1.Research progress on retinal protection of bone marrow mesenchymal stem cells

?Bone marrow mesenchymal stem cells ( BMSCs ) is a kind of adult stem cells mainly enriched in bone marrow, which possesses multiple differentiation potential and can differentiate into trans-germinal layer. lt is easy for BMSC to be isolated and cultured, which has the ability of repairing various tissues with efficient proliferation and expression. BMSC could be used as seed cell for the transplantation therapy of retinal disease because of its properties of immunoregulation and neurotrophin secretion. This review focuses on research progress on retinal protection of BMSCs.
2.Vibrio vulni f icus pollution condition in marine products in Zhuhai region and drug susceptibility analysis

Objective To understand the vibrio vulnificus pollution condition in marine products in Zhuhai region and the drug resistance situation to provide the basis for further study of vibrio vulnificus and clinical medication .Methods 89 samples were col-lected from the marine products wholesale market ,pedlars′market and supermarkets in Zhuhai region and the method published by the US Food and Drug Administration (FDA) was adopted to perform the qualitative detection of vibrio vulnificus .The identifica-tion of bacteria and the drug sensitivity test were performed by using the ATB instrument .Results In 89 samples of marine prod-ucts ,vibrio vulnificus was detected in 39 samples ,the total positive rate was 43 .82% .The detection rate of oyster was as high as 60 .87% (28/46) ,followed by marine fish (39 .13% ,9/23) and marine shrimp (20 .00% ,2/10) .The drug sensitive test showed that vibrio vulnificus had the resistant strains to cephalosporins and aminoglycoside drugs .Conclusion The seafood vibrio vulnificus pollution in Zhuhai region can not be ignored .The monitoring of vibrio vulnificus in the marine products and the drug resistance a-nalysis should be strengthened for effectively preventing the outbreak of vibrio vulnificus .
3.Research on serum TSGF levels in healthy people and cancer patients in Guangzhou area

Objective To investigate the results of tumor specific growth factor(TSGF) levels in in healthy people and cancer patients in Guangzhou areas .Methods The serum TSGF levels of 6 615 health check-up receivers and 287 patients with cancer in the hospital from July 2012 to June 2013 were detected .Results In health check-up receivers ,the level and positive rate of TSGF were higher in >40-50 age women .The level and positive rate of TSGF in men were increased with age .The levels of TSGF in pa-tients with lung cancer ,liver cancer and digestive system tumor were higher than that in health check-up receivers(P<0 .01) ,but there has no statistically difference between genital system cancer patients and healthy check-up receivers(P>0 .05) .Conclusion Performing TSGF test in health examination is great significance to the early diagnosis of malignant tumors .The reference value range of TSGF should be set according to different gender and age .
5.Analysis of insulin release pattern in patients with polycystic ovary syndrome and normal glucose tolerance

Objective To investigate the release pattern of insulin after the load of glucose in patients with polycystic ovary syndrome ( PCOS ) and normal oral glucose tolerance.Methods Sixty-three patients with PCOS were undertaken oral glucose tolerance test (OGTT) and insulin release test,while 34 women with normal menstrual cycle served as control.Results Among 63 patients with PCOS,33 cases were obese with body mass index over 25 kg/m2,including 5 with abnormal OGTT.All 30 non-obese patients with PCOS had normal OGTT.The prevalences of insulin resistance were 78.8%,16.7%,and 9.0% in obese PCOS,non-obese PCOS,and control groups,respectively.Abnormal insulin release curve were found in 84.5%,70.0%,and 14.7% of subjects in these 3 groups,respectively.In 58 PCOS patients with normal OGTT,the prevalence of insulin resistance was 44.8%,and 75.9%with abnormal insulin release curve. Among them,body mass index of 32 patients,whose homeostasis model assessment for insulin resistance (HOMA-IR) and fasting insulin remained in normal range,was similar to those of control group [ ( 20.52 ± 2.86 vs 20.01 ± 2.54 ) kg/m2,P>0.05].Conclusion These findings indicate that insulin release test is useful in detecting insulin resistance.Insulin release is elevated in PCOS patients even with normal OGTT.
6.Curing method affecting the formation of oxygen inhibition layer on the surface of resin cement.

OBJECTIVE:
To explore the conversion of resin monomer, the change of inorganic component and the influencing factors on the oxygen inhibition layer formed on the cured surface of resin cement.
METHODS:
Three kinds of resin cement were divided into three groups: (1) light-cured group: RelyX Veneer, NX3 (light-cured), Variolink N; (2) dual-cured group: RelyX U200 Automix, NX3 (dual-cured), Multilink Speed; (3) chemically-cured group, and the above 3 types of dual-cured resin cement cured without illumination could be used as chemically-cured resin cement. Each sample was provided with and without oxygen exposure of two matching surfaces, cured respectively, and the variables of light intensity and illumination time were set in the light-cured group and the dual-cured group. Scanning electron microscopy was used to observe the samples' surface morphology. Energy dispersive spectrometer was used to analyze the samples' composition of surface elements. Confocal Raman spectroscopy was used to measure the monomer conversion of resin cement and to obtain the thickness of the oxygen inhibition layer.
RESULTS:
(1) On the surface of cured resin cement, the weight percentage of oxygen element in the aerobic side was higher than that in the anaerobic side (P < 0.05), and the weight percentage of inorganic element was lower than that in the anaerobic side (P < 0.05). (2) The surface monomer conversion of the cured resin cement on the aerobic surface was significantly lower than that on the anaerobic surface (P < 0.05), and the surface monomer conversion on the aerobic surface and the anaerobic surface was the lowest in the chemically-cured group (P < 0.05), the dual-cured group was the highest (P < 0.05), and the light-cured group was between them. With the increase of light intensity or illumination time, the surface monomer conversion increased (P < 0.05). (3) The thickness of the oxygen inhibition layer was the thickest in the chemically-cured group [(40.27±2.81) μm](P < 0.05), the thinnest in the dual-cured group [(21.87±5.42) μm](P < 0.05) and light-cured group [(23.73±3.84) μm] was between them. With the increase of light intensity or illumination time, the thickness of the oxygen inhibition layer of resin cement decreased (P < 0.05).
CONCLUSION
When resin cement is exposed to oxygen, it will form an oxygen inhibition layer, its surface's inorganic filler is less, the surface monomer conversion is lower. The surface monomer conversion and the thickness of oxygen inhibition layer are affected by curing mode and illumination factors.

7.Clinical characteristics and prognostic analysis of secondary hemophagocytic syndrome in adults

Objective:To investigate the clinical characteristics and prognosis of secondary hemophagocytic syndrome (HPS) in adults.Methods:The clinical data of 43 adult patients with primary secondary HPS in Mianyang Central Hospital of Sichuan Province from January 2014 to June 2020 were retrospectively analyzed. The patient's clinical manifestations and biochemical indicators were summarized. The clinical characteristics of patients between high-level ferritin (≥5 000 μg/L) group and low-level ferritin (<5 000 μg/L) group, lymphoma-related HPS group and infection-related HPS group were compared. Logistic regression model was used for multivariate survival analysis.Results:All patients had fever as the main complaint, and infections were manifested at the onset or during treatment. In all patients, 88.4% (38/43) had hemophagocytosis in the bone marrow; 88.4% (38/43) had varying degrees of cytopenia; 36.6% (15/41) had reduced fibrinogen [median value 0.93 g/L (0.50-1.35 g/L)]; 76.7% (33/43) had elevated aminotransferase. Compared with the low-level ferritin group (19 cases), the high-level group (21 cases) had a lower proportion of monocytes (0.081±0.040 vs. 0.163±0.076, P = 0.016), the incidence of fibrinogen reduction and the incidence of aminotransferase elevation were high [60.0% (12/20) vs. 11.1% (2/18), P = 0.002; 95.2% (20/21) vs. 52.6% (10/19), P = 0.006]. Compared with infection-related HPS patients, lymphoma-related HPS patients had a higher incidence of lymphadenopathy [72.7% (16/22) vs. 27.8% (5/18), χ2 = 8.021, P = 0.005] and a lower incidence of aminotransferase elevation [59.1% (13/22) vs. 94.4% (17/18), χ2 = 4.848, P = 0.028]. Multivariate analysis showed that the high proportion of neutrophils ( OR = 0.886, 95% CI 0.786-0.998, P = 0.046) and high-level albumin ( OR = 0.376, 95% CI 0.156-0.907, P = 0.030) after treatment were protective factors for survival of patients. Conclusions:Among adult patients with primary secondary HPS, the proportion of monocytes in patients with ferritin ≥5 000 μg/L is low, and liver damage and fibrinogen reduction are more likely to occur. Lymphoma-related HPS patients are more likely to have lymphadenopathy, and infection-related HPS patients are more likely to have liver damage. After treatment, a low proportion of neutrophils and hypoproteinemia indicate a poor prognosis in adult patients with secondary HPS.
8.Surface labeling of bone marrow mesenchymal stem cells by biotin-streptavidin

BACKGROUND:Currently, there is a lack of efficient, non-invasive way to transplant stem cels to the target organ or tissue. Exploring a way to guide targeting transplantation of stem cels and to improve the efficiency of stem cel homing is now one of focuses in the field of stem cels research.
OBJECTIVE: To establish a simple and feasible method to chemicaly modify the cel surface using biotin-streptavidin reaction system, and to evaluate the efficiency of this method to label bone marrow mesenchymal stem cels (BMSCs) and its effects on cel biological functions.
METHODS: Passage 3 BMSCs were obtained by whole bone marrow culture method and verified by flow cytometry. Biotin, streptavidin, sulfonated biotin-N-hydroxy-succinimide were used to equip the adhesion molecule ligand, sialyated LewisX (SLeX), to the BMSCs surface. The labeling rate of BMSCs was assessed using fluorescence microscope, the vitality of BMSCs was evaluated by trypan blue staining, and the proliferation of BMSCs was evaluated by cel counting kit-8 assay. Adipogenic and osteogenic inductions were used to evaluate the effect of the method on the multi-differentiation function of BMSCs.
RESULTS AND CONCLUSION: After culture for 2 weeks, passage 3 BMSCs were obtained and confirmed by expressing CD90, CD29 and lack of CD34, CD45. Biotin, streptavidin, sulfonated biotin-N-hydroxy-succinimide were successfuly used to equip sialyated LewisX (SLeX) to the BMSCs surface and had minor effects on the vitality, proliferation, and differentiation of BMSCs. This method was simple for surface modification and had a high modification rate of 88%. The homing of BMSCs modified by this method to the target organ or tissue could be greatly enhanced. Therefore, this method potentialy could have extensive and important applications.
9.Determination of valsartan in human plasma by HPLC

AIM To establish a high performance liquid chromatographic method (HPLC) to determine the concentration of valsartan in human plasma. METHODS Separation was achieved on the lichrospher C 18 column. The mobile phaseconsisted of pH 3 1 phosphate buffer acetonitrile (53∶47, V/V) was used at a flow rate of 1 0 ml?min -1 . The fluorimetric excitation and emission wavelengths were set at 265 nm and 378 nm, respectively. The plasma samples were acidified with HCl, extracted with ethyl acetate. Separate the organic phase, remove the solvent and then residue was dissolved in mobile phase. RESULTS The retention time of valsartan was 12 5 min. The calibration curves were linear in the range of 5 9~ 2 360 ?g?L -1 . The precision values (RSD) of intra day and inter day were determined to be 2 83%~7 07% and 1 57%~8 41% respectively. The absolute recovery rate were 80 30%?5 13%. The method was applied to determine the peak and valley concentrations in plasma of the hypertensive treated with 80mg valsartan per day. CONCLUSION The assay was sensitive and simple. It is suitable for the study of the pharmacokinetics of valsartan.
10.Efficacy and safety of liraglutide compared with insulin glargine in patients with type 2 diabetes mellitus inadequately controlled with metformin

Objective To observe the effect and safety of the human glucagon-like peptide-1 analogue,liraglutide,versus insulin glargine in patients with type 2 diabetes mellitus inadequately controlled with metformin alone.Method Ninty patients with type 2 diabetes mellitus(aged 18-79 years,HbA1C 7.5%-10.0%,body mass index<40 kg/m2) who had inadequate glycaemic control on metformin were allocated for the research with an open,randomized,parallel controlled clinical research method.The patients kept the original dose of metformin unchanged and were randomly assigned to the liraglutide group or the insulin glargine group according to a proportion of 1 ∶ 1.Liraglutide group started with a dose of 0.6 mg subcutaneous injection qd,changed to 1.2 mg subcutaneous injection qd after one week and kept unchanged until the end of the research.Insulin glargine group started with a dose of 0.1-0.2 U/kg according to the fingertips peripheral blood glucose level before breakfast on the continuous 3 d before every follow-up.At the baseline,after 4 weeks,12 weeks,20 weeks,and 26 weeks of treatment,HbA1C,blood glucose,lipids weight,blood pressure were arranged to measured.86 patients finally completed the study.Results Mean HbA1C and the success rate of HbA1C <7% were similar between liraglutide group and insulin glargine group [(7.06 ± 0.87) % vs (7.25 ± 1.20) %,47.73 % vs 45.23 %,P>0.05],while the percentage of subjects reaching the composite endpoint of HbA1C<7% with no hypoglycemia and no weight gain was significantly higher in liraglutide group than insulin group(P<0.05) ; Fasting plasma glucose decreased more markedly in insulin glargine group,2 h postprandial plasma glucose was decreased more markedly in liraglutide group(P<0.05 or P<0.01).Liraglutide significantly reduced mean body weight by (3.21 ± 1.18) kg,waist circumference by (3.82 ± 1.21) cm,and body mass index by (1.95 ± 0.61) kg/m2 (P<0.01 or P<0.05),while in the insulin glargine group there sere rise of respective figure of(2.86 ± 0.43) kg,(1.52 ± 0.56) cm,and (0.61 ± 0.25) kg/m2 (P<0.05),systolic blood pressure and serum triglyceride declined.There was no serious adverse affect in both groups,the incidence of mild hypoglycemia was significantly less in liraglutide group and has a statistically significant difference (4.55% vs 21.43%,P<0.05).Conclusions Liraglutide showed a good effect on reducing weight,systolic blood pressure,blood lipid and in addition to blood glucose control which is comparable to insulin glargine.What is more,liraglutide had good safety and tolerability,which can be regarded as a good choice for patients with type 2 diabetes mellitus inadequately controlled with metformin alone.
