Objective To study the expression of Vascular Endothelial Growth Factor (VEGF),Malondialdehyde (MDA),Hypoxia Inducible Factor-1 α (HIF-1 α) in the Non-Steroidal Anti-Inflammatory Drug (NSAID)-associated gastric mucosa injury tissue, and to understand the mechanism of NSAID induced gastric mucosa injury. Methods We collected 114 biopsy specimen of gastric mucosa under endoscope from patients admitted to our hospital, including 70 cases with NSAID-associated chronic erosive gastritis or gastric ulcer (NSAID group) and 44 cases with chronic superficial gastritis who never take NSAID as controls (the control group).Immunohistochemistry technique was used to detect the positive expressions of VEGF,MDA,HIF-1α in patients. Results The positive rate of VEGF expression in the NSADI group was significantly lower than that in the normal control group (25.7% (18/70) vs.54.5% (24/44),x2 =9.70,P < 0.05).The positive rates of MDA and HIF-1α in the NSADI group were significantly higher than these in the normal control group (MDA:62.9% (44/70) vs.27.3% (12/44),x2 =13.70,P <0.05; HIF-1α:45.7% (32/70) vs.13.6% (6/44),x2 =12.50,P < 0.05).Conclusion NSAID drugs may induce gastric mucosa injury by decreasing the expression of VEGF and increase the levels of MDA and HIF1-α in gastric mucosa tissue.

Background contamination is a major problem in the analysis of organophosphate esters (OPEs). In this study, the possible sources of OPEs pollution were screened and several different ways were applied to minimize the blank contamination. Under the strict quality control measures, the cleanup efficiency of different solid phase extraction (SPE) was investigated for OPEs in different environmental matrices. A method was developed for the detection of 7 OPEs in dust, soil and sediment samples by gas chromatograph coupled with mass spectrometry ( GC / MS). Target compounds were extracted by hexane:dichloromethane (1 : 1, V/ V) followed by aminopropyl silica gel SPE column cleanup for dust, and target compounds in soil and sediment were Soxhlet extracted and cleanuped by two-step SPE. The results showed that the aminopropyl silica gel SPE column displayed the best purification performance among the three employed columns. Instrumental detection limits among the 7 OPEs ranged from 2. 5 to 25. 8 μg / L, and the method limits of quantification (MLOQs) in dust and soil sample ranged from 1. 4 to 15. 7 ng / g and 0. 3 to 2. 9 ng / g, respectively. The average recoveries of 7 OPEs in different matrices ( dust and soil) at two spiked concentration levels ranged from 67. 9% to 117. 4% . The proposed method was successfully applied to detect OPEs in different environmental matrices collected in Shanghai.
7.Serum levels of IL-12, TGFbeta1 and IgE in children with asthma.
Chinese Journal of Contemporary Pediatrics 2008;10(2):146-148
OBJECTIVEThis study examined the changes of serum levels of interleukin 12 ( IL-12), transforming growth factor beta 1 (TGFbeta 1) and immunoglobulin E ( IgE) in children with asthma as well as the correlation of IL-12 and TGFbeta 1 with IgE in order to investigate their roles in asthma.
METHODSSerum levels of IL-12 , TGFbeta 1 and IgE were detected using ELISA in 85 asthmatic children at the acute and the remission stages. Thirty healthy children served as control group.
RESULTSCompared with the control group, serum IL-12 and TGFbeta 1 levels were significantly lower and serum IgE levels were significantly higher in the asthmatic group through the acute to the remission stages. Serum IL-12 and TGFbeta 1 levels (40.42+/-15.26 ng/L and 65.41+/-22.38 pg/mL) significantly increased in the asthmatic group at the remission stage compared with those at the acute stage (28.42+/-10.73 ng/L and 40.25+/-11.73 pg/mL) (P<0.01), but remained lower levels than those in the control group (67.42+/-20.58 ng/L and 178.54+/-90.56 pg/mL) (P<0.01). The asthmatic patients at the remission stage showed significantly decreased serum IgE levels (145.67 +/-51.25 IU/mL) compared with those at the acute stage (280.35 +/-80.54 IU/mL) (P<0.01), but the IgE level in the remission stage was obviously higher than in the control group (53.61+/-13.32 IU/mL) (P<0.01). Serum IL-12 and TGFbeta 1 levels were negatively correlated with serum IgE level in asthmatic children.
CONCLUSIONSThere might be an imbalance in serum IL-12, TGFbeta 1 and IgE levels in asthmatic children. IL-12, TGFbeta 1 and IgE may play an important role in the pathogenesis of asthma. They may be useful in the diagnosis and severity evaluation of asthma.
