Objective To observe the dynamic levels of IL-2,IL-5 and IL-6 produced by Balb/ c mice infected with DEN_2 clinical strains and to study their relation.Methods The Balb/c mouse in- feetion model was established by multiple-site subcutaneous injection with various doses of DEN_2 clini- cal strain.Mouse plasma samples collected from different experiment groups at various time after in fection were tested for IL-2,IL-5 and IL-6 levels with sandwich ELISA.Results After primary in- feeted with DEN_2 B strain,the levels of IL-2,IL-5 and IL-6 of all experimental groups were not sig- nificantly higher than the normal control group while the levels of experimental groups increased sig- nificantly after re-infection.The level of IL-2 reached to peak[average value of(101 522.44?10 465.375)pg/ml]at the 4th day after re-infection(the 20th day after the primary infection),and then the level gradually reduced.The levels of IL-5 in the Balb/c mice of the group 1 and 2 reached to peak at the 1st day after re infection(the 16th day after the primary infection),and there was signifi- cant difference between these two groups and the control group(P＜0.05).The levels of IL-6 in all experimental groups reached to peak at the 1st and the 2nd day after re-infection.The peak value of the third group is the highest comparing with the normal control group(P＜0.05).Conclusion Th2 response was predominant in the second infection phase.

Objective To construct human cytomegalovirus pp65 prokaryotic expressing vector and induce specific T lymphocyte immune response with recombinant pp 65 protein of human cytomegalovirus(HCMV) to observe the effect of CTL on infected HEL cells. Methods The whole cDNA of HCMV pp65 was amplified by PCR and inserted into prokaryotic expressing vector pRSET by gene engineer technique. The product was purified by affinity chromphotography and identified with Western blot after that recombinant plasmid was expressed by the induction of IPTG. With MTT technique, we observed the stimulating and proliferating effect of recombinant HCMV pp65 protein on PBMC in vitro. Cytotoxicity of PBMC on HCMV-infected HEL cells was detected. Results The pp65 prokaryotic expressing vector was successfully constructed and could express in engineering bacteria DE3. High dose of pure recombinant protein was acquired and had been identified. The rhHCMV pp65 protein can activate PBMC and cause the proliferation of it in vitro. The proliferated PBMC have the specific cytotoxicity to HEL cells infected by HCMV. Conclusions The acquired recombinant HCMV pp65 protein could induce specific T cells immune response in vitro to kill the HCMV infected HEL cells. And it is very important for the immune therapy of the HCMV infections.

Objective To investigate the effect of isoflurane preconditioning on the nuclear factor kappa B (NF-kB)activity of leukocytes in patients undergoing cardiac valve replacement with cardiopulmonary bypass (CPB).Methods Twenty ASAⅡorⅢpatients of both sexes aged 20-60 yrs undergoing cardiac valve replacement under CPB were randomly divided into 2 groups(n=10 each):control group(C)and isoflurane preconditioning group(I).Anesthesia was induced with midazolam 0.08-0.12 mg?kg~(-1),fentanyl 5-10?g?kg~(-1) and vecuronium 0.1 mg?kg~(-1).The patients were mechanically ventilated(FiO_2=100%)after tracheal intubation.Anesthesia was maintained with intermittent i.v.boluses of fentanyl and midazolam in group C or with 2 MAC isoflurane and intermittent i.v.boluses of fentanyl and midazolam in group I before CPB.Systolic BP was kept between 90-120 mm Hg,diastolic BP between 50-80 mm Hg and HR between 60-90 bpm in both groups. Isoflurane was discontinued at the initiation of CPB.Arterial blood samples were taken after tracheal intubation and before inhalation of isoflurane(T_0)at 30 min(T_1),1 h(T_2)and 2 h(T_3)after aortic unclamping for determination of NF-kB activity of leukocytes using electrophoretic mobility shift assay(EMSA).The amount of fentanyl,midazolam,dopamine and sodium nitroprusside(SNP)consumed during operation and the rate of recovery of spontaneous heart beat in both groups were recorded.Results The NF-kB activity was significantly increased after aortic unclamping in C group but there was no significant difference in NF-kB activity before CPB (T_0)and after aortic unclamping(T_(1-3))in I group.The NF-kB activity was significant lower at T_(1-3) in group I than in group C.The total amount of fentanyl consumed was 40-60?g?kg~(-1) in C group and 20-30?g?kg~(-1) in group I. Significantly less amount of dopamine was used in group I than in group C.There was no significant difference in SNP consumption between the 2 groups.The rate of recovery of spontaneous heart beat was significantly higher in group I than in group C(P＜0.01).The amount of dopamine consumed was positively correlated with the highest level of NF-kB activity in both group[r=0.962 in group C;r=0.908 in group I(P＜0.01)].Conclusion Isoflurane preconditioning can attenuate the NF-kB activity of leulocytes in patients undergoing cardiac valve replacement under CPB.

In order to improve the metabolism yield of cordycepin and adenosine,we studied nitrogen sources ,the levels of nitrogen sources and carbon sources,dynamic development in liquid culture ,as well as the total yield of cordycepin and adenosine in culture system through determining the contents of cordycepin and adenosine by HPLC .The results are as follows: not only animal protein but also some plant protein is very good nitrogen sources; in culture solution the suitable levels of nitrogen sources and carbon sources is respectively 3% and 4%; the content of adenosine in culture solution is very low, while it is quite high in mycelia; it is much higher that the total yield of cordycepin in culture solution than in mycelia; when the culture system is oscillated and cultured after 7~9 days, the total yield of cordycepin and adenosine are both high.
