1.Imaging Diagnosis of Uveal Melanoma

Objective To investigate CT and MRI manifestations of uveal melanoma as well as their diagnostic value.Methods 33 cases of uveal melanoma proved by operation and pathology were studied retrospectively.CT and MRI scanning were performed in 15 and 18 patients respectively.Results The round occupying lesions within eyeball were the characteristic imaging finding of uveal melanomas, most of them were found in the location of choroid membrane. These lesions appeared as homogenous density on CT and short T 1 and short T 2 intensity on MRI. MRI was superior to CT because it could provide more accurate information such as location, shape,extension,and differentiation of the complication (retina detachment) from lesion itself. The preoperation diagnostic accuracy of CT was 53% compared with that of 88% of MRI. Conclusion MRI is the optimal examination method in confirming suspected uveal melanoma.

3.The Study of MSCT Volume Reconstruction in the Ear Lesions

Objective To investigate the value of volume reconstruction ( VR ) in showing the ear lesions and to compare with otherpost-processed methods.Methods 15 patients with hearing loss underwent MSCT ultra-high resolution scan with collimation 0.5 mm,pitch 0.5~0.875 and increment 0.1mm.An additional enhanced CT was performed in 10 of 15 cases. The various reconstructions including VR , MPR, SSD, VE, Ray Sum and MIP/MinIP were performed for the comparison of their ability in showing the ear lesions and the invasion of the auditory ossicals and the inner ear of the adjacent tumors. Results VR was the most optimal way, in showing the ear lesions,successionally in descending order was VR, MPR, SSD, MIP/MinIP. Ray Sum and VE were only suitable for showing the lesions of the inner ear and auditory ossicles respectively.Conclusion The data acquired from the scan of MSCT using 0.5 mm collimation can be used in various reconstruction methods including VR, MPR, SSD and MIP/MinIP to display the lesions of the ear and the invasion of the ear fine structures from the plane and three-dimensional view. VR is the best way, which can display the relationship between tumor and facial/auditory nerve, labyrinth .
4.Comparison of health related quality of life of ileal conduit versus orthotopic ileal neobladder

Objective To compare the health related quality of life (HRQoL) of ileal conduit versus orthotopic ileal neobladder using the FACT-BL scale.Methods One hundred and thirty patients underwent radical cystectomy and urinary diversion for invasive bladder cancer from Jan 2006 to Dec 2010 at our hospital.According to different urinary diversions,patients were divided into ileal conduit (IC) and orthotopic ileal neobladder (NB).HRQoL was assessed using FACT-BL.The following scores calculated from the FACT-BL questionnaire and list:physical well-being ( PWB ),social/family well-being ( SWB ),emotional well-being(EWB),functional well-being( FWB),overall bladder-special subscale and total FACT-BL were included.A high FACT score indicated a high level of HRQoL.The medical records of each patient were then reviewed.Clinical parameters including age,sex,type of diversion and pathological status were recorded and analyzed.Results A total of 94 questionnaires were collected,of these patients 50 had an IC and 44 had an NB.Among the respondents,the age at surgery was significantly younger in NB group than IC group (P =0.014 ).While the percent of males,follow-up years and pathological stage T3 or greater revealed no significant difference.The NB group got significantly higher scores in PWB,SWB,EWB and FACT-BL than the IC group.Which indicated that the HRQoL was higher in patients following orthotopic ileal neobladder.Conclusions The HRQoL was better in patients who followed orthotopic ileal neobladder than those who followed ileal conduit.This may indicate that orthotopic ileal neobladder should be considered first when making a decision about which urinary diversion should be used.
5.Compared empirical evaluation of artery stiffness by echo-tracking and M-mode ultrasonography

Objective To study the worth of echo-tracking and M-echo in evaluating artery stiffness comparatively.Methods Fifty two rabbits were divided into 4 groups randomly:control group 1,hypercholesterinemia group,control group 2,and artherosclerosis group,there were 13 rabbits in each group.The rabbits were bred with normal or 2% cholesterol animal feeds according to their groups.The femoral artery blood pressure were measured with catheter.Artery stiffness of abdomen aorta of every rabbit was evaluated by echo-tracking and M-echo at the baseline and the end of the study by 2 doctors,and each doctor did twice.Results All the rabbits of artherosclerosis group had lipid plaques in the thorax and abdomen aortas,and the interior elastic fibers board under the plagues were disrupted.Echo-tracking was more stable and sensitive than M-echo in evaluating artery stiffness.pressure-strain elastic modulus(E?) and stiffness index(?) were sensitive stiffness indexes,but arterial compliance(AC) was not.The indexes of artery stiffness evaluated by echo-tracking with short view of the artery were stable too,but not sensitive compared with long view.E? was relative to diastolic diameter of artery and HDL-C positively(P0.05).Conclusions Echo-tracking is more stable and sensitive than Mecho in evaluation of artery stiffness,long view of the artery should be applied in the evaluation of artery stiffness with the technique of echo-tracking.E? and ? were sensitive artery stiffness indexes,but AC was not.

7.Protective effect of total flavonoids of epimedium on the kidney in experimental diabetic rats.

OBJECTIVETo investigate the influence of total flavonoids of epimedium (TFE) on the streptozocin (STZ)-induced kidney injury in diabetic rats and discuss the possible mechanism.
METHODSDiabetes was produced by a single injection of streptozocin (40 mg/kg, iv) in male SD rats. The rats were randomly divided into three groups (n = 10): control group, model group and TFE group (100 mg/kg, ig). Animals were sacrificed 12 weeks later. The level of blood glucose, blood urea nitrogen (BUN) and creatinine (Cr) as well as the renal index were determined. Detect the specific biochemical of renal tissue: superoxide dismutase (SOD), malondialdehyde (MDA). Use masson staining to observe the morphology of the renal tissue. Immunohistochemistry was employed to determine the protein levels of transforming growth factor-beta1 (TGF-beta1).
RESULTSCompared to control group, the enhancement of blood glucose, renal index, BUN and Cr was found in model group, which was significantly attenuated by treatment with TFE. Meanwhile, elevated MDA level in renal tissue as well as decreased SOD activities in renal tissue were significantly remitted by TFE. Furthermore, TFE decreased the expression of TGF-beta1.
CONCLUSIONTFE can evidently relieve renal damage in rats with diabetic nephropathy induced by STZ, which might be related to antioxidation and modulating the expression of TGF-beta1 protein.

ObjectiveTo optimize human Hexastatin gene,to express,purify protein and conduct activity experimental research,and to provide a theoretical basis for further study of Hexastatin.MethodsHuman Hexastatin gene was optimized and synthesized.It was connected to the pET28a expression vector,induced to express by isopropyl β-D-1-thiogalactopyranoside(IPTG),and optimized induction conditions.After the ultrasonication of bacterial cells and inclusion bodies,the recombinant fusion protein was purified with Ni-NTA chromatographic column,analyzed and identified by SDS-PAGE and Western Blot,and conduct activity experimental research in vitro by MTT.ResultsConstructed production was pET28a-Hexastatin expression plasmid.The human Hexastatin protein was expressed in E.coli BL21 the high level and accounted for 45.1% of the total bacterial protein.The purification of recombinant protein purified with Ni-NTA chromatographic column reached 90%,and the concentration was 80 μg/ml.Human Hexastatin protein can restrain the growth of C6,MCF-7 and human vascular endothelial cell (HMEC) cells,and inhibition ratio reach to 72.9%±3.6%,48.8%±2.9%,52.7%±2.5%,respectively through MTT test.ConclusionThe optimized human Hexastatin protein was expressed successfully,which confirmed the inhibition to tumour cells.It is a new way for anti-angiogenesis therapy of tumour.
