from December 2010 to January 2011,then inoculated into Madin-Darby canine kidney (MDCK) cells.The types of influenza were identified by direct immunofluorescence assay (DIF) and influenza A (H1N1) subtype was determined by reverse transcriptase-polymerase chain reaction (RT-PCR).The mutations of gene and amino acid locus were analyzed through the whole genome sequencing of hemagglutinin (HA),neuraminidase (NA) and polymerase (PB2) segments from some influenza A (H1N1) isolates.Results Total of 53 human influenza virus strains were isolated including 48 influenza A (H1N1) virus strains.Nineteen strains were selected for sequencing by simple random sampling.The phylogenetic tree of HA gene revealed that the latest isolates and most of influenza A (H1N1) viruses isolated before June 2010 were not in the same stem.Analysis of amino acid residues in HA protein showed that mutations were found in antigenic determinant region in some strains.Residues at the enzyme active sites of NA protein were strictly conservative,no change was observed in amino acid residues which were related to drug resistance against oseltamivir and zanamivir.The 627 and 701 residues in PB2 protein were glutamic acid and aspartic acid,respectively,which was still the feature of avian influenza virus,but E677G mutation was detected.Conclusion Compared to influenza A (H1N1) strains isolated in spring and summer,some variations have been detected in the strains isolated in Shanghai area in winter of 2010,some antigen drift and adaptive evolution in mammalian hosts have appeared.
10.Effects of Chinese herbal medicine Huaiqihuang Granule on nephrin and podocin expressions in renal tissues of rats with adriamycin-induced nephrosis.

Object: To investigate the effects of Huaiqihuang Granule, a compound Chinese herbal medicine, on expressions of nephrin and podocin of slit diaphragm of glomerular podocytes in rats with adriamycin-induced nephrosis and to explore the mechanism in reducing the proteinuria. Methods: Twenty SD rats were randomly divided into five groups: control group, model group, glucocorticoid group, Huaiqihuang Granule group and Huaiqihuang Granule plus glucocorticoid group. The 24-hour urine was collected 7, 14, 21 and 28 days after adriamycin injection respectively to measure 24-hour urinary protein, and all rats were sacrificed after 28-day treatment. Pathological changes in renal tissues were observed under a light microscope and an electron microscope. Expressions of nephrin and podocin mRNAs in renal cortex were determined by real-time polymerase chain reaction, and protein levels of nephrin and podocin were detected by Western blotting. Results: (1) In the model group and the treatment groups, the level of urinary protein increased significantly from the 14th day. (2) Under the light microscope, inflammatory cells and slight fibroplasia were found in renal interstitium of the model group, but there were less inflammatory cells in renal interstitium in the intervention groups than in the model group. Under the electron microscope, 29 days after adriamycin injection, extensive fusion of foot processes was observed. (3) The expressions of nephrin and podocin were higher in treatment groups than in the model group. (4) Proteinuria level was negatively correlated with the expressions of nephrin mRNA and nephrin and podocin proteins. Conclusion: The above results indicate that Huaiqihuang Granule can maintain the integrity of the slid diaphragram in podocyte, alleviate the lesion of glomerular filtration membrane, and decrease the proteinuria by up-regulating the expressions of nephrin and podocin. Huaiqihuang Granule plus glucocorticoid maybe has better effects than glucocorticoid alone.
