2.Biosynthesis-based production improvement and structure modification of erythromycin A.

Erythromycin A is a clinically important macrolide antibiotic with broad-spectrum activity. Its biosynthesis involves the formation of the 14-membered skeleton catalyzed by polyketide synthases, and the modification steps such as hydroxylation, glycosylation and methylation. Based on the understanding of the biosynthetic mechanism, it is reliable to genetically manipulate the erythromycin A-producing strain for production improvement and structure modification. In this paper, we reviewed the progress regarding erythromycin A in high-producing strain construction and chemical structure derivation, to provide insights for further development.

3.THE TREATMENT OF RETINAL REDETACHMENT AFTER VITRECTOMY AND SILICONE OIL TAMPONADE

OBJECTIVE: To investigate the treatment of retinal redetachment after vitrectomy and silicone oil tamponade, METHODS: Investigating retrospectively on the treatment effect of 8 cases of redetachment of retina with proliferative vitreoretinopathy(PVR) in which the retinas had been attached formerly after vetrectomy and silicone oil tamponade opertion. The reoperative procedures included pars plana vitrectonly, membranes peeling, retinotomy, inner exchange of the fluid and silicone oil tamponade and subretinal membranes were removed out in 2 eyes. RESULTS: Six eyes had anatomical reattachment postoperatively and another 2 eyes still had inferior shallow retinal detachment. Visual acuity was improved in 6 patients and remained unchange in 2 patients. CON CLUSIONS: The cause of recurrent retinal detachment might be the formation of the proliferative epiretinal and subretinal membranes, and vitrectomy to release the traction of proliferative membranes and full fluid silicone oil exchange should do good to reattachment.
4.Application of colonoscopy in pediatric surgery

Objective To investigate the application of colonoscopy in pe diatric surgery. Methods The colonoscopy was performed in 776 patients of ped iatric surgery with colonoscope type Olympus PCF 20. Results 659 cases of large intestinal polyposis,including 4 cases of familial polypsis coli,2 cases of Peu tz- Jegher's syndrome,had been diagnosed and polypoectomy was performed.42 cas es of congenital megacolon had been diagnosed and classified.75 cases of intussu sception had been diagnosed and treated. Conclusion Colonoscopy is important and useful in diagnosis and treatment for colon disease in pediatric surgery.
5.Diagnostic value of neurovascular compression in trigeminal neuralgia with 3D-TOF MRA and postprocessing techniques

Objective To evaluate diagnostic value of 3D time-of-flight (3D-TOF) magnetic resonance imaging (MRA) and postprocessing techniques in neurovascular compression (NVC) in trigeminal neuralgia.Methods 38 patients with trigeminal neuralgia were examined by 3D-TOF MRA.MRA fundament images and multiplanar reconstruction (MPR) images were blindly interpreted by 2 neuroradiologists.Results (1) Of all the patients, 31 cases showed vascular compression or contact on MR imaging, 26 cases showed the vascular compression or contact at the same side of clinic symptoms. 6.Blood analyzer Sysmex XN-9000 testing in low whole blood platelet performance evaluation

Objective:To evaluate the performance of Sysmex XN-9000 automatic blood analyzer (hereinafter referred to as XN-9000) in whole blood with the low platelet (PLT). Methods:A total of 48 whole blood samples with platelet count less than 70×109/L were randomly collected from XN-9000 correlation analysis to evaluate XN-9000 the accuracy and precision of detection in low whole blood PLT and carry pollution rate, use XN-9000 and microscope count, respectively.Results: XN-9000 detection in low whole blood PLT between batch and batch of CV% 1.13% and 1.29%, respectively; XN-9000 with microscope PLT count results have high correlation, The regression equation forY=0.917X+3.217,correlation coefficientr=0.964; XN-9000 detection in low whole blood PLT carry pollution at a rate of 0.12%. Detection accuracy of PLT Bias, %(-1.25%~1.8%); Linear range(0~1206)×109/L; Clinical reportable range (0~6030)×109/L.Conclusion: The detection of XN-9000 blood analyzer is in low whole blood PLT has high precision and accuracy, carry pollution rate is extremely low, the result has the reliability, can be directly applied to clinical.
7. Mitochondrial nitric oxide synthase: Biological function

Increasing evidences have shown the existance of a mitochondrial nitric oxide synthase (mtNOS), which binds to the matrix face of the mitochondrial inner membrane and produces nitric oxide (NO) through a Ca2+ sensitive pathway. Under physiological condition, the NO catalyzed by mtNOS regulates mitochondrial oxygen consumption and transmembrane potential via reversible competition with cytochrome C oxidase. The reaction of NO with superoxide anion, which was produced by mitochondrial respiratory chain, yields peroxynitrite. Peroxynitrite irreversibly modifies susceptible targets in mitochondria and induces oxidative and/or nitrative stress. In addition, NO has also been implicated in the programmed cell death. This article reviews the current understanding of mtNOS's role in the regulation of mitochondrial functions.
8.The Mechanism Behind the Anti-Apoptotic Effects of TNK on Islet β Cell in Type 2 Diabetic ZDF Rats

This study aimed to investigate the anti-apoptotic effects of Tangnaikang (TNK) on islet β cells in Zucker diabetic fatty (ZDF) rats.Six male fa/+ ZDF rats were took as the control group,while other thirty male fa/fa ZDF rats were divided into five groups at random:the model group,the metformin group,the high-,mediumand low-dose TNK groups,depending on their body weight and random blood glucose.Prior to the administration,fasting blood glucose and fasting insulin were measured by drawing blood with inner canthusl.Materials were prepared when administered for six weeks.Fasting blood glucose and fasting insulin were detected again.When the sections of the rat pancreatic tissue were embedded,the morphological changes of the islet were observed via HE staining,and the apoptosis of islet β cell were observed using TUNEL.Positive expression of Caspase-3,the transduction enzyme of cell death signal,was tested by immunohistochemical method.It was found that the fasting blood glucose of the (fa/fa) ZDF rats in the high-,medium-and low-dose TNK group was significantly improved after administration (P < 0.01).The serum insulin of rats in the high-,medium-and low-dose TNK group arised compared with the model group,while the high-and low-dose TNK group showed differences in a statistical sense.Compared with the model group,the HOMA-IR of all the treatment group decreased,while significant difference was presented between the high-dose TNK group and the metformin group.HE staining showed that the morphology of the islet β cell of the rats in all the treatment group was improved.The results of TUNEL showed significantly apoptotic changes on islet β cell of the fa/fa ZDF rats.Compared with the model group,the positive expression of TUNEL in the metformin group and the high-dose TNK group were significantly reduced (P < 0.05).The result of immunohistochemistry method showed that the protein levels of Caspase-3 in the metformin group and the high-dose TNK group decreased (P < 0.05).In conclusion,it was demonstrated that TNK effectively reduced the apoptosis of islet β cells in fa/fa ZDF rats,which presented a protective effect.
9.Comparing the characteristics of tissue culturein vitro and different cell types of cancer microenvironment in tissues at different distances from colorectal cancer lesions

Background and purpose:Cancer microenvironment has become a hot topic of cancer research. It is important in the initiation of colorectal cancer. This study aimed to discuss the correlation between the characteristics of tissue culturein vitro and different cell types in cancer microenvironment.Methods:Samples were collected at different distances from the colorectal cancer lesions, which were named as positions 1, 2 and 3 from distal to proximal. Tissues were cut into 1-2 mm3 forin vitro culturing. HE staining was used to observe the structure of crypts. Immunohistochemistry was used to detect the expressions of Cyclin D1 (CD1), CD133, cytokeratin18 (CK18), vimentin and α-smooth muscle actin (α-SMA).Results:Position 3 grew faster than position 2 and position 1. As getting closer to the colorectal cancer lesions, expressions of CD1, CD133, vimentin and α-SMA were increased while expression of CK18 was decreased.Conclusion:The tissue structure and the expression of different cell types in cancer microenvironment changed more seriously as get-ting closer to the colorectal cancer lesions. This indicated that the change of cancer microenvironment may contribute to the initiation of colorectal cancer.
