2.Diffusion-weighted imaging combined with dynamic contrast-enhanced MRI in parotid gland tumors

Objective To explore the value of DWI combined with dynamic contrast-enhanced MR imaging (DCE MRI) in differential diagnosis of benign and malignant parotid gland tumors.Methods DWI and DCE-MRI was performed in 90 patients with parotid gland tumors (94 lesions),included 75 benign tumors and 19 malignant tumors.The ADCs were measured,and the type of time-intensity curve (TIC) was assessed.ROC curve was used to analyze the diagnostic efficacy of ADC values,TIC types and combination of the two methods in diagnosing benign and malignant tumors.Results The ADC values of malignant tumors ([1.02 ± 0.25] × 10a mm2/s) were significantly smaller than that of benign tumors ([1.38±0.44] × 10-3 mm2/s;t=5.170,P<0.001).The sensitivity was 0.62 and specificity was 0.92,when type A (persistent),B (washout),and D (flat) tumors regarded as benign and type C (plateau) tumors regarded as malignant.Sensitivity and specificity were significantly improved in diagnosis of malignant tumors when combined type C and ADC<1.36× 10-3mm2/s,which were 0.92 and 0.95,respectively.Conclusion Due to an overlap within benign and malignant parotid tumors,the diagnosis should not be addressed on the basis of ADC values alone.The TIC manifestations type A,B or D on DCE-MRI indicates benign parotid tumors,and type C indicates malignant diseases.When the TIC shows type C,combining ADC value can elevate the differential diagnostic efficacy of benign and malignant parotid tumors.
3.Effect of resveratrol on the oxidative stress and antioxidant levels of different skeletal muscle mitochondrial subpopulations and insulin sensitivity in rats fed with high-fat diet

ObjectiveTo observe the effect of resveratrol on the oxidative stress and antioxidant levels of different mitochondrial subpopulations in the skeletal muscle and insulin sensitivity of rats fed with high-fat diet.MethodsMale SD rats,aged 8 weeks,were divided into normal chow (NC) group,high-fat diet(HF) group,high-fat diet plus resveratrol ( HFR ) group.After intervention for 8 weeks,the impacts of resveratrol on oxidative stress levels and antioxidant enzymes activities in subsarcolemmal ( SS ) and intermyofibrillar ( IMF ) mitochondria from skeletal muscle as well as general and skeletal mascle insulin sensitivity were assessed.ResultsCompared with NC group, insulin sensitivity was significantly reduced while reactiveoxygenspecies (ROS)and malondialdehyde(MDA) levels in SS and IMF mitochondria increased in HF group ( all P<0.01 ).In addition,antioxidant enzyme activities were significantly decreased in SS mitochondrial and increased in IMF mitochondrial ( both P < 0.05 ).Compared with HF group,the insulin sensitivity in HFR group was significantly improved.Moreover,the activities of antioxidant enzymes in SS and IMF mitochondrial were increased,and the oxidative stress levels returned to normal ( P < 0.05 ).ConclusionResveratrol notably improves the oxidative stress of different skeletal muscle mitochondrial subpopulations and insulin resistance in rats fed with high-fat diet.
4.Analysis of influencing factors of platelet transfusion during bone marrow empty window period on patients with allogeneic peripheral blood stem cell transplantation

Objective To analyze the influence of different factors and their relating correlation results on platelet transfusion during the bone marrow empty window period on the patients who have undergone allogeneic peripheral blood stem cell transplantation (allo-HSCT) with retrospective analysis of case-control data.Methods Clinical data of 153 cases were collected by the clinical blood management and evaluation information system with discharge diagnosis of allo-HSCT in the hematology department of The First Affiliated Hospital of Nanchang University within a time frame from January 2014 to December 2016.A total of 90 cases were considered valid for retrospective analysis according to the case exclusion criteria.The average transfusion dose for patients with allo-HSCT during the bone marrow empty window period was defined as the threshold value which divided the 90 cases into the observation group of 38 cases receiving more than 6 Units of platelet transfusion and the control group of 52 cases with less than 6Units of platelet transfusion.The amount of platelets transfused during the bone marrow empty window period,clinical indexes include Hb,ANC,Plt,SF before pretreatment,platelet engraftment time and the number of mononuclear cells implanted were compared and analyzed by Logistic regression.Results (1) There was no significant difference between the two groups in gender,age,primary diagnosis,HLA matching,Hb before pretreatment and the number of mononuclear cells implanted (P>0.05).The ANC(×109/L) (1.24±0.57 vs 3.36±1.33) and Plt(×109/L) (43.55±68.29 vs 126.62±84.73) counts before pretreatment in the observation group were significantly lower than those in the control group(P<0.05).SF(μg/L) (2351.05 ± 1 587.96 vs 1 000.96± 362.97)before pretreatment and P LT recovery time (d) (16.84± 2.47 vs 12.73 ± 1.65)was significantly higher than that in the control group(P<0.05).Donor-recipient ABO blood group typing incompatibility (15 vs 10) was significantly higher than the control group (P<0.05);(2) Single factor Logistic regression analysis showed that ABO blood group matching,clinical indexes include ANC,Plt,SF before pretreatment,PLT recovery time were statistically significant,Only ANC before pretreatment and PLT recovery time had significant effect on the platelet transfusion during bone marrow empty window period in patients with allo-HSCT in multivariate Logistic regression analysis(P<0.05).Condusion The ANC before pretreatment and PLT recovery time are independent factors for platelet transfusion of the bone marrow empty window period in patients with allo-HSCT.The PLT recovery time is an independent risk factor,which indicates that the longer the duration of PLT implantation,the greater the amount of platelet transfusion will be needed.Besides,the ANC before pretreatment is the independent protective factor,which indicates that the greater the ANC,the smaller the amount of platelet transfusion is required.

6.Effect of iterative reconstruction techniques on the volume measurement accuracy of pulmonary ground-glass density nodule on low-dose CT

Objective To evaluate the potential of three commercial CT iterative reconstruction methods (ASIR,MBIR and iDose4) for the image quality and accuracy of volumetric measurement of ground-glass opacity (GGO) nodules on low-dose chest computed tomography (CT) using a phantom study.Methods A phantom containing GGO nodules was examined in volumetric mode on 2 multi-detector CT scanners at low dose.CT images were reconstructed with adaptive statistical iterative reconstruction (30% ASIR),model-based iterative reconstruction (MBIR),iDose4 L3,L6 reconstruction algorithm.Then two radiologists measured CT value of the region of interests (ROI) on reconstruction images,and calculated average noise and contrast to noise ratio (CNR) of reconstruction images.And they applied the lung nodules evaluation software to measure the volume of GGO nodules on the reconstruction images,and calculated the absolute percentage error (APE) of nodule volume measurement.The difference of noise and CNR at different reconstructions was compared with ANOVA test,and Kruskal-Wallis H ranking test was used to the difference of GGO measurements at different reconstructions.Results The average noises of CT image reconstructed with the standard FBP algorithm,30% ASIR algorithm,MBIR algorithm were 40.38 ± 4.49,30.06 ± 20.17,10.55 ± 1.56,and the CNRs were 5.00 ± 2.36,8.43 ± 3.89,20.27 ± 7.35,respectively.Compared to CT image reconstructed with the standard FBP algorithm and 30% ASIR algorithm,CT image noise reconstructed with MBIR algorithm decreased by 73.9% and 64.9%,while the CNR increased by 75.3% and 58.4% with effective dose of 0.36 mSv in HDCT,respectively(F =13.053,9.459,P <0.01).When effective dose was equal to 0.64 mSv in iCT,the image noise reconstructed with iDose L6 algorithms decreased by 36.5% compared with the standard FBP algorithm,and CNR increased by 78.4%.The reconstruction algorithm,NI and tube current were no significant difference in lung nodule volume measurement of absolute percentage error(P > 0.05).Conclusions Three kinds of iterative reconstruction algorithms are able to reduce the image noise in low-dose CT and improve the image quality.
7.Effect of Pyruvate-enriched ORS on intestinal mucosal blood flow, activity of Na+-K+-ATPase and intesti-nal absorption rate during enteral resuscitation of scalded rats

Objective Gastrointestinal rehydration is a simple and effective method in treatment of burn shock during war-time, fire disaster and other harsh conditions , and practice has proved the exact curative effect of HCO 3 salt sugar liquid .This article was to investigate the effect of pyruvate-enriched oral rehydration solution ( Pyr-ORS ) on intestinal mucosal blood flow ( IMBF ) , activity of Na +-K+-ATPase and intestinal absorption rate during en-teral resuscitation of a 35% TBSA third-degree scald in rats . Methods 90 male rats were randomly divided into 5 groups: scald without fluid resuscitation ( S group ); sham scald resuscitated with HCO3 salt sugar liquid ( SS HCO3 group ); sham scald resuscitated with Pyr-ORS ( SS Pyr-ORS group ); scald resuscitated with HCO 3 salt sugar liquid (S HCO3 group); scald resuscitated with Pyr-ORS (S Pyr-ORS group) (n=18).Each group was divided into 2 subgroups of 1.5 and 4.5 h after scald injury.Intestinal absorption rate of water and Na +, IMBF and activity of Na +-K+-ATPase were detected on each group . Results Compared with shame scald groups , the intestinal absorption rates of water and Na +decreased ob-viously in scald groups with fluid resuscitation (P<0.05);at 1 h after scald injury, the intestinal absorption rates of water and Na +in S Pyr-ORS groups were both higher than those in S HCO 3 groups(P<0.05).Compared with shame scald groups , IMBF and activity of Na+-K+-ATPase at 1.5 and 4.5 h after scald injury decreased obviously in scald groups with fluid resuscitation (P<0.05); at 1.5 and 4.5 h after scald injury, IMBF in S Pyr-ORS groups (95.250 ±5.096/112.765 ±7.215) were greater than those in S HCO3 group (80.764 ±7.852/94.671 ±8.469), which was of statistical significance (P<0.05). Conclusion Pyr-ORS is a simple and effec-tive method in treatment of burn shock during wartime , fire disaster and other harsh conditions .

9.Application of smell identification test in Parkinson's disease in China

Objective To evaluate the olfactory functions in patients with Parkinson's disease (PD) with a smell identification test developed for the Chinese population and to examine possible factors contributing to PD patients' olfactory performance.Methods Fifty-five patients with idiopathic and nondementia PD and 55 demographically matched healthy controls (HC) were recruited and their olfactory identification tested using the Chinese Smell Identification Test developed by the Institute of Psychology at Chinese Academy of Sciences.Results Olfactory identification score was significantly lower (t =-6.827,P<0.01) in the PD group (19.3 ±5.4) as compared with the HC group (26.5 ±5.7).Of all PD patients,63.6% scored over 1 SD below the age norm.In particular,of those with Hoehn-Yahr ratings between 2 to 3,77.8% scored over 1 SD below the age norm.Receiver operating characteristic (ROC) curve analysis indicated that a cut-off score of 22.5 on the Chinese Smell Identification Test distinguished between PD patients and HC with a sensitivity of 74.6% and specificity of 70.9%.Within the PD group,age,education,Hoehn-Yahr rating and Mini-Mental State Examination score were not found to correlate with olfactory performance (all P > 0.05).Conclusion PD patients in China show significant impairment of olfactory identification,which is effectively captured by the Chinese Smell Identification Test.
10. Expression of apolipoprotein D in autosomal dominant polycystic kidney disease and its influence on proliferation of cyst-lining epithelial cells

Objective To investigate the role of apolipoprotein D (ApoD) in the pathogenesis of autosomal dominant polycystic kidney disease (ADPKD). Methods The expression of ApoD protein in renal cystic tissues of ADPKD patients was examined by immunofluorescence and Western blotting analysis. Western blotting analysis and real-time PCR were used to detect the expression of ApoD protein and mRNA in the renal tissues of Han: SPRD rats. The cystic lining immortalized epithelial cells (WT9-12 cells) of ADPKD were stimulated with human recombinant ApoD protein and their proliferation was examined by MTT assay; flow cytometry was used to determine the cell apoptosis and cell cycle distribution. Results Immunohistochemisty and Western blotting analysis showed that ApoD expression was significantly lower in the renal tissues of ADPKD patients than that in healthy controls, and that in the Han: SPRD cy/+ rats was significantly lower than that in the Han: SPRD +/+ rats (P<0. 05, P<0. 01). ApoD mRNA expression in the kidneys of Han: SPRD cy/+ rats of 8, 12, 16, and 24 weeks old was significantly lower than that of Han: SPRD +/+ rats of same ages (P<0. 05). Recombinant ApoD protein at 100, 200, and 400 ng/ml inhibited the proliferation of WT9-12 cells by 8. 21%, 7. 59% and 8. 07% after 48 h stimulation (P<0. 05), and by 8. 62%, 6. 43% and 9. 42% after 72 h stimulation, respectively (P<0. 05). Treatment with 400 ng/ml human recombinant ApoD protein for 72 h increased Gi phase WT9-12 cells by 8. 26% and decreased S phase cells by 8. 09% (P<0. 05), but it had no significant effect on cell apoptosis. Conclusion ApoD may inhibit WT9-12 cell proliferation by regulating cell cycle, and decreased ApoD expression may participate in the pathogenesis of ADPKD.
