3.Treatment of rheumatoid arthritis by tumor necrosis factor inhibitor
Basic & Clinical Medicine 2006;0(09):-
Tumor necrosis factor alpha(TNF-?) is one of the key proinflammatory factors in driving and attending chronic inflammatory process in rheumatoid arthritis(RA).TNF-? acts on different kinds of cell in synovial membrane, such as synoviocytes, macrophages, osteoclasts and chondrocytes, which can produce metalloproteinase, collagenase, stromelysin and so on,further induce pannus formation, joint inflammation, bone erosion and cartilage degradation. The TNF inhibitors have been proved by a lot of clinical trials to be an important new group of agents that significantly improve symptoms and signs, induce remission, reduce objectively measured damage in chronic inflammatory conditions, such as RA.There are three kinds of TNF inhibitors: Etanercept,Infliximab and Adalimumab.Etanercept is a fusion protein of human IgG and two p75 TNF receptors.Infliximab is an IgG1 monoclonal antibody(a chimera of human constant and mouse variable regions).Adalimumab is a humanized IgG1 monoclonal antibody with fully human constant and variable regions. The side effects include injection site infusion reactions, infection, lymphoproliferative disease, demyelinating disease, and SLE-like syndromes.

China Oncology 2001;0(03):-
Background and purpose:Hsp90 is cell chaperone protein that interacts with many proteins, but itself can not degrade its client proteins.Hsp90 inhibitor can inhibit tumor cell proliferation, induce cell apoptosis, cell growth arrest and increase the degradation of Hsp90 client proteins like Survivin. In order to explore the co-effects of Hsp90 inhibitor 17-AAG and Survivin on LoVo cells and the possible mechanisms,we observed the effects of 17-AAG on proliferation and cycles of LoVo cells and the protein level of Survivin. Methods:LoVo cells were treated with 17-AAG.The cell proliferation inhibition rate was evaluated by MTT assay. The cell cycle was detected by ? ow cytometry. The expression of Hsp90 client protein Survivin was detected by Western blot.Results:17-AAG time-dose-dependently inhibit the proliferation of LoVo cells, after 100 ng/ml,500 ng/ml and 800 ng/ml 17-AAG exposure for 24 hrs,the cell proliferation inhibition rate was 21.00%,40.81%,60.34% respectively, after exposure for 48 hrs,the cell proliferation inhibition rate was increased to 27.29%,48.17%,80.97% respectively, after exposure for 72 hrs,the cell proliferation inhibition rate was to 34.45%,67.81%,88.42%;17-AAG arrested cell cycle, when LoVo cells were exposed to 100 ng/ml 17-AAG for 72 hrs,the cell ratio of G0/G1 phase was(61?3)%,when to 500 ng/ml for 72 hrs, cell ratio of G0/G1 phase was increased to(74?3)%,compared to(48.2?0.8)% LoVo cells without 17-AAG(P
5.Anti-light aging effect of Polygala tenuifolia saponins on the skin
Chinese Journal of Biochemical Pharmaceutics 2017;37(1):18-22
Objective To study the anti-aging effect of total saponins of Polygala tenuifolia (PTS). Methods The effects of total saponins of Polygala tenuifolia on oxygen free radicals were investigated by the methods of scavenging hydroxyl radical, superoxide radical and DPPH oxygen free radicals in vitro. The photoaging model of mice skin was established to observe the effect of total saponins of Polygala tenuifolia on mouse skin pathology. Results In the experiment of antioxidation in vitro, the total saponins of Polygala tenuiflora significantly scavenged hydroxyl free radicals (P<0.05, P<0.001) and superoxide radical (P<0.01) at 6.0 and 12.0mg/mL, DPPH free radicals (P<0.001). In the light aging experiment, the light aging model of mice skin was established successfully in group B (model + medium tea oil). Compared with group B, the density of the skin epidermis in D group (30 min before irradiation, 6mg/mL PTS) was thickened, the elastic fibers were arranged uniformly and tightly, the collagen fibers proliferated significantly and densely, and the inflammatory cells infiltration was not found. The treatment effect was most remarkable in group G (30min after irradiation, PTS 12mg/mL PTS), and the skin of the mice was thickened, the elastic fibers were evenly arranged, the collagen fibers had some hyperplasia, evenly distributed, and there was no infiltration of inflammatory cells, which had significant therapeutic effect. Conclusion The total saponins of Polygala tenuifoliahas significant antioxidant activity and anti - light aging effect.

8.Study on the effect of genistein and adriamycin on the proliferation of human ovarian cell A2780 and growth of tumor bearing nude mice
Chinese Journal of Biochemical Pharmaceutics 2014;37(4):28-32
Objective To study the effects of genistein and adriamycin on the proliferation of human ovarian cell A2780 and growth of tumor bearing nude mice. Methods The ovarian cancer cell A2780 suspension were inoculated into nude mice (n=40 )to establish the animal models with ovarian tumor. These model rats were randomly divided into four groups, saline group and different genistein concentrations combined with adriamycin treatment group (high-dose,middle-dose and low-dose group)by intraperitoneal injection once a week. After treatment for four weeks, the ovarian tumors in all rats were peeled off and weighted for the measurement of tumor inhibition rate. The expression of c-myc in ovarian tumors were detected by immunohistochemistry. In vitro, after affected by different genistein concentrations combined with adriamycin, the expression of c-myc in A2780 cell of each group were detected by western blot. The proliferation of A2780 cell were determined by MTT method, and the invasiveness of cell by cell adhesion and transwell assay, the apoptosis rate and cell cycle were measured by flow cytometry. Results The tumor inhibition rates in middle-dose (40.26 ± 4.25)% and high-dose groups[,(44.81 ±5.74)%])were significantly lower than saline group (P<0.05).Immunohistochemistry results showed that c-myc monoclonal antibody in middle-dose and high-dose groups can significantly inhibit the activation of c-myc in ovarian carcinoma tissue.Compared with control group, the expression of c-myc in A2780 cell were significantly decreased, the proliferation and invasiveness of cell were inhibited, cells in G0/G1 phase were significantly increased(P<0.05)and in S phase were(P<0.05)decreased,and the apoptosis were induced by genistein combined with adriamycin in middle-dose and high-dose (P <0.05 ).Conclusion Genistein combined with adriamycin in certain doses could inhibit the proliferation and invasiveness of ovarian carcinoma, and induce its apoptosis. The mechanism may be related to the decreased expression of c-myc.
9.Risk factors and prognosis of cardiac arrest in children
Chinese Pediatric Emergency Medicine 2014;21(4):241-243
Cardiac arrest in children is one of the most critical clinical situations, posing a serious threat to life. With the progress of cardiopulmonary resuscitation and the advanced life support technology, children's cardiac arrest survival rate is higher than before, but many of the survivors suffer from neurological sequelae.Since the causes of out-hospital cardiac arrest and in-hospital cardiac arrest are different, their prognosis and influence factors are also different.Death risk factors for cardiac arrest in children are cancer, hepatic insufficiency, acute renal failure, sepsis, systemic inflammatory response syndrome, etc.Cardiopulmonary resuscitation,as the main emergency treatment of cardiac arrest,is also one of the main factors affecting survival of cardiac arrest child.
