2.Effects of maternal hypercholanemia on fetal myocardium in rats

Objective To explore the effects of maternal hypercholanemia on the myocardium changes in rat fetus. Methods Thirty clean SD female rats were equally randomized to three groups after mating successfully.From the 13th to 20th day of gestation,group A and B were injected injected with sodium chloride(NS) as control.Total bile acid(TBA) and cardiac troponin I(cTnI) were measured in the maternal and fetal serum on the 21st day when all rats were killed.Fetal cardiac muscle cells were also collected for examination with light microscope and electronic microscope.Results (1)TBA in maternal and fetal serum were(22.32±8.12)μmol/L and(28.84±8.06) μmol/L,respectively in group A,(9.77±3.56)/μmol/L and(9.34±3.54) μmol/L in group B,and (3.60±1.78) μmol/L and(3.95±1.19) μmol/L in group C.Significant differences were found among groups(P<0.01).(2)Fetal death rates were significantly different among the three groups (P<0.05),with 30.11%,16.85%,and 7.05%,respectively.(3)Fetal cTnl were also found significant difference among groups(P<0.01),with(19.98±7.75)ng/ml,(11.41±3.64)ng/ml and(4.38±1.19)ng/ml,respectively.(4)The integrated scores of fetal necrosis area were significantly different in three groups(P<0.05),with 1.92±0.43,1.36±0.37 and 0.44±0.12,respectively.(5)Under electronic microscope,the number density of mitochondria in group A was lower than that in group C(P<0.05)while the average volume of mitochondria was larger in group A (P<0.05).The average volume of mitoehondria in group B was larger than that in group C(P<0.05) while no difference was found with regard to the number density between the two groups.The number density and average volume of myofibril in group A were lower than those in group C(P<0.05).The number density of myofibril in group B was higher than that in group C(P<0.05) while no difference was found with the average volume.(6)Positive correlations were found in maternal TBA,fetal TBA,fetal cTnI and the integrate of fetal necrosis area when comparing every two of the above factors. Conclusions Fetal myocardium is impaired obviously in hypercholanemia rats.The serum level of TBA and cTnI in fetal rats are positively correlated with each other.
3.Protective Effect of Erythropoietin on Anoxia of Motoneurons after Spinal Cord Injury in Rats

Objective To observe the protective effect of erythropoietin (EPO) on anoxia of motoneurons after spinal cord injury in rats.Methods The motoneuron was dissociated from neonate Wistar rats and cultivated in vitro with deprivation of oxygen in the medium. The changes of morphology were observed by phase-contrast microscopy, and the expression of EPO receptor (EPOR) was detected with Western blotting. The survival rate of motoneuron was measured with methyl thiazolyl tetrazolium (MTT) assay, and the level of lactate dehydrogenase (LDH) was observed. Results Compared with the control group, the expression of EPOR and the level of LDH increased (P<0.01),and the survival rate of motoneuron decreased (P<0.01) in the anoxia group. Compared with the anoxia group, the expression of EPOR and the level of LDH decreased (P<0.05), and the survival rate of motoneuron increased (P<0.01) in the EPO group, which was related with the level of EPO. Conclusion EPO could relieve the damage of motoneuron caused by anoxia, especially by upregulating EPOR.
4.Effect of dual track continuous health education on patients with endometrial cancer after operation

Objective:To investigate the effect of individualized and generalized dual track continuous health education on patients with endometrial cancer after operation.Methods:From January 2019 to January 2020, 96 patients with endometrial cancer in the Second Affiliated Hospital of Henan University of traditional Chinese Medicine were enrolled. They were all treated with surgery. According to the random number table method, they were divided into the control group and the study group, with 48 cases in each group. However, during the whole study period, some patients were affected by various factors, and the final number of patients in the study group was 45 cases (dual track continuous health education combined with individualization and generalization) and 45 cases in the control group(routine continuous health education). Before the intervention and 3 months after the intervention, the obstetrics and Gynecology nurses who had received relevant training adopted self-made health knowledge awareness questionnaire, self-care ability assessment scale (ESCA), Medical Coping Style Questionnaire (MCMQ) and simple quality of life scale (SF-36).The patients' mastery of disease knowledge,self-care ability,coping style and quality of life were evaluated.Results::After the intervention,the awareness of health knowledge was 42.23±2.58 in the study group and 35.71±3.26 in the control group, and the difference was statistically significant ( t value was 4.508, P<0.01). After the intervention, the self-care skills, self responsibility, self-concept, health knowledge level and total score of the study group were 9.45±1.58, 11.82±1.63, 61.12±6.52, 10.56±1.33 and 31.83±2.21 respectively, while those of the control group were 5.34±0.75, 7.85±0.92, 68.23±6.45, 6.35±0.94 and 19.54±1.26 respectively, and the difference between the two groups was statistically significant ( t value was 11.025-16.444, P<0.05). After the intervention, the scores of facing,avoiding and yielding in the study group were 19.39±3.29, 10.58±2.40, 9.20±3.12 respectively, and those in the control group were 15.97±3.77, 14.56±3.00, 13.02±2.50 respectively, with significant difference between the two groups( t value was 4.459, 6.762, 6.220, P<0.01). After the intervention, the scores of physiological function,physical pain,social function,mental health,emotional function,vitality and general health in the study group were 80.23±8.12, 22.19±2.63, 44.98±4.25, 76.25±7.64, 69.58±6.23, 70.05±7.22 and 78.31±7.52 respectively, while those in the control group were 59.63±5.14, 52.37±5.38, 41.16±4.73, 57.28±5.36, 55.25±5.27, 46.89±4.52 and 48.96±4.37 respectively, the difference between the two groups was statistically significant( t value was 3.630-11.870, P<0.05). Conclusions:The implementation of individualized and generalized dual track continuous health education in patients with endometrial cancer after surgery has a significant effect, can significantly improve the patient's mastery of disease health knowledge, enhance their self-care ability and positive coping ability, so as to improve their quality of life, which is worthy of clinical promotion.
7.Optimization of Purification and Isolation of Punicalagin from Pomegranate Peel

Objective: To establish the methods for extraction and isolation of punicalagin from pomegranate peel, and further study the purification and quantification of punicalagin. Methods: Using an ultrasonic-assisted extraction method, punicalagin in pomegranate peel was extracted at room temperature by 50% ethanol with 20-fold volume of raw material. The content of punicalagin in the crude extract was determined by HPLC. To optimize the purification process of punicalagin, static adsorption and desorption experi-ments were employed to study five kinds of macroporous adsorbent resins (D101, A8-8, NKA-9, HPD-100 and HPD-500) for the one with the highest purification efficacy of punicalagin. In addition, the technical parameters of the macroporous adsorbent resin were opti-mized to obtain punicalagin with higher purity. Punicalagin was further separated and purified by using a reverse phase MCI GEL CHP20P column. Results:HPD500 resin showed the best ability to absorb and separate punicalagin in among five kinds of macro-porous adsorbent resins. The best technical parameters were as follows:the mass concentration of sample solution was 15 mg·ml-1 , the loading amount was 2BV, the pH was 2 and the eluting solvent was 8BV of 30% ethanol. With the best process as described a-bove, the content of punicalagin extracted from pomegranate peel increased from 10. 3% to 30. 7%. The obtained punicalagin could be further purified to 61. 3% from 30% in ethanol eluate by the reverse phase MCI GEL CHP20P column. Conclusion:HPD500 resin is the most effective in the purification of punicalagin from pomegranate peel, and the content of punicalagin can be dramatically increased after the purification by a reverse phase MCI GEL CHP20P column. The optimized process shows good reproducibility and stability.

Objective To investigate dyslipidemia and the use of lipid-lowering drugs in patients with chronic kidney diseases(CKD)and analyze the related risk factors.Methods A total of 397 out-patients with CKD in our Renal Department from March 2006 to March 2007 were enrolled;258 healthy people with similar age and sex were taken as the control group.Lipid levels were compared between the two groups and related factors were analyzed.Results Among 397 CKD patients,there were 52.1% patients with high triglyceride(TG),41% with high total cholesterol(TC),18.8% with low high-density lipoprotein cholesterol(HDL-C),47.6% with high low-density lipoprotein cholesterol(LDL-C).Compared with controls,rates of hypertriglyceridemia and low high-density lipoprotein cholesterol in CKD group were higher(P

ObjectiveTo investigate the relationship between the oxidative injury induced by low glucose and mitochondrial membrane potential in HUVEC-12 cells. Methods Human umbilicalvein endothelial cells HUVEC-12 were cultured in low concentration glucose for 4 h.Cell viability of HUVEC-12 cell was assessed with MTT assay.Dihydroethidium (DHE) was used as a reactive oxygen species (ROS)capture, which was detected the mean fluorescence intensity of samples and Rhodamine 123 as a fluorescence detector was to measure the level of mitochondrial membrane potential (MMP) in cells.Results Comparing to HUVEC-12 cells viability in 5.5 mmol/L glucose group (96.80 ±3.20)%, cells exposed to 2.8 mmol/L glucose group (66.40 ± 1.60) % and 0 mmol/L glucose group (58.93 ± 1.67) % were decreased by 32% and 40% respectively (P < 0.01).ROS level of 5.5 mmoL/L glucose group, 2.8 mmol/L glucose group and 0 mmol/L glucose group were 0.59 ± 0.02, 0.74 ± 0.04 and 0.88 ± 0.05,respectivdy, increased by 25% in cells exposed to 2.8 mmol/L glucose and by 48% in cells without glucose exposure comparing to 5.5 mmol/L glucose group (P <0.01) ; MMP levels of 5.5 mmol/L glucose group,2.8 mmoL/L glucose group and 0 mmoL/L glucose group were 148.83 ± 3.51, 271.07 ± 19.54 and357.74 ±51.32 respectively, increased to 1.8 times in cells exposed to 2.8 mmol/L glucose and to 2.4times in cells without glucose exposure comparing to 5.5 mmoL/L glucose group (P < 0.01).Conclusion Low glucose leads to injury in HUVEC-12 cells, which is probably induced by the oxidative stress via the increasing MMP.
10.Butylphthalide improves cerebrovascular reactivity in patients with severe intracranial internal carotid artery stenosis

Objective To investigate the effect of butylphthalide on cerebrovascular reactivity (CVR) in patients with severe intracranial internal carotid stenosis.Methods The patients with severe intracranial internal carotid stenosis diagnosed by cerebral angiography in Nanjing Stroke Registry Program from January 2010 to November 2010 were randomly divided into either the butylphthalide group or the control group.All patients received the oral drugs for ischemic cerebrovascular disease that met the guideline criteria,and butylphthalide 20 mg,3 times a day orally was added in the butylphthalide group.The CVR was evaluated by transcranial Doppler and CO2 inhalation challenge test,and reexamined 3 months after treatment.Results A total of 81 patients met the inclusion criteria,11 (13.6%) were lost to follow-up,and finally 70 (n =35 in each group) were included in the analysis.The mean age of the patients was 55.2 ± 9.0 years,47 of them were male and 23 were female.The degree of vascular stenosis was 72% to 99% (mean 79.4% ± 9.5%).The CVR was improved significantly after 3 months of treatment in the butylphthalide group than that in the control group (P =0.007),but it was not improvedsignificantly in the control group (P =0.330).All the patients did not have ischemic stroke events during the medication.No serious adverse reactions occurred in the butylphthalide group.Conclusions Butylphthalide may improve the CVR in patients with severe intracranial internal carotid stenosis.
