Chinese Journal of Virology 2014;30(4):483-488
Epidemics of hand, foot and mouth disease (HFMD) have mainly been caused by Coxsackievirus A16 (CVA16) and Enterovirus A 71 (EV-A71), which circulated alternatively or together in the affected area. CVA16 has caused numerous outbreaks and epidemics in multiple countries and geographical regions, and has become an important public health problem. Based on an analysis of the complete VP1 coding region, all CVA16 strains can be divided into genotypes A, B1, and B2. Furthermore, genotype B1 can be divided into subgenotypes B1a, B1b, and B1c. After 2000, no reports of genotype B2 virus strains have been reported. All of the CVA16 strains reported in mainland China have belonged to subgenotypes B1a and B1b. Most CVA16-associated infections cause only mild symptoms; however, some CVA16 infections can lead to severe complications and even death. Vaccination is considered to be the most effective method to control the transmission and infection rate of this virus. A number of research groups are studying various vaccine types, including inactivated vaccines, genetic engineering vaccines, and DNA vaccines, amongst others. In this review, an overview is provided of the research advances in molecular epidemiology and vaccines of CVA16.

Chinese Journal of Experimental Ophthalmology 2015;33(10):915-918
Background Researching the pathological characteristics and components of cells in internal limiting membrane of idiopathic macular hole (IMH) has an important clinical significance for the prevention of IMH.However, the study results are still disputable. Objective This study was to investigate the histopathological features of internal limiting membrane of IMH and the types of cells inside it, and explore the pathomechanism of IMH.Methods Seven specimens of internal limiting membrane were obtained during the vitrectomy with IMH patients in the Second Affiliated Hospital of Guangzhou Medical University from February 2012 to August 2013 under the informed consent of patients. The histopathological examination was performed for the structural observation and cellular identification of internal limiting membrane. The expression and location of glial fibrillary acidic protein (GFAP), CD45 and CD44 in internal limiting membrane were examined by using immunochemistry and immunofluorescence technology. Results All the seven specimens showed continuous undulating membrane with red staining. Two specimens appeared to be uniform in thickness and few cells were distributed in the specimens. The internal limiting membranes were uneven in thickness in the other specimens with retinal pigment epithelial cells, neuroglia cells, fibrocytes, macrophages and lymphocytes in them.Immunochemistry showed the positive expression of GFAP in the outer layer of the specimens.CD45 positive cells were detected in the internal limiting membranes, and CD44 was detected in the inner layer of the specimens. Conclusions Few cells exist in the internal limiting memranes of IMH.However, neuroglia cells and CD45 positive cells emerge in the internal limiting memranes of stage 3 or above IMH eyes, indicating the proliferation of cells and immuno-inflammation response exist during the IMH development. The up-regulation of CD44 expression promotes inflammatory response of internal limiting memranes.

Objective To observe the dynamics of F1 antibody and immunoglobulin M (IgM) in cats after vaccinated Yersinia pestis,to discuss the significance of detection of F1 antibody and IgM in surveillance of animal plague. Methods The 3 cats were vaccinated Yersinia pestis on their backs with muhipoint hypodermic injection and blood samples were collected via femoral vein on 3rd to 521st days after injection.F1 antibody was detected by sandwich Enzyme-linked immunosorbent assay (ELISA),and IgM of F1 antibody was determined with a capture antibody. Results After vaccinated Yersinia pestis for 3rd days,F1 antibody and IgM appeared slightly positive, with the titer of 1 ∶ 22.00 and 1 ∶ 20.33,respectively.The titer of F1 antibody increased to 1 ∶ 25.33 on the 4th day,and reached the peak of 1 ∶ 29.67 on the 97th day, kept 1 ∶ 25.33 on the 521st day. While the titer of IgM reached peak of 1 ∶ 212.00 on the 7th to 10th days, and decreased rapidly to below the positive standard on the 30th day. Conclusions Detection of F1 antibody in cats with plague by sandwich ELISA can trace plague prevalence in animals back to a long time, which may be applied for investigation of plague foci.A single serum sample can determine early animal plague with a capture antibody to detect IgM of F1 antibody in cat with the plague, and a single serum sample tested with the two methods at the same time for detection of F1 antibody and IgM can precisely verify the infection time of plague in animals for 3 to 7 days.

Background Recent studies indicated that human amnion mesenchymal stem cells (hAMSCs) can be induced to differentiate into multiple types of cells in vitro, but whether the hAMSCs can differentiate into ocular surface cells has not been reported yet. Objective This study was to investigate the feasibility of inducing differentiation of hAMSCs into ocular surface cells by co-culturing with human bulbar conjunctiva fibroblasts (hBCFs).Methods This study was approved by Ethic Committee of Affiliated Second Hospital of Guangzhou Medical University.HAMSCs were isolated from placenta under the informed consent of healthy delivery women.hAMSCs were cultured, passaged and identified by detecting the expressions of CD44,CD45,CD73,CD90 in the cells with flow cytometer, osteogenesis and adipogenic differentiation experiments.Human conjunctival tissue was obtained during the eye operation under the informed consent of patients and hBCFs were isolated and cultured with explant culture. The cells were divided into the hAMSCs culture group and the hAMSCs and hBCFs co-culture group and cultivated in Transwell chambers for 7 days. The expressions of cytokeratin 19 (CK19) and α-smooth muscle actin (α-SMA) in the cells were assayed by immnofluorescence technique. Results Cultured hAMSCs showed the slender shape and cell body enlarged with passage.CD44,CD73 and CD90 were expressed in the cells, and the expression of CD45 was absent. After 3-4 weeks of osteogenesis and adipogenic induce, the cells showed red staining for alizarin and oil red O.In the co-culture group of hAMSCs and hBCFs,hAMSCs presented the epithelioid cell-like in shape and showed the positive response for CK19 and weaker response for α-SMA.However,in the hAMSCs culture group, the cells showed the positive response for α-SMA and absent response for CK19.Conclusions The hAMSCs can differentiate into ocular surface cells after being induced by hBCFs.And the differentiation mechanism is possibly relevant to mesenchymal cells epithelium.

China Journal of Chinese Materia Medica 2015;40(3):560-563
To study the name of Wenyujin Rhizoma Concisum and Film Turmeric by literature research methods provide the basis for correct application of Cuba in modern clinical application. Wenyujin Rhizoma Concisum and Film Turmeric often called each other mutual generation and used as the same kind of medicine in the ancient prescriptions books. They were often recorded and stated as the same species of Curcumae Longae Rhizoma. Wenyujin Rhizoma Concisum and Curcumae Longae Rhizoma also often called each other mutual generation in the ancient prescriptions books and used as the same kind of medicine. Wenyujin Rhizoma Concisum was often recorded and stated under the Curcumae Longae Rhizoma articles in the ancient materia medica literatures. Ancient literatures on Wenyujin Rhizoma Concisum records were almost the same, the modern literatures on the records of the sources of Wenyujin Rhizoma Concisum were inconsistency. Inconsistency of Wenyujin Rhizoma Concisum source records in modern literatures was related to the changes of the modern records on source records about Curcumae Longae Rhizoma, Root-tuber of Aromatic Curcumae Longae Rhizoma and Zedoray. The author thinks that Wenyujin Rhizoma Concisum is the ancient Film Turmeric which referes to the same medicine as Curcumae Longae Rhizoma. The source of it just as the Curcumae Longae Rhizoma is not only one kind. Wenyujin Rhizoma Concisum and Curcumae Longae Rhizoma have been recorded as two medicines at the present, and the source of them simply referes to the original plant Curcuma wenyujin. When using ancient prescriptions, we need to understand the changes that Curcumae Longae Rhizoma and Wenyujin Rhizoma Concisum are the same in ancient but different today in order to choose medicine correctly in the clinical.
