1.Relationship between tumor tolerance to TRAIL and its receptors

Tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) preferentially induces apoptosis in tumor cells but not in normal cells. It induces apoptosis in its target cells by interacting with surface death receptors DR4 and DR5,which contain a cytoplasmic region designated as thedeath domain. Clinical use of rhTRAIL and its agonistic antibodies is limited due to partial or completetolerance to TRAIL in human tumor cell lines. As TRAIL binding to its death receptors is the initial and crucial step in initiating apoptosis, the mechanisms of TRAIL-tolerance in tumor cells is closely related to the expression,location,distribution and function of TRAIL receptors . Personalized care combined with other therapies targeting TRAIL receptors may be expected to overcome the TRAIL-tolerance of tumor cells eventually.
3.Clinical analysis of patients with eosinophilia-myalgia syndrome

Objective To analyze clinical features of patients with eosinophilia-myalgia syndrome (EMS).Methods Clinical manifestations,response to treatment and prognosis of 89 cases with eosinophilia hospitalized at Peking Union Medical College Hospital(PUMCH)during the past nine years were analyzed retrospectively,including four cases conformed with the diagnostic criteria for EMS set by the Centers for Disease Control and Prevention of the US.Results Infection,tumor and other diseases that could cause increased eosinophil count were excluded in all but four cases,with all female,average age of 48.8 years,and mean count of eosinophil of 42.75 percent of blood cells.Multi-organs and systems were involved in the patients,with the most common clinical manifestations of myalgia,skin rash,joint swelling,arthralgias,respiratory symptoms and paresthesia.Patients responded well to prednisone at acute phase of illness,but prognosis was poor as severe pulmonary hypertension occurred.Occurrence of EMS closely associated with environmental factors and special attention should be paid to patient's history of medication and diet in its diagnosis.Conclusion EMS is characterized by the increasing of eosinophils and myalgia,while multi-organs and systems were involved.Glucocorticoid plays an important role in the treatment at acute phase of illness.
4.The inverse PCR:good method for deletion breakpoint mapping

Objective To map the deletion breakpoint of 9p21 in breast cell line MCF-7 use the inverse PCR .Methods After di-gestion ,ligation and PCR reaction ,the breakpoint was confirmed by sequencing .Results The deletion breakpoint started at chr9 :21819532 and ended at chr9 :21989622 with a small insertion of ACTGG ,which was consistent with the result confirmed by the long range PCR .Conclusion The inverse PCR is one good method for deletion breakpoint mapping and suitable for large sample size detection .
5.Clinical Observation on Intractable Rotavirus Enteritis with Wenpi Yiyang Decoction

Objective To evaluate the clinical effect of treating intractable rotavirus enteritis with Wenpi Yiyang Decoction. Methods Sixty cases with intractable rotavirus enteritis were recruited in this study and divided into a treatment group and a control group randomly.Traditional Chinese drugs of warming spleen and invigorating yang were adopted in the treatment group(30 cases).Results The total effective rate was 93% in the treatment group and 49% in the control group.There was no significant statistical difference between two groups(P<0.05).Conclusion There was good clinical effect of treating intractable rotavirus enteritis with Wenpi Yiyang Decoction.
6.Analysis of reproductive performance after loop electrosurgical excision procedure for the management of CIN3

Objective To investigate the effects of loop electrosurgical excision procedure(LEEP) on preg- nancy and delivery outcome in patients with CIN3.Methods Fifty-eight patients with CIN3 who were considering future pregnancies underwent LEEP from January 2003 to December 2006,and their following reproductive perfor- mances and delivery outcomes were observed.Results The mean time of the follow-up survey was 21 months(range from 6~42 months).There were 34 cases getting intrauterine pregnancies and 4 cases getting ectopic pregnancies. Among the 34 cases of intrauterine pregnancies,4 terminated by induced abortion and 1 by delayed abortion in early stage.21 cases delivered termed baby vaginally and 7 underwent cesarean section.No cervical dystocia had been re- ported.No disease recurrence had been found until the end of follow-up survey.Conclusion Loop electrosurgical ex- cision procedure is a safe and effective treatment for patients with CIN3.It will not effect the pregnancy and delivery outcome after LEEP.
7.The observation of clinical efficacy of rituximab in combination with DHAP chemotherapy in patients with relapsed non-hodgkin lymphoma

Objective To study the safety and efficacy of the combination of rituximab and DHAP chemotherapy in patients with relapsed non-Hodgkin lymphoma(NHL). Methods To choose 16 patients with relapsed B-cell NHL as research objects, they were divided into two groups: treatment and control group. The treatment group(8 cases) received the combination of rituximab and DHAP chemotherapy; the control group (8 cases) received combination chemotherapy. The treatment was first infused a dose of rituximab (375 mg/m2) the day before receiving the DHAP chemotherapy, and such treatment being repeated for 4 ~ 6 times every other week. The DHAP chemotherapy lasts 4 ~ 6 cycles and twenty-one days in each cycle. The control group was primitively administrated 4 ~ 6 cycles of CHOP chemotherapy, methotrexate, Ara-C and etoposide as additional drug. Results The complete response and overall response rate were 75 % and 100 % while in the control group were 37.5 % and 62.5 %. 8.The Effect of Quercetin Targeted p53 Gene and Protein Expression in NB4 Cell Lines

Objective In order to study the value of drug treatment for leukemia, quercetin was delivered to the cultured NB4 cell line. MethodsUsing RT- PCR and western blot, we studied the expression of mutant p53 gene and protein after inducing 24, 48, 72, 96 and 120h by the quercetin with different concentrations (30, 60, 90μmol/L). Results We verified the results that quercetin with concentration of 30 ~90μmol/L could successfully inhibit the expression of mutant p53 protein, but the mutant p53 gene did not have. ConclusionThat data suggested that quercetin would be an effective method for the therapy of leukemia.
9.Quality Study of Marketed Rose Bengal Agar Culture Medium for Drug Control

Objective:To investigate overall quality status of rose bengal agar culture medium for drug control in China to provide technical support for the drug standard revision. Methods:Four quality control microorganisms, namely Aspergillus niger, Candida al-bicans, Staphylococcus aureus and Escherichia coli, were used to evaluate the quality of rose bengal agar culture medium from 7 domes-tic and foreign manufacturers. Results:Including the control medium, the pH value of all the 8 media was different. The growth rate of Aspergillus niger and Candida albicans in the seven tested media were all above 0. 7 without significant difference (P>0. 05). The selectivity factors of Staphylococcus aureus in the seven media and the control medium was more than 6. The eight media showed differ-ences in the selectivity factors of Escherichia coli. Among them, A, B, C, E, G and the control medium had no inhibitory ( selectivi-ty) , while D and F showed certain selectivity. Conclusion:The rose bengal agar media from different manufacturers for drug control show similar growth-promoting ability with different pH value and selectivity to Escherichia coli.
10.Clinical value of serum HDL-C in the evaluation of synthetic function of the liver

Objective To evaluate the clinical value of serum high density lipoprotein cholesterol (HDL-C)in the evaluation of synthetic function of the patients with liver diseases.Methods Abbott Aeroset automatic biochemical analyzer was used to detect serum HDL-C level in control group (30 cases) and liver disease group(87 cases).T test was used to analyze the differences of serum HDL-C level in liver disease group and control group,different liver disease groups and control group,and different liver disease groups.Results HDL-C level was (1.28 ±0.20)mmol/L in the control group,(0.77 ± 0.15)mmol/L in the liver disease group,and (0.81 ± 0.13)mmol/L in the hepatitis group,(0.68 ± 0.14)mmol/L in the liver cirrhosis group,(0.54 ± 0.05)mmol/L in the liver cancer group.The HDL-C level in liver disease group and hepatitis group,liver cirrhosis group,liver cancer group was significantly lower than that in the control group(t =15.5569,14.2463,45.4393,25.6344,all P < 0.01).The HDL-C level in liver cirrhosis group and liver cancer group was significantly lower than that in hepatitis group (t =3.6583,16.9057,all P < 0.01).The HDL-C level in liver cancer group was significantly lower than that in liver cirrhosis group(t =4.4103,P < 0.01).Conclusion Serum HDL-C level can be used to evaluate synthetic function of the liver,the lower of serum HDL-C level,the more serious liver synthesis function is impaired.
