1. Optimization of processing method for Lonicera macranthoides of Guizhou by multi-index evaluation

Objective To investigate the effects of different processing methods on the appearance, quality components and processing efficiency of Lonicera macranthoides that produced in Guizhou Province, in order to provide basis for the optimization of processing methods. Methods Eleven processing methods were used such as the traditional method group (sun drying, sun drying after steaming, drying in shade, and drying in shade after steaming), constant temperature drying group (50, 60, 70, and 80 ℃) and variable temperature drying group (50→60 ℃, 50→70 ℃, 50→80 ℃). Then the indicators such as color, aroma, moisture, total ash, acid insoluble ash, chlorogenic acid, total saponins, processing time, and drying rate were measured. Results According to all the indicators, the comprehensive correlation degree of the 50 ℃ and the variable temperature drying method was between 0.94 and 0.96, which was much higher than other methods, and the comprehensive correlation degree of 50→60 ℃ degrees was the highest, reaching 0.959 8. Conclusion In conclusion variable temperature drying method is a better processing method for L. macranthoides. This method can be used as an important technical means for large-scale processing.
2.Correlation between patient's age and expression of Ki-67 or microvessel density in the tissues of human hepatocellular carcinoma

Objective To investigate the correlation between patient' s age and expression of Ki-67 or microvessel density in primary hepatocellular carcinoma.Methods The expression level of Ki-67 and the microvessel density in the hepatocellular carcinoma tissues were detected by immunohistochemical method,and the correlation between age and expression of Ki-67 or microvessel density were analyzed.Results In 48 cases of hepatocellular carcinoma tissues,Ki-67 expressed to varying degrees,but the positive rate was 83.3 % (40/45),and there was no expression in corresponding noncancerous tissues (P < 0.01).Microvessel density marked by CD34 in cancer tissues and noncancerous tissues were 32.17±8.06 and 10.83±2.85 respectively.Microvessel density of cancer tissues was higher than noncancerous tissues (P < 0.01).Both levels of Ki-67 expression and microvessel density in hepatocellular carcinoma were negatively correlated with the age (r =-0.418,P =0.003 and r =-0.443,P =0.002 respectively).Conclusion Both levels of Ki-67 expression and microvessel density in hepatocellular carcinoma are negatively correlated with patient' s age,which may suggest worse prognosis for the younger patients.
3.Construction,Identification and Expression of Recombinant Eukaryotic Vector pCAG-IRES-SHIP-GFP on Porliferation of Leukemia Cell Line K562

The aim was to construct and identify the mammalian expression vector of pCAG-IRES-SHIP-GFP and to detect whether it could express in human acute leukemia cell line K562.The cDNA fragment of SHIP obtained by RT-PCR was inserted into pCAG-IRES-GFP.The recombinant plasmid was confirmed by restriction enzyme digesiton,PCR and DNA sequecing.pCAG-IRES-SHIP-GFP was transfected into K562 cells with lipofectamine 2000.The expression of SHIP was determined by GFP fluorescence and Western blot analysis.FQ-PCR was used to quantitate SHIP mRNA.The expression of p-Akt,Akt were determined by Western blot.PI were tested by flow cytometry and MTT to verify whether exogenous SHIP could inhibit proliferation of K562 cells.The results showed that the correct constrution of the recombinant plasmid pCAG-IRES-SHIP-GFP has been shown by restriction enzyme digestion,PCR and DNA sequencing.pCAG-IRES-SHIP-GFP could express SHIP protein in K562 cells.The K562 cells viability after transfected with SHIP gene droped down.Western blot analysis showed that phospha-Akt308 and Akt473 were reduced to 38.7% and 68% respectively.It was concluded that the vector of pCAG-IRES-SHIP-GFP has been successfully constructed and it can be expressed in K562 cells.The expression of exogenous SHIP gene can lead to apoptosis of K562 cells by down-regulating the p-Akt expression.What found here might be one of the mechanisms involved in the pathogenesis of leukemia.
4.Correlation between Organ Weight and Body Weight in Outbred Wuzhishan Mini-Pigs

Objective To measure the organ weight and body weight of outbred Wuzhishan mini-pigs (WZSP), and calculate the organ coefficient and the linear and multiple linear regression equations between body weight and major organ weights. Methods 30 common WZSP (16 males and 14 females) were chosen,the body weight and 7 organ weights were determined, and the organ coefficients were calculated. The organ coefficients between males and females were compared. The correlation and regression analysis was performed using the SAS software. Results The coefficient of heart had remarkably significant difference between males and females (P<0.05).The linear analysis showed that there were apparent linear correlations between body weight and all the organ weights except for the stomach of males and the lung of females.The weights of liver and kidney showed great influence on the body weight of males, while the body weight of females relied on the weights of heart, liver and kidney greatly. Conclusion Differences of organ coefficients are not significant between males and females,and there are linear relationships between body weight and some major organ weights.
5.DICOM in radiotherapy.

Digital Image Communication in Medicine (DICOM) defines a standard method to store and transmit digital medical image information, in which there is a piece of implemented protocol named DICOM-RT that specially addresses both the transmission of radiation therapy image data and the ancillary data. In this paper, we firstly introduce the DICOM-RT with the emphases on its components, relationship with radiotherapy and how to produce the DICOM-RT object that refer to some certain radiotherapy information. Then we expatiate on the impact that benefits from applying DICOM-RT to radiotherapy, with an aid to accelerate its application in China.

6.Effects of indomethacin on human choroidal melanoma OCM-1 xenografts in nude mouse

Background Choroidal melanoma(CM)is a common form of primary ocular cancer in adults.It is reported that indomethacin has inhibitory effect on many tumor in vitro and in vivo,but whether it can inhibit the growth of CM has not been published. Objective This study was to investigate the anti-tumor activity of indomethacin on the growth of human CM OCM-1 cell xenografts in nude mice. Methods OCM-1 cells were subcutaneously implanted on 24 SPF female BALB/C.nu/nu nude mice to establish ectopic models of human CM.The nude mice with the tumor 5 mm were randomly divided into 4 groups:untreated group,normal saline solution(NS) group,indomethacin 1 ms/kg group,indomethacin 2 ms/kg group.The 1 mS/kg or 2 ms/kg indomethacin was intraperitoneally injected for 14 consecutive days in indomethacin 1 ms/kg group and indomethacin 2 me/kg group respectively.and 0.2 ml of 2%NS-DMSO was used at a same way in the NS group.No any agent was used as the untreated group.The volume and weight of implanted tumor as well as inhibitory rates of indomethaein on tumor were calculated.The expression of ki67 and survivin proteins were measured with immunohistochemistry,and the expression of survivin mRNA in CM was assessed by RT-PCR. ResuIts The tumor of indomethacin treatment group was reduced in volume and weight with a significant difference between treatment group and control group as well as indomethacin 1 ms/ks group and indomethacin 2 ms/kg group(P<0.05).The inhibitory rate of indomethacin 1 ms/kg and 2 ms/kg for tumor was 22.86%,48.00%respectively.The prolifiration index (PI)of ki67 in these 4 groups were (76.73±3.34)%,(73.30±2.95)%,(55.97±2.24)%,(32.87±2.91)%respectively,and significant difference was found in PI between indomethacin 2 mg/kg group and untreated group or NS group(P<0.05),but there was not significant difference between indomethacin 1 mg/kg and 2 ms/kg group(P>0.05).Compared with the control group,the indomethacin treatment groups showed the decreased expression of survivin protein and mRNA,and significant difference was found between indomethaein 2 ms/kg group and untreated group or NS group(P<0.05),however,no significant difference was found between indomethacin 1 mg/kg and 2 mg/kg group(P>0.05). Conclusion Indomethacin inhibits the growth of CM in nude mice through inhibiting the expression of survivin in the tumor and accelerating cell apoptosis and inhibiting tumor cell proliferation.
7.Cellular expression profile of RhoA in rats with spinal cord injury.

RhoA, a small GTPase, is involved in a wide array of cellular functions in the central nervous system, such as cell motility, cytoskeleton rearrangement, transcriptional regulation, phagocytosis and cell growth. It is not known how spinal cord injury (SCI) affects the expression of RhoA in different nerve cells. In the present study, we investigated the changes of RhoA expression in remote areas of the injury at the 3rd, 7th and 30th day after SCI, which was established by T10 contusion method. Moreover, we examine its expression profile in neurons, astrocytes and microglia. RhoA was found to be weakly expressed in these nerve cells in normal spinal cord. Western blotting showed that, after SCI, the total RhoA expression was up-regulated, and the RhoA expression was increased and peaked at the 7th day. Double immunostaining revealed specific and temporal expression patterns of RhoA in different nerve cells. The expression of RhoA in neurons started to increase at day 3, peaked at day 7 and then decreased slightly at day 30. Expression of RhoA in astrocytes increased moderately after SCI and peaked at day 7. There was no obvious change in RhoA expression in microglia after SCI in remote areas. This study demonstrated that, after SCI, RhoA expression exhibited different patterns with different nerve cells of spinal cord. RhoA expression patterns also changed with time after SCI, and among different nerve cells in the injured spinal cord. These findings can help us better understand the roles of RhoA in SCI.
8.Exploration and Practice in Bilingual Teaching of Medical Organic Chemistry Experiment

Objective Research the bilingual teaching of medical organic chemistry experiment. Methods PBL teaching method was used in bilingual teaching of medical organic chemistry experiment. Results As a result, we improved the effect of bilingual teaching in medical organic chemistry experiment. Conclusion Teaching research found that the teaching mode with a problem-centered and students-orientation approach has improved students' analysis and problem solving skills. During the process of teaching, the students have enhanced their enthusiasm of study.
9.Application of emotion interaction on the clinical nursing education

Objective To evaluate the effects of emotion interaction in the clinical nursing education.Methods 120 nursing students who studied in the emergency department were divided into two groups:experimental group and control group. Each group has 60 students. Added the emotion interaction to the experimental group, but did notion to the control group. One month later, all of the students were evaluated in the clinical technology with SPSS 16.0 software. Results Self-management, learning enthusiasm and selfcontrol were different between the two groups (t = 2. 537, 3.980, 2. 858; P < 0. 05 or P < 0. 01) ; practical skill scores between the two groups were also statistically different (t =7. 562, 5. 496, 6. 590,4. 267;P <0. 01).Conclusions Emotion interaction can inspire the sense of participation and independent study, and students' potential and promote the development of clinical nursing education.
