Objective To investigate the effects of lipid microspheres prostaglandin E1(Lipo PGE1) on subarachnoid hemorrhage(SAH).Methods Ninty-three patients with SAH were randomly divided into control group(treated with nomal drugs) and Lipo PGE1 treatment group(treated with nomal drugs and Lipo PGE1).Changes in neuroimaging, biochemical indexes and incidence of cerebral vasospasm were measured. Results A lower incidence rate of cerebral vasospasm was observed in Lipo PGE1 treatment group(P0.05).The levels of vWF and GMP-140 were significant lower in the Lipo PGE1 treatment group than those in the control group after treatment for 3 and 7 d(P

Objective To investigate the changes of somatosensory evoked potential(SEP)in subarachnoid hemorrhage(SAH)and in-fluence from lipid microspheres prostaglandin E1(Lipo PGE1). Methods Ninty-three patients with SAH were randomly divided into control group(treated with nomal drugs) and Lipo PGE1 treatment group(treated with nomal drugs and Lipo PGE1).Clinical outcomes and changes of SEP before and after the treatment were observed. Results After the treatment, the latency of N20 wave was prolonged in both groups(P

Objective To study the alterations in neurobehavior of rats with diffuse axonal injury(DAI) and to evaluate the potential role of basic fibroblast growth factor(bFGF). Methods A weight-drop device was employed to build the DAI model. An injection of bFGF subdurally and subcortically was given to the bFGF therapeutic group(n=60).Besides,normal control(n=20) and injured control group(n=60) were established. The elevated walkway test, prehensile traction test, sensorimotor integration test and the Morris' water maze test were adopted to examine the motor and memory abilities. After the implantation of skull electrodes,P3-like potential was explored in rats before and after injury. Results After DAI,the scales of the elevated walkway test, prehensile traction test, sensorimotor integration test and the Morris' water maze test were decreased, and the rats in the bFGF therapeutic group presented a better behaviour in the early stage. The latency of P3-like potential prolonged significantly in rats with DAI,with the P3-like potential in the injured control group longer than that in the bFGF therapeutic group(P

Objective To study the alterations in pathology and immunohistochemistry in rats with diffuse axonal injury(DAI) and the effects from basic fibroblast growth factor(bFGF). Methods A weight-drop device was employed to produce DAI in rats.In the treatment group(n=60),bFGF was injected subdurally and subcortically.Besides, normal control group(n=20) and injury-control group(n=60) were also established. The pathological changes were observed by light microscopy and electromicroscopy, and the expression of brain-derived neurotrophic factor(BDNF),growth associated protein-43(GAP-43) and glial fibrillary acidic protein(GFAP) were also examined by immunohistochemistry. Results Typical pathological changes were observed in the basal portion of pons, corpus callosum and white matter of cerebral hemisphere in the rats with DAI.And an upregulation of GFAP,GAP-43 and BDNF was also found. In the treatment group, better outcomes of pathological changes were observed.bFGF increased the expression of BDNF and GAP-43,while inhibited the immunoreactivity of GFAP. Conclusion Topical application of bFGF can improve brain tissue regeneration and speed function recovery in rats with DAI,though its long-term effect warrants further study.

This study investigates whether kappa-opioid receptor and ORL1 receptor may interact to form a heterodimer. In immunofluorescence and co-immunoprecipitation experiments, differentially epitope-tagged receptors, colocalization and heterodimerization of kappa-opioid receptor and ORL1 receptor were used and examined in primary culturing rat neurons, Chinese hamster ovary (CHO) or human embryonic kidney 293 (HEK293) cells. The results show that fluorescence of both kappa-opioid receptor and ORL1 receptor were overlapping in primary culturing hippocampal and cortical neurons. Similarly in co-expressing CHO or HEK293 cells, HA-KOR and Myc-ORL1 were almost exclusively confined to the membranes, revealing extensive colocalization. When Flag-KOR and Myc-ORL1 were co-expressing in CHO cells, heterodimerization was identified to have the ability to co-immunoprecipitate ORL1-receptors with kappa-opioid receptor and vice versa. In the current study, further evidence was provided for the direct interaction of two subtypes of opioid receptors, kappa-opioid receptor and ORL1-receptor, to form the heterodimerization. The finding represents the novel pharmacological mechanism for modulation of opioid receptor function as well as diversity of G protein-coupled receptors.

Objective To determine the contents of free ferulic acid and total ferulic acid in different processed products of Rhizoma Chuanxiong. Methods With methanol-formic acid(95∶5) as the extraction solvent for free ferulic acid and with methanol-2% NaHCO3 water solution(95∶5) as the extraction solvent for total ferulic acid, ultrasonic method was used for the extraction. The contents of free ferulic acid and total ferulic acid were determined by HPLC,and the chromatographic conditions were as follows: C18 column(250mm?4.6mm,5?m),the mobile phase consisting of acetonitrile-1%acetic acid solution(20∶80),the detecting wavelength at 320nm,flow rate being 1.0 mL/min and detection at room temperature. Results The average contents of free ferulic acid and total ferulic acid in Rhizoma Chuanxiong roasted by wine were higher than those in raw and other processed Rhizoma Chuanxiong, and free ferulic acid content in raw and different processed Rhizoma Chuanxiong was lower than the total ferulic acid content. Conclusion The method is simple, accurate and can be used for the quality control standard for Rhizoma Chuanxiong, and the chemical assay of total ferulic acid content would be a better choice of assessing the herbal quality of Rhizoma Chuanxiong.

Objective To investigate the mechanisms and clinical characteristics of fatal asthma attack induced by amiodarone in the elderly, and to discuss its serious potent risks. Methods We reported two cases of fatal asthma attack in the elderly with fast atrial fibrillation induced by amiodarone, and discussed the clinical manifestation and emergency treatment, with literatures review. Results The two patients presented with respiratory arrest. After targeted therapies, including artificial ventilation and hormone injection, the patients were capable of spontaneously breathing, while still in severe asthma attack. And further anti-asthma treatment led to a complete remission. Conclusions Amiodarone may cause extremely severe asthma attack in elder patients with tachyarrhythmia complicated with bronchial hyper-responsiveness diseases such as asthma or chronic obstructive pulmonary diseases.Clinicians should pay great attention to this situation and be prepared to cope with serious adverse reactions.
