1.Effect of ganglioside on nerve function in hypertensive cerebral hemorrhage after operation

Objective To investigate the effect of ganglioside on nerve function in patients with hypertensive cerebral hemorrhage after operation. Methods 90 cases with hypertensive cerebral hemorrhage after operation from March 2014 to March 2016 in our hospital were divided into control group and observation group, the control group was treated with routine treatment, the observation group was given ganglioside nerve cell nutrition therapy, the two groups of patients after treatment of clinical efficacy, neurological function score, ADL score, hematoma, edema were observed.Result The total effective rate(91.11%) in the observation group was significantly higher than that in the control group(66.67%)(P <0.05), the difference was statistically significant.After 7 days and 28 days after treatment, the volume of hematoma and edema in the observation group was significantly decreased, and the control group had obvious advantages compared with the control group ( P <0.05 ).Compared with seventh days and twenty-eighth days after treatment, the NIHSS score and ADL score of the observation group were significantly better than the control group ( P <0.05 ) , the difference was statistically significant.Conclusion The application of the ganglioside in the treatment of hypertensive cerebral hemorrhage can significantly improve the neurological function, improve the quality of life of patients and clinical treatment effect, has good clinical application value.
2. The effect of age on local recurrence of patients with breast cancer undergoing breast-conserving therapy

Objective: This study was designed to evaluate the effect of age on local recurrence after BCT (breast-conserving therapy) in patients with breast cancer. Methods: The clinical records of 548 female patients with breast cancer undergoing BCT between January 2000 and December 2007 were analyzed retrospectively, and the follow-up of these patients was performed. The univariable and multivariabe analyses were conducted to evaluate the potiential factors related to local recuurence after BCT. Results: For 548 patients, the recurrence rate within 5 years after BCT accounted for 4.01% (22/548). There were statistical differences in local recurrence rate among different ages (< 45 years vs ≥ 45 years, P = 0.041), tumor sizes (T1 vs T 2 vs T3, P = 0.014) and histologic grades (grades I-II vs grade III, P = 0.003). The multivariable analysis displayed that the factors of younger age (< 45 years, P = 0.013) and higher histologic grade (grade III, P = 0.046) were independent factors in prediction of poor prognosis of breast cancer after BCT. Conclusion: Younger age (< 45 years) is a reasonable justification to consider more aggressive treatment to prevent local recurrence in patients with breast cancer after BCT. 5.Complications of obesity in children

With the growth of people's living standard,the prevalence of childhood obesity is increasing year by year.Meanwhile,the adverse effect caused by obesity occurred more frequently as well.Therefore,obesity in childhood has become a critical issue in public health area.Complications of obesity may lead to enormous implications on children,including their endocrine,cardiovascular,digestive system,etc.,which will have an impact on the body growth in the adolescence.Obesity not only has a huge threat to children's physical and psychological health,but also has a long-term effect on body health condition in the future.Consequently,it is significant to diagnose the occurrences of complications of children,and implement the guiding and curing at an early stage.
6.Reform of laboratory diagnostics teaching mode for students of clinical medicine

7.Construction of vascular endothelial growth factor modified muscle-derived stem cells and its expression

Objective To construct the lentiviral vector encoding vascular endothelial growth factor gene and detect the vascular endothelial growth factor (VEGF) expression in muscle-derived stem cells (MDSCs). Methods To culture MDSCs and detect the CD34,CD45,Bcl-2 and Desmin expression in MDSCs by immunofluorescence. A cDNA encoding VEGF gene was amplified by PCR. This fragment was cut with EcoRI and BamHI and ligated with an EcoRI- and BamHI-reated lentiviral vector pCDH-CMV-MCS-EF1-copGFP. Then DNA sequencing analysis was performed to confirm successful construction of pCDH-CMV-MCS-EF1-copGFP -VEGF. The expression of VEGF was confirmed using enzyme-linked immunosorbent assay (ELISA), Western blot, and Real-time PCR analyses. Results The pCDH-CMV-MCS-EF1-copGFP-VEGF lentiviral vector was constructed successfully. When MOI values in the transfection efficiency MDSCs by FCM. were 1,5,15, the transfection rate reached to 16.7%, 45.6%, 66.3% and 85.6% respectively. When MOI value was of 20, the rate was up to 90.1%. Real-time PCR, Western blot and ELISA showed stable expression of VEGF in MDSCs. Conclusion We successfully constructed lentiviral vector carrying the VEGF and stable expression in MDSCs.
8.Application of Clinical Data Interchange Standards Consortium (CDISC) Standards to Electronic Data Capture System

Data acquisition is one of the key links that influencing the quality of clinical research.Electronic data capture system (EDC) embodies the advantages of saving time,manpower and material resources and improving efficiency and reliability by data acquisition.CDISC has established worldwide industry standards to support the electronic acquisition,exchange,submission and archiving of clinical research data.The application of CDISC standards to EDC system is favorable in ensuring the validity and standardization of clinical data.This paper takes the Oracle's OC/ RDC (Oracle Clinical / Oracle Remote Data Capture) system as an example to discuss the application of CDISC standard to EDC system from the two aspects:direct application and indirect application.We suggest that data collection should be taken into account during the design phase of a clinical trial,and the CDISC standard be applied at the CRF design stage.A design for eCRF takes time and effort by the combination of EDC system and CDISC standard,while thoughtless design may collect the wrong data.Therefore,it is suggested that a specialized personnel should be put in charge of eCRF design and maintenance during the operation of EDC system,and a set of standardized eCRFs based on CDISC standard and standard operating procedures should be built in one organization.
