1. Signal transduction mechanism of macrophages polarization induced by methionine enkephalin

OBJECTIVE: To investigate the signal transduction mechanism of macrophages polarization induced by methionine enkephalin (MENK), which can promote tumoricidal responses in vitro. METHODS: The phenotype of macrophages were assessed by the quantitative analysis of key surface molecules on macrophages with flow cytometry (CD64, CD206). The expressions of NF-kB/STAT6 signal transduction were analyzed with Western-blotting. RESULTS: MENK(10-12 mol · L-1) could significantly decrease the expression of CD206 (P < 0.01), and at the same time, it could increase the expression of CD64 significantly (P < 0.01). MENK could increase the expression of NF-κB compared with MENK-untreated group. Furthermore MENK could inhibit the activation of STAT6 which is mediated by IL4 (P < 0.05). CONCLUSION: MENK could inhibit the activation of STAT6 signaling pathway mediated by IL4. Meanwhile, MENK could increase the expression of NF-κB and be conducive to the combination of NF-kB sites with M1-type cytokines, consequently MENK could induced the conversion of macrophages from M2 to M1 phenotype effectively.
2.Therapeutic Effect of Entecavir Combined with Vitamin E in the Treatment of Chronic Hepatitis B Complicated with Non-alcoholic Fatty Liver

Objective To explore the clinical efficacy of entecavir combined with Vitamin E in treating patients with Chronic hepatitis B complicated with non-alcoholic fatty liver.Methods Retrospective analysis was done by reviewing the clinical data of 103 patients, who suffered with chronic hepatitis B complicated with non-alcoholic fatty liver and treated in our hospital from 2015 to 2016.The patients were divided into the control group with 61 cases and the observation group with 42 cases based on different therapies.The control group was treated with only entecavir while the observation group was treated with entecavir and Vitamin E, and both the courses of treatment lasted for 6 months.The negative conversion ratio of HBV-DNA, levels of ALT, TBIL, ALP, the disease state and the adverse reactions of the two groups were compared before and after treatment.Results The negative conversion ratio of HBV-DNA of the observation group was obviously higher than that of the control group, and the patients of the observation group showed significant improvement.Meanwhile, the levels of ALT, TBIL and ALP in the two groups were both lower than before treatment.Compared with the control group, those indexes in the observation group were significantly lower (P<0.05).Conclusion Entecavir combined with Vitamin E for the treatment of chronic hepatitis B complicated with non-alcoholic fatty liver could significantly decrease the viral infections so that the state of patients can be improved.
4. Active part of Achyranthes promotes maturation and regulates the functions of dc in tumor microenvironment

OBJECTIVE: To study the active part of Achyranthes bidentata promotes maturation and regulates the functions of DC in tumor microenvironment. METHODS: The total saponins and the polysaccharides from dry powder of Achyranthes bidentata were extracted and identified. The tumor microenvironment by co-culturing breast cancer MCF-7 cells with DCs, and add polysaccharides, total saponins or total saponins with polysaccharides of Achyranthes bidentata to stimulate DCs were established. The expression of surface molecules on DCs and the killing activity of cytotoxic T lymphocytes (CTL) induced by DCs by Flow Cytometry were detected. ELISA was used to detect the levels of IL-β, TNF-α, IL-10 and IL-12 in the culture supernatant. Detect the protein expression of JAK, p-JAK, STAT and p-STAT3 in DCs by Western blot. RESULTS: In the tumor microenvironment, the active parts of Achyranthes bidentata, total saponins and ABPS, alone or combined used, can trigger JAK/STAT3 signaling pathway of DCs through TLR4 and Dectin-1 receptor and upregulate the expression of surface molecules, such as CD80, CD86 and CD40, to induce the maturation and differentiation of DCs, promote the secretion of TNF-α, IL-12, IL-10 and IL-β. CONCLUSION: As an effective adjuvant, the active parts of traditional Chinese medicine can enhance the effect of DC vaccine to correct the immune deficiency in tumor microenvironment, activate tumor-specific CTLs and then produce specific anti-tumor effects.
5.GSH changes in HepG2 cells infected by dengue virus or stably expressing viral protein E and NS3

Objective To investigate the changes of intracellular GSH content in HepG2 cells infected with dengue virus(DV)or stably expressing E or NS3 protein.Methods HepG2 cells were cocultured with DV or inactivitated DV,l h later the viral supernatant was removed.The infected HepG2 cells were collected 10,20,30,40,60 min,and 2,6,12,24,48 h after the beginning coculture and intracellular GSH content was detected by spectrophotometry.Intracellular GSH content was also detected in HepG2 cells stably expressing E protein/NS3(pReceiver-E/HepG2,pReceiver-NS3/HepG2)and those containing empty plasmid(pReceiver/HepG2).Results GSH content showed a decreasing tendency after DV-2 infection.The lowest values were seen 30 min and 2,24 h after infection,which were of significant difference in comparison with those in inactivated DV infected HepG2 cells as well as at other time points.GSH levels in pReceiver-E/HepG2,pReceiver-NS3/HepG2 were significantly lower than those in pReceiver/HepG2.Conclusion DV-2 infection might lead to the GSH depletion in HepG2 cells,and GSH lost from HepG2 cells might undergo a three-step process:virus adsorption/penetration,protein synthesis and budding.E or NS3 protein stably expressed in HepG2 cells can also decrease the GSH levels.

The present study identified chemical constituents of Honghua Xiaoyao Tablet (HXT) and explored its biological connotation and characteristics on the premenstrual syndrome (PMS) treatment from the "disease-syndrome-symptom" association network. UHPLC-Q Exactive Orbitrap HRMS technology was applied to analyze the chemical constituents in HXT. According to the composition principles, the compatible herbs of HXT were divided into the Shugan Jieyu group, Huoxue Tiaojing group and Yiqi Jianpi group. The candidate targets of the corresponding prescriptions of HXT efficacy groups were collected from the Pharmmapper database and Integrative Pharmacology-based Research Platform of Traditional Chinese Medicine (TCMIP) v2.0. The gene set related to the clinical symptoms included in Traditional Chinese and Western Medicine diagnosis and treatment standards were obtained from SoFDA, GeneCards, DisGeNET, MalaCards and literature published. The "HXT candidate targets-PMS (liver depression, Qi stagnation, and blood stasis syndrome) genes" network was constructed based on the gene interaction information, and further, the core network targets were screened out by topological characteristics of calculating network, and the functional exploration was carried out based on Kyoto Encyclopedia of Genes and Genomes (KEGG) for exploring the therapeutic advantages in PMS treatment of HXT efficacy groups, which were further verified experimentally

BACKGROUND:Conventional rehabilitation evaluation call not provide more objective data for analysis,so all innovative method to monitor rehabilitation of the cervical spine is introduced.OBJECTIVE:The monitoring rehabilitation system of the cervical spine develops independently under Visual Basic 6.0 environment.It can gather both video and photos at dynamic and static states.According to the suitable feature points of the video,the system could calculate characteristic parameters,such as moving angles,distances,speed,acceleration,and integral curves.Thus,the system could create patients'database.RESULTS:The system could calculate different kinds of patients'characteristic parameters through data recording.Through the objective comparison of characteristic parameters,the turning flexibility of head and controlling capability for head turning could be reflected.CONCLUSl0N:The design is an innovation in the monitoring rehabilitation systems of the cervical spine.It makes up the shortcomings of conventional rehabilitation evaluation and objectively reflects the rehabilitation status of patients.
8.Association between functional genetic polymorphisms of IL-1B and IL-1RN and susceptibility to gastric cancers

Objective To study the association between functional genetic polymorphisms of IL-1B(T-31C,C-511T),IL-1RN and the susceptibility to gastric cancers. Methods A case-control study was conducted in 180 gastric cancer cases and 308 age-and sex-matched cancer-free controls.Genotypes were detected by PCR-restriction fragment length polymorphism(PCR-RFLP) assays,and association between genotypes,environmental factors and risk of gastric cancers were determined. Results IL-1B T-31C was in strong linkage disequilibrium with IL-1B C-511T(D'=0.862,R2= 0.721,P=0.000).Multivariate logistic regression analysis revealed that the variant genotypes of IL-1B T-31C and C-511T were not significantly associated with risks for gastric cancers(adjusted OR,0.95 and 95% CI,0.62-1.47 for IL-1B T-31C;and adjusted OR,0.85 and 95% CI,0.55-1.31 for IL-1B C-511T).The variant genotypes(1/2,2/2) in IL-1RN were associated with a non-significantly increased risks for gastric cancers(adjusted OR,1.32 and 95% CI,0.71-2.36) in all subjects and with a significantly increased risks for gastric cancers in subjects with H.pylori infection(adjusted OR,2.03 and 95%CI,1.02-4.80).Conclusion The functional genetic polymorphisms of IL-1RN may contribute to the risks of gastric cancers in high-risk population,particularly in those with H.pylori infection.
