1.Extracelluar matrix stimulates neurite outgrowth of dorsal root ganglion neurons differently depending on whether extracted from degenerated or normal intervertebral disk

BACKGROUND:Compared with the normal intervertebral disk, the density of nerve fibers and number of nerve endings and neuropeptides appear to be more in the degenerated intervertebral disk. However, this phenomenon does not occur in the normal y aged disk.
OBJECTIVE:To evaluate the axonal growth and induction of a painful neuropeptide and substance P using rat dorsal root ganglion neurons and degenerated human disc cells in vitro.
METHODS:The human intervertebral discs were harvested from patients with discogenic low back pain and normal people. And extracelluar matrix extracted from human degenerative intervertebral discs was cultured with rat dorsal root ganglion neurons. The promotion of axonal growth and induction of substance P of dorsal root ganglion neurons in extracted medium were evaluated through morphology observation and enzyme-linked immunosorbent assay, respectively.
RESULTS AND CONCLUSION:Compared with the normal group, the content of nerve growth factor in the degenerative group was significantly higher and the average length of neuritis was significantly longer in the experimental group (P<0.05). After intervention with anti-nerve growth factorβ, the average length of neuritis became remarkably shorter. The percentage of substance P-immunoreactive cells was significantly higher in the degenerative group compared with the normal group (P<0.001). Nerve growth factors that highly express in the extracellular matrix from the degenerative intervertebral dick can promote neurite outgrowth of dorsal root ganglion neurons and induce release of neuropeptides related to pain transmission.

4.Effectiveness of the Spot Vision Screener on vision screening of children without cycloplegia

AlM: To evaluate the effectiveness of Spot Vision Screener on vision screening of children without cycloplegia.METHODS:A total of 87 children (174 eyes) aged from 2~9 years old were examined with Spot Vision Screener and optometrist before cycloplegia.RESULTS: Statistical analysis demonstrated that the cylinder diopter and axis, the equivalent spherical diopter in both eyes, and the spherical diaopter in left eye had no significant change ( P>0. 05 ). However, the spherical diaopter in right eye had statistical significance. CONCLUSlON: Spot Vision Screener is a suitable instrument in vision screening of children without cycloplegia.
7.Clinical assessment and significance of peritoneal lavage cytology on prognosis of gastric carcinoma

Objective To evaluate the clinical value between peritoneal lavage cytology (PLC) and prognosis of gastric cancer. Objective To explore the effect of sex difference on learning and memory function and the brain tissue damage of neonatal SD rats with hypoxic-ischemic brain damage.Methods Sixty,7-day-old neonatal Sprague-Dawley rats,were divided into randomly:male control group (M group,n=15),female control group (F group,n=15),male hypoxic-ischemic brain damage group (MHIBD group,n=15) and female hypoxic-ischemic brain damage group (FHIBD group,n=15).A modified newborn rat model that had a combined hypoxic-ischemic brain damage as described by Rice-Vannucci was used.The Morris water maze was used to evaluate the ability of learning and memory.The brain MRI and transmission electron microscope(TEM) was used to evaluate the scope of brain tissue damage and the change of the synaptic ultrastructure.Results There were no differences in swimming distance,escape latency,synaptic cleft and damage brain volume between M group and F group(P>0.05).Compare with M group and F group,MHIBD group and FHIBD group showed significant brain injury,longer escape latency ((39.38±11.40) s vs (14.86±4.42) s,(30.14±7.18)s vs (18.41±5.03) s),longer swimming distance ((15.31± 1.77) cm vs (3.68±1.50) cm,(13.18±1.79) cm vs (4.61±1.61) cm),and TEM showed the synaptic cleft was widened ((23.18± 1.36) nm vs (19.24± 1.51) nm,(21.40± 1.71) nm vs (19.87±0.94) nm),P<0.05).MHIBD group was more seriously compromised than the FHIBD group(P<0.05).The brain MRI showed the damage brain volume of MHIBD group were significantly larger than FHIBD group(P<0.05).Conclusion After bypoxic ischemic brain damage,the tolerance of brain damage and / or post-injury recovery capabilities of female rats are stronger than males.
9.Stability of the salt iodization agent potassium iodate: a differential thermal analysis

Objective To investigate the thermal stability of solid potassium iodate and potassium iodate as additive in the sodium chloride,vitamin E,vitamin C and yellow prussiate.Methods HCR-2 type Differential Thermal Analyzer was used to carried out the differential thermal analysis of the potassium iodate and the potassium iodate in the sodium chloride,vitamin E,vitamin C and the yellow prussiate,and differential thermal curves were obtained and analyzed.Results The decomposition temperature of solid potassium iodate was 525℃ ; when mixed with sodium chloride,potassium iodate was stable below 300 ℃ ; vitamin C was unstable at 170-200 ℃ and underwent chemical changes; iodate and vitamin C underwent oxidation-reduction reaction at 145 to 160 ℃;potassium iodate with vitamin E at 300 ℃ was stable; yellow prussiate at 300 ℃ was stable; iodized salt was stable at cooking temperature below 300 ℃.Conclusions The potassium iodate has good stability below 525 ℃,however,potassium iodate iodized salt in the cooking process is easy to react with vitamin C in vegetables causing iodine losses,so iodized salt should be added just before the dish is done.
10.Effect of Foreign Plasmid DNA on Material and Energy Metabolism of Spleen in Mice

To study the influence of foreign plasmid DNA on spleen metabolism in immune-stimulated mice via the gastrointestinal tract. Mice were oral administered by pipette with 200?g of plasmid pcDNA3 and spleen were isolated at 4h and 18h after oral administration. Total RNA were extracted from spleen and spleen gene expression profile of Balb/c mice was analyzed by using Affymetrix oligonucleotide genechip after oral plasmid pcDNA3 administration. Functional cluster analysis was conducted by Genmapp and MAPPFinder software. By functional cluster analyzing the genes which were up-regulated more than twofolds, Genmapp results showed that plenty of metabolic pathway were induced in spleen after oral administration of plasmid pcDNA3. These metabolic process included purine metabolism, pyrimidine metabolism, protein synthesis, cholesterol synthesis, fatty acid synthesis, Glycolysis, TCA cycle and mitochondria oxidative phosphorylation pathway. The similar results also took place at 18h after oral administration. The result indicated that foreign plasmid DNA can modulate metabolism process in spleen of mice via the gastrointestinal tract, and may help understand the mechanism of action of foreign plasmid DNA uptaked via the gastrointestinal tract.
