1. Research progress on pharmacological effects of Polygonati Rhizoma

As a traditional Chinese herbal medicine, Polygonati Rhizoma widely distributed in most areas south of the Yangtze River. It has the function of nourishing liver and kidney, prolonging life and so on. Importantly, it is a Taoist Holy medicine since ancient times. Polygonati Rhizoma has high medicinal value and nutritional value because it contains polysaccharides, saponins, flavonoids, lignin, amino acids, quinones, vitamins, alkaloids and a variety of trace elements and so on. The domestic research institutions have carried out a deeper exploration, while its research is still at an early stage for foreign countries. At present, the experimental studies are mainly concentrated on the polysaccharides, ethanol, the extracts of saponins or the aqueous extracts of Polygonati Rhizoma. The experimental type is mainly based on the animal experiments and the clinical researches of Polygonati Rhizoma or its compound preparations. Various Polygonati Rhizoma preparations have been widely used in clinic, such as Polygonati Rhizoma Oral Liquid, Polygonati Rhizoma Tea, Cistanche and Polygonati Rhizoma Granules, Polygonati Rhizoma Zanyu Capsules, Polygonati Rhizoma essence oil patch and so on, which play different roles in individual products. In this paper, a comprehensive analysis was carried out on the basis of the latest experimental research on Polygonati Rhizoma, and its utility value was summed up from various angles, which provides a reference for the deep development and application of the Polygonati Rhizoma.
2.Joint application of EBL and PBL in clinical teaching of emergency medicine

Objective To observe and analyse the application effect and value of problem based learning (PBL) combined evidence-based learning (EBL) in emergency medicine clinical teaching.Methods A total of 53 clinical medicine students were selected.The combined teaching method was applied in emergency clinical teaching.After the end of the teaching,the teaching effectiveness survey and theory test of the combined teaching group was performed,and the theory test scores was compared with the traditional teaching group.Results The combined teaching method could stimulate learning enthusiasm,improve learning efficiency,the abilities of selfstudy,literature retrieval,and the ability to analyze and solve problems.77.4% of students thought this new teaching method had good application value in clinical teaching.Furthermore,the excellent rate of the theory test scores combined teaching group was better than traditional teaching group (41.5% vs 15.6%,x2 =7.868,P =0.007).Conclusions The PBL joint EBL pedagogy can overcome diadvantages of tradional teaching and provide higher interest,ability of self-study as well as higher teaching quality of the medical students.It will be worth to spread in medical clinical teaching.

Objective To compare the changes of serum levels of heme oxygenase‐1(HO‐1) ,C‐reactive protein(CRP) and Chitinase‐3‐1ike protein 1(YKL‐40) in patients with rectal cancer after laparoscopic‐assisted radical operation versus open radical surgery .Methods According the integration and elimination standard ,60 cases of patients with rectal cancer were selected and di‐vided into laparoscopic group (30 cases) and open group (30 cases) ,the concentrations of HO‐1 ,YKL‐40 and CRP in peripheral blood of patients were detected on the first and third postoperative day and preoperative day(P<0 .05) ,and were compared between the two groups .Results The serum levels of HO‐1 ,YKL‐40 and CRP in the laparoscopic group were significantly lower than those in the open group on the first and third postoperative day(P<0 .05) .The serum levels of HO‐1 ,YKL‐40 and CRP were significant‐ly increased after surgery ,and serum levels of HO‐1 and CRP on third postoperative day were significantly increased compared with that on the first postoperative day in the two groups after surgery ,while serum level of YKL‐40 on the third postoperative day was significantly lower than that on the first postoperative day in the two groups after surgery .Conclusion The stress reaction is com‐paratively mild in patients with rectal cancer after laparoscopic assisted radical operation ,and the postoperative period may be shor‐ter in patients treated with laparoscopic assisted radical operation than those treated with open radical surgery .
5.The effect of high-level of glucose on ghrelin expression in human umbilical vein endothelial cells

Objective To investigate the influence of high-level of glucose on the expression of ghrelin in human umbilical vein endothelial cells (HUVECs). Methods After 12 h glucose free culturing, the effects of different concentrations of glucose and different incubation times on expressions of ghrelin were observed in HUVECs. ( 1) The cells were treated with 0, 5, 10, 15, 20 mmol/L glucose for 2 h, then ghrelin mRNA expression levels were detected by RT-PCR, and the protein levels of ghrelin were detected by ELISA. (2) The cells were treated with 10 mmol/L glucose for 0, 0.5, 1, 2, 6, 12 hours, ghrelin mRNA and protein levels were detected respectively. Results (1) The expression levels of ghrelin mRNA and protein decreased along with increased glucose concentrations, which showed no obvious changes when the glucose was above 15 mmol/L. (2) The expression levels of ghrelin mRNA and protein decreased with the prolonged incubation time. But more than 6 h culturing time showed no further effect on reducing the expression level of ghrelin. Conclusion High levels of glucose can inhibit the expression level of ghrelin, which may be one of the mechanisms of atherosclerosis.

OBJECTIVE:To establish a HPLC method for the determination of stavudine and related substances METHODS:The Hypersil C18(4 6mm?150mm,5?m)was used as analysis column The mobile phase was methanol-water(13∶87,V/V) Detection wavelength was 264nm The flow rate was 1ml/min The column temperature was 20℃ RESULTS:The calibration curves was linear in the concentration range of 0 025~0 3mg/ml(r=0 9 999,n=6) The average recovery was 99 26%(RSD=0 76%) The contents of stavudine in 3 batches of semifinished materials and stavudine capsules(imported and domestic)were 99 48%,99 55%,99 32%(materials);97 21%,101 54%,98 92%(imported stavudine capsule);100 57%,97 86%,102 33%(domestic stavudine capsule)respectively CONCLUSION:The method is simple,accurate,sensitive and repeatable and it is suitable for the determination of stavudine and its preparations

Protein arginine methyltransferase (PRMT) can methylate a variety of proteins,including histones and non-histones,affecting a variety of cellular processes such as transcription,RNA splicing,DNA repair,and cell cycle control.The activity of PRMT is affected by a variety of regulatory mechanisms,and its abnormal expression plays an important role in the development of the diseases,especially in tumors such as breast cancer and leukemia.PRMT has a unique value in the diagnosis or treatment of tumors.With the further exploration of PRMT methylation mechanism,PRMT selective inhibitors have made some progress.PRMT-specific inhibitors are expected to be targeted drugs for the treatment of tumors.
8.Study on molecular mechanism of Euodiae Fructus on liver toxicity in MICE.

OBJECTIVETo study the molecular mechanism of extracts from Euodia rutaecarpa on hepatotoxicity in mice.
METHODTotally 30 KM mice were divided into 3 groups and orally administrated extracts from E. rutaecarpa for consecutively 15 days. The expressions of Erkl/2, CDK8, CK1e, Stat3 and Src were detected by Western blotting method.
RESULTThe extracts from E. rutaecarpa could up-regulated Erkl/2, CDK8 and CK1e expressions (P <0.01) and down-regulate Stat3 and Src (P <0.01).
CONCLUSIONThe molecular mechanism of E. rutaecarpa on hepatotoxicity may be correlated with Erkl/2, CDK8, CKle, Stat3 and Src signal molecules.

9.The comparison of methodologies ot assessing myocardial reperfusion in patients with acute myocardial infarction after primary angioplasty

Objective To assess the predictive value of coronary angiography TIMI myocardial perfusion grade (TMPG), electrocardiogram (ECG) single-lend ST segment resolution (STR), ECG Max-ST-segment deviation (MaxSTE) on judging myocardial reperfusion after primary angioplasty in the patients with acute myocardial infarction (AMI). Methods Primary percutaneous coronary intervention(PCI) was performed in 42 patients within 12 hours after AMI onset. Coronary angiography and ECG was done before and after angioplasty. TMPG, ECG single-leed STR and MaxSTE were used to assess myocardial reperfusion immediately after PCI. Myocardial perfusion scan was examined in all the patients using99mTc-MIBI SPECT on day 7±2. Results Compared with the level of myocardial perfusion demonstrated in myocardial scan of 99mTc-MIBI SPECT, the sensitivity, specificity and accuracy of TMPG, single-lead STR and MaxSTE was calculated. The sensitivity of TMPG, single-lead STR and MaxSTE was 93.75%, 87.50% and 81.25%, respectively, the specificity of them was 20.00%, 80.00% and 80.00%, respectively, and the accuracy was 76.19%, 85.71% and 80.95%, respectively. The findings in single-lead STR and MaxSTE matched well with the results of myocardial scan of 99mTc-MIBI SPECT. Conclusion In the patients suffering from AMI treated with PCI, ECG is an effective method to assess myocardial tissue reperfusion.
10.Expression and significance of aquaporin 1 in placenta, placental membranes and peritoneum of patients with hypertensive disorder complicating pregnancy

Objective To explore the role of aquaporin 1 (AQP1) in the initiation and development of hypertensive disorder complicating pregnancy ( HDCP), and to analyze the relationship between AQP1 expression and ascites formation of patients with eclampsia. Methods Sixty inpatients with HDCP were recruited in the study, including 20 patients with gestational hypertension, 20 with mild preeclampsia, and 20 with severe preeclampsia. And 20 healthy pregnant women were taken as control. Immunohistochemistry was used to analyze AQP1 expressions in placenta, embryolemma and peritoneum, and B-mode uhrasonography was used to detect the ascites level of the patients. Results ( 1 ) AQP1 expression was detected in placenta, embryolemma and peritoneum. AQP1 was mainly located in endotheliocytes of blood vessels and blood capillaries in placenta, endothelial cells of amniotic membrane in embryolemma, and endotheliocytes of blood capillary and small veins in peritoneum- (2)The ascites incidence of HDCP patients ( 63%, 38/60) was higher than that of controls( 10% ,2/20 ; P < 0. O1 ). ( 3 ) The positive expressive rate of AQP1 in placenta of patients with HDCP ( 85% ) was higher than that of controls ( 70%, P < 0. 01 ).Furthermore, the AQP1 positive expressive rate from severe preeclampsia (90%)was obviously higher than that from gestational hypertension patients ( 80%, P <0. 05 ). (4) The AQP1 positive expressive rate in embryolemma from HDCP patients( 87% ) was lower than that of controls ( 95%, P < 0. 05 ). The expressive rate from severe preeclampsia patients (80%) was obviously lower than that from gestational hypertension patients(95%, P <0. 05) and that of controls. (5) The AQP1 expressive rate in peritoneum from HDPC patients(82% )was higher than that of controls(70%, P <0. 01 ). The expressive rate of AQP1 from severe preeclampsia patients ( 90% ) was obviously higher than that from gestational hypertension patients (75%,P <0. 05) and that of controls. Conclusions The expression level of AQP1 of patients with preeclampsia increases in placenta and peritoneum and decreases in embryolemma, and holds correlation with the degree of HDCP. All these suggest that the changes in AQP1 expression may play an important role in the initiation and development of HDCP and may be one of the mechanisms for ascites formation.
