1.Establish of animal model of nonarteritic anterior ischemic optic neuropathy

Nonarteritic anterior ischemic optic neuropathy (NAION)is a common optic neuropathy seriously affecting the visual function in the middle-aged and elderly population.However,its pathogenesis is not completely clear,and therefore its treating efficacy is dissatisfactory.The current study on NAION is focused on the establishment of suitable animal model and the pathogenesis.In recent years,the relatively ideal animal models(including rodent and primate) have been established by photodynamic methods,which make people have more in-depth understanding on the pathophysiologic mechanism of NAION and lay the basis for the research of therapies.The selection of experimental animals,various induction methods and existing problems in the creation of NAION animal model were reviewed and analyzed in this artical.
3.Effect of allicin on the radiosensitivity of human pancreatic carcinoma BXPC3 cells

Objective To study the effect of allicin on the growth and radiosensitivity of human pancreatic carcinoma BXPC3 cells.Methods BXPC3 cells were exposed to X-rays in the presence or absence of allicin.Cell proliferation was measured by MTT assay.Cell cycle distribution and apoptosis were detected by flow cytometry assay.Cell radiosensitivity and the influence of allicin on it was evaluated by colony formation assay.The expressions of Bax and Bcl-2 proteins were assayed by RT-PCR and Western blot.Results IC50 values of allicin on cell growth were 76.24,58.34 and 43.58 μmol/L under 12,24 and 48 h treatment,respectively.Treatment of cells with allicin obviously inhibited cell growth after irradiation and hence increased radiosensitivity (t =2.74,P < 0.05).This treament also enhanced radiation-induced cell cycle arrest at G2/M phase (t =11.41,P <0.05),apoptosis induction (t =12.36,P < 0.05),and Bax expression (t =4.83,P < 0.05),but it decreased Bcl-2 expression (t =3.69,P < 0.05).Conclusions Allicin could inhibit cell growth,induce cell cycle arrest and apoptosis via Bax/Bcl-2 pathway and hence increases radiosensitivity of BXPC3 cells.
4.Evaluation of SYTO13 as fluorescent dye for high resolution melting based single nucleotide polymorphism genotyping

Objective To evaluate the utility of fluorescent dye SYTO 13 for high -resolution melting ( HRM) detection in single nucleotide polymorphism ( SNP) genotyping and its clinical application . Methods This is a performance verification study .36 genotype defined samples were divided into three groups:SNP rs3125734 C>T (class Ⅰ SNP) ,rs255758 A>C (class ⅡSNP) and rs688C>T.These samples were used to evaluate SYTO 13′s SNP genotyping capability of class ⅠSNP, classⅡSNP, and two PCR products of different lengths (52 and 107 bp) covering the same SNP of rs688C>T.The commercial HRM dye of LCGreen Plus was used as the control .The genotyping capability is indicated by the Tm difference(ΔTm) between wild type and homozygous mutant genotypes .The Tm differences between wild genotype and homozygous mutant genotype were compared using the Independent Samples t test.Paired t test was used to evaluate genotyping capability of the two dyes .The clinical applicability is evaluated by synchronously performing PCR amplification and HRM analysis on thirty -five randomly selected DNA samples with known genotypes of the three SNPs .Results The SNPs of class Ⅰ and class Ⅱ can be genotyped directly and clearly with SYTO13 (ΔTmclas Ⅰ =0.36 ±0.05,tclas Ⅰ =14.827,Pclas Ⅰ =0.000;ΔTm clas Ⅱ =0.42 ±0.110,tclasⅡ =9.539,Pclas Ⅱ =0.000).The classⅠSNP genotyping results was better using SYTO13 (ΔTmSYTO13 =0.39 ±0.027), while the SNP genotyping for small amplicon did not discriminated clearly in this study .Long amplicons of class ⅠandⅡSNPs can be identified directly except for several samples which can be genotyped accurately after having performed reexamination .Conclusion SYTO13 can apply for HRM analysis of genotyping classⅠand ⅡSNPs with long amplicon and for clinical routine detection.
5.To study the clinical pathway of enhanced recovery after surgery in patients undergoing laparoscopic hysterectomy

Objective To investigate the effect of clinical pathway of enhanced recovery after surgery in patients undergoing laparoscopic hysterectomy.Methods All patients with uterine fibroids,uterine adenomyosis,cervical lesions and endometrial lesions were selected in Qinghai Provincial Traffic Hospital during the period of January 2015 to December 2016.Among them,108 cases undergoing laparoscopic hysterectomy were analyzed and enrolled in the study.They were randomly divided into two groups.The observation group was treated with clinical pathway of enhanced recovery,while the control group received routine clinical pathway.The general situation,clinical pathway related indicators were recorded and compared between the two groups.Results The average age,BMI,abdominal surgery history and disease composition of the two groups were not statistically significant (P > 0.05).The exhaust time [(19.5± 5.6)h vs (24.2 ± 7.5) h],activity time [(17.2 ± 7.5) h vs (26.4 ± 5.3) h],indwelling catheter time [(18.1 ± 3.9) h vs (30.5 ± 4.7) h],average hospitalization days [(5.2 ± 1.1) days vs (6.3 ± 1.7) days] and hospitalization expenses [(13 688.2 ± 709.6)yuan vs (15 793.4 ± 1 021.3)yuan] of the observation group were less than those of the control group,with statistically significance difference (P < 0.05).Conclusions Clinical pathway of enhanced recovery after surgery can speed up the rehabilitation of laparoscopic hysterectomy,improve the patient's medical experience and shorten the average length of stay.
6.HPLC determination of the related substances in erdosteine.

An HPLC method was established for the determination of the related substance in erdosteine. Waters ODS-SunFire (250 mm x 4.6 mm ID, 5 microm) column was used, the mobile phase was composed of methanol-acetonitrile-0.01 mol x L(-1) citric acid (20:4:76, the pH value was adjusted by triethylamine to 2.5). The flow rate was 1 mL x min(-1). The detection wavelength was 254 nm. The related substances in the sample of erdosteine taken were calculated by self control with or without the response factor of impurity relative to that of erdosteine. Under the chromatographic condition developed, the impurities in erdosteine were isolated well. The detection limit was 0.2 microg x mL(-1) (signal/noise = 3) by principal component calculated. The method can be adopted to control the related substances in erdosteine.

7.Taohong Siwu Decoction regulated functions of endothelial cells and treated arteriosclerosis obliterans: an experimental study.

OBJECTIVETo discuss the effect of Taohong Siwu Decoction (TSD) in regulating functions of endothelial cells and treating arteriosclerosis obliterans (ASO).
METHODSThe ASO model was prepared by using high-fat diet plus intimal injury. They were randomly divided into the model group (n = 10), the normal control group (n = 9), the low dose TSD group (group A, n = 12), the middle dose TSD group (group B, n = 10), and the high dose TSD group (group C, n = 9). Eight weeks after modeling, the limb blood perfusion was observed using laser Doppler flowmetry. The arterial morphology was observed using light microscope and transmission electron microscope. The number of circulating endothelial cells (CECs) was determined using Percoll density gradient centrifugation method. Serum levels of TNF-alpha, IL-1, ET-1, and NO were detected using double antibody sandwich assay of enzyme linked immunosorbent assay (ELISA).
RESULTSThe ASO rat model was successfully established. Blood lipids levels significantly increased, the blood perfusion of left hind limbs significantly decreased, the number of CECs in the peripheral blood significantly increased, the arterial lumen was irregularly narrowed, the ultra-structure of vessel walls was damaged, serum levels of TNF-alpha, IL-1, and ET-1 significantly increased, and the serum level of NO significantly decreased in the model group, showing statistical difference when compared with the normal control group (P < 0.01). Compared with the model group, significant improvement in the aforesaid indices was shown in group B and C (P < 0.05, P < 0.01).
CONCLUSIONSThe injury and abnormal functions of endothelial cells is an important pathological process of ASO. As an effective recipe for treating ASO, TSD could protect vascular endothelial cells and improve the secretion function of vascular endothelial cells.

8. Clinical research on the simulation accuracy of zygomatic reduction using an L-shaped osteotomy by Proplan CMF software

Objective:
To evaluate the surgical prediction accuracy of Proplan CMF software for zygomatic reduction surgery using L-shaped osteotomy.
Methods:
Pre-and-postoperative 1-year CBCT data of 26 patients with zygomatic arch hypertrophy were imported in Proplan CMF software during 2014 Jan. to 2016 Jun., the 3D models were reconstructed for simulation of L-shaped osteotomy, characteristic landmarks were selected and 3D point measurement system was established. The measurement result were analyzed by one-way ANOVA. Meanwhile, the overlap color grading charts of preoperative and simulated images were also observed.
Results:
The facial width, bilateral zygomatic process angle and facial width index were [(128.56±2.72) mm, (106.87±2.53)°, (108.56±3.02)°and 1.41±0.03] in postoperative result, [(129.49±2.26) mm, (108.68±2.40)°, (108.85±3.02)°and 1.42±0.03]in simulated result and [(135.45±2.45) mm, (102.50±2.60)°, (103.41±2.56)°and 1.48±0.05] in preoperative result, with significant difference between preoperative and postoperative result, or between preoperative and simulated result (
9.Construction of Nonenzymatic Glucose Sensor Based on One Step Preparation of Poly(3,4-ethylenedioxythiophene) Graphene Composite Materials by Pulse Potentiostatic Method

The Poly(3,4-ethylenedioxythiophene) graphene ( PEDOT-rGO) composite film was prepared by one step pulse potentiostatic method, based on the π-π* interaction and the polymerization reaction of hydrogen bond between the graphene ( rGO) and the aromatic ring of 3,4-ethylenedioxythiophene ( EDOT) monomer. Then the nickel nanoparticles ( NiNPs ) were electrodeposited on composite film to obtain a composite film-modified glassy carbon electrode ( NiNPs/PEDOT-rGO/GCE ) . The catalytic performance of NiNPs/PEDOT-rGO/GCE towards the oxidation of glucose was studied by electrochemical method. The experimental results showed that the NiNPs/PEDOT-rGO/GCE could be used as a nonenzyematic sensor for detection of glucose. This method had high stability, good selectivity, wide linear range ( 2 μmol/L-58 mmol/L) and low detection limit (0. 7 μmol/L), realizing the rapid and sensitive detection of glucose.
10. Finite element study of the mandibular angle width changes in retrogression surgery by bilateral sagittal split ramus osteotomy

0bjective:
To establish a three-dimensional finite element model of mandible and study the transverse displacement of proximal segment after Bilateral Sagittal Split Ramus Osteotomy (BSSRO) with different retrogression amounts during mastication.
Methods:
DICOM data of a skull model were processed with MIMICS and ANSYS software, reconstructing the 3D model including the teeth and temporomandibular joint in order to simulate BSSRO and evaluate the transverse displacement of proximal segment with different retrogression amounts during mastication.
Results:
The mean of proximal segment width change were 2.955 mm and 3.490 mm, when retrogression amounts of distal segmentwere 3 mm and 8 mm, respectively.No significant difference between the two groups were found (P=0.131). Meanwhile the displacement color scale of the 3D finite element models showed that the apparent transverse displacement distribution of the proximal segment was measured around the gonial area, decreased from the exterior to the interior.
Conclusions
The mandibular angle width was significantly expanded right after BSSRO. The masticatory muscle system and single cortical fixation system played an important role in expanding the width of proximal segment. However there was no correlation between the widening effect and retrogression amounts of distal segment of mandible.
