1. Prostacyclin signal pathway: Research advances
Journal of International Pharmaceutical Research 2010;37(4):276-278
Prostacyclin (PGI2) is a member of the prostaglandins family and the main product of arachidonic acid metabolism by cyclooxygenase and prostaglandin synthase. PGI2 mediates cellular activity through binding to G-protein coupled receptor, prostacyclin receptor, therefore leading to activation of adenylyl cyclase, accumulation of cAMP and activation of PKA. PGI2 plays an important role in vascular biology such as regulating vasodilation, platelet aggregation, vascular permeability, and vascular smooth muscle cell proliferation. At high concentrations, PGI2 is able to interact with peroxisomal proliferator-activated receptor, a class of nuclear receptors, and modulate specific cellular functions such as angiogenesis. This paper reviews the research advances in PGI2 synthesis and signal pathway.

Osteoarthritis (OA) is a complex chronic progressive disease attacked by biological and mechanical factors and a result from the anabolic and catabolic imbalance in chondrocyte, subchondral bone and extracellular matrix(ECM). Etiology and pathological of OA are not yet entirely clear. The degradation and destruction of collagen II caused by matrix metalloproteinase -13 (MMP-13) is considered the core factor in the occurrence and development of OA. The research of MMP-13 inhibitor provide ideas and methods for the treatment of OA. In this article, the role and determination of MMP-13 in OA and the development prospect of MMP-13 inhibitor in the treatment of OA research progress were reviewed.

Objective To analyze the etiological factor and genetic feature of a familial hemophagocytic lymphohistiocytosis patient with PRF1 mutation (FHL2) with human herpesvirus 7 (HHV7)infection and its family constellation. Methods Clinical characteristics, laboratory examinations of a FHL2 case with HHV7 infection were reported. HHV1-HHV8 virus DNA was screened by PCR; NK cell function was analyzed by flow cytometry; PRF1 gene mutations were analyzed by PCR and direct sequencing, structure of mutant PRF1 proteins were analyzed using ExPasy and I-TASSER server and genetics pedigree were analyzed. Results The patient's HHV7 viral was detected positive with DNA copy number of 350/106 peripheral nucleated cells. Flow cytometry analysis showed decrease both in proportion of perforin positive NK cells and perforin protein expression. Genetic testing showed PRF1 biallelic heterozygote mutations (c. 503G > A/p. S168N and c. 1177T > C/p. C393R) and pedigree analysis showed they were inherited. The patient was then treated with antivirus therapy, dexamethasone and VP16 therapy, but only achieved partial response. The patient was then followed by human leukocyte antigen 10/10 allele identical nonconsanguinity allogeneic hematopoietic stem cell transplantations (allo-HSCT) and soon the successful implantation of donor hematopoietic cells and persistent recovery was achieved. The patient was now surviving without recurrence for 9 months after allo-HSCT. Conclusions FHL is prone to be misdiagnosed as lymphoma. Genetic analysis of related gene mutation and herpes simplex virus detection will help in early and accurate diagnosis. Allo-HSCT is a fundamental treatment of FHL.

OBJECTIVETo compare the effects of olecranon plate and tension band in the treatment of olecranon fracture.
METHODSSixty-three patients with olecranon fracture were treated with olecranon plate and tension band. Thirty-five patients (21 male and 14 female, aged from 15 to 62 years, with an average of 48.9 years) were involved in tension band group and 28 patients (20 male and 8 female, aged from 25 to 67 years, with an average of 50.6 years)were involved in plate group. According to the Delee, JC classification in 1984, there were type 1A in 5 cases, type 1B in 8 cases, type 2 in 17 cases, type 3 in 7 cases and type 4 in 1 case in tension band group; there were type 1A in 3 cases, type 1B in 5 cases, type 2 in 6 cases, type 3 in 11 cases and type 4 in 3 cases in plate group.
RESULTSSixty-three cases were all followed up from 6 to 15 months. According to the criterion of PANG Gui-gen, the results were excellent in 20 cases, good in 7 cases, fair in 1 case and no bad in plate group; the results were excellent in 23 cases, good in 10 cases, fair in 21 case and no bad in tension band group. The therapeutic effect of two groups has no significantly statistic difference.
CONCLUSIONBoth olecranon plate and tension band are the effective treatments for olecranon fracture. The selection of two methods should depend on to the state of fracture.

ObjectiveTo evaluate the curative effect difference between Tong's anterior mesh suspension and modified tension-free vaginal tape-obturator (TVT-O). Methods75 SUI cases were random divided into two groups: Group A (35 cases) were accepted Tong's anterior mesh suspension and group B (40cases) were accepted modified tension-free vaginal tape- obturator. Curative effects, operation safety, and complications and so on, were recorded and compared. ResultsThere had no statistical differences between these two groups on operation time, bleeding lose, operation injury, immediate postoperative urinary retention andcure rates, but the operation expense of group A was cheaper than group B [ (980. 74 ±212.45)yuan vs (2879.06 ±467. 13)yuan , P <0.05). ConclusionThe curative effect between two methods were similar. It's a little complicated and fit to be popularized in large hospital to modified tensionfree vaginal tape-obturator. Tong's anterior mesh suspension was performed under completely direct vision, not through pelvic cavity, and it was an economic, convenient and easy way, which not only fit to be popularized in basic hospital, but also a surgical remedial measure to the failures who accepted these operations, such as TVT, SPARC, IVS, MONARC, TOT, TVT-O, etc.

OBJECTIVEGene sequence data were clustered to explore evolution lineages of H3 antigen of influenza A virus.
METHODSAll data of H3 RNA sequence in NCBI Genbank and Influenza sequence database were downloaded and aligned in ClustalX while two step cluster method were applied to explore the data.
RESULTSAll sequences were aggregated into ten clusters, while seven of them mainly were human virus. Human virus and avian/other mammal virus were separated into different clusters distinctively, but coexisted into same clusters with swine virus. Time and host distribution were very distinctive in these clusters, but no geographic distribution features were found.
CONCLUSIONWith the interaction of human immunity system, H3 antigen mutated significantly every 5 - 7 years, and the speed of mutation had accelerated with the application of influenza vaccines in recent years. Mean while, human and swine influenza virus were not separated distinctly between clusters indicating that they had short inheritance distance. Result showed again that swine served as the mixer for antigenic recombination of different influenza virus.
