1.Study of correlation of diabetic nephropathy with serum endothelin(ET-1) and C-reactive protein(CRP)

Objective To investigate the relationship of diabetic nephropathy with serum endothelin-1(ET-1) and C-reactive protein(CRP). Methods Levels of blood pressure, glucose, lipid, creatinine,ET-1,CRP and urinary albumin excretion(UAER) were measured and calculated in 86 patients with type 2 diabetes mellitus(T2DM)and 30 healthy subjects. Results Serum ET-1 and CRP were significantly higher in T2DM than in control(P<0.05,P<0.01).Serum ET-1 and CRP were increased with increment of serum creatinine and UAER(P<0.05,P<0.01). Conclusions Serum ET-1 and CRP may be involved in the development and progression of diabetic nephropathy.
2. Insulin-like growth factor- II induces mTOR pathway change in Rh1 sarcoma cells

Objective To observe the effect of insuiin-iike growth factor-II (IGF-2) on the growth and the mTOR pathway of Rh1 sarcoma cells. Methods Rh1 cells were cultured routinely, and were treated with IGF-2 at a final concentration of 10 ng/ml after starving with pure RPMI 1640 medium. The growth of cells was analyzed by flow cytometry 72 h after IGF-2 treatment. The phosphorylation of S6 and Akt (s473) proteins were examined by Western blotting analysis at 5, 10, 20, 30, and 60 min after IGF-2 treatment. Results IGF-2 treatment promoted the survival and inhibited the apoptosis of Rh1 cells compared with the control group. IGF-2 also increased the phosphorylation of S6 in a time-dependent manner. However, the phosphorylation of Akt(s473) was relatively stable in Rh1 cells. Conclusion IGF-2 can gradually increase the function of S6 in the mTOR pathway, and the function of Akt (s473) is kept relatively stable.
3.Analysis of Immunogenicity of ENA in Activated Lymphocytes

Objective To study the immunogenicity of extractable nuclear antigens(ENA)in activateed lymphocytes.Methods The ENA of the normal and activated lymphocytes was extracted according to Sharp's method,then syngeneic BALB/C mice were immunized.The dynamic fluctuation of serum IgG anti-dsDNA antibody level in mice was analyzed by ELISA,so did the ENA polypeptide spectrum.The immunofluorescent staining pattern of ANA and renal immunopathologic changes of the mice were investigated.Results ANA could be detected in the sera of the immunized mice by the ENA extracted from the activated lymphocytes,including anti-dsDNA and anti-ENA.The immunofluorescent staining patterns for ANA manifested as homogeneous pattern,peripheral pattern,speckled pattern and nucleolar pattern.Moreover,marked immune complex deposits in glomerulus could be observed in ANA positive mice.The results in those mice immunized by the normal-lymphocyte-ENA were negative.Conclusion The ENA extracted from activated lymphocytes is immunogenic,can drive the production of ANA and cause SLE-like syndrome.

5.Effect of Prolactin and Growth Hormone on the Secretion of Th1/Th2 Type Cytokines in the Patients with Systemic Lupus Erythematosus

Objective To study the effect of growth hormone (GH) and PRL (prolactin) on the secretion of Th1/Th2 type cytokines in the patients with systemic lupus erythematosus (SLE). Methods The secretion of cytokines in PBMCs stimulated by GH and PRL was detected by ELISA in vitro. Results The results showed that the secretion levels of IL-6 and IL-10 from PBMCs of the SLE patients in active stage were higher than those of the normal controls, and the secretion level of IFN-? was lower than that of SLE patients in resting stage and the normal controls, but it is of no statistical significance. The secretion levels of IL-6 and IL-10 were significantly increased in PBMC stimulated by GH and PRL, but GH and PRL had no effect on the secretion of IFN-?. There was no difference on the secretion level of IL-6, IL-10 and IFN-? when the PBMC was stimulated by GH and PRL. Conclusion GH and PRL might play an important role in the pathogenesis of SLE.

8.Clinical significance of NMP 22 in early diagnosis of bladder cancer

10 U/ml was determined as positive value.Urinary NMP 22 protein was elevated in 22 cases.Bladder cancer was diagnosed in 11 cases.The sensitivity and specificity of the NMP 22 test were 100%(11/11) and 81%(46/57),respectively.Cystoscopy alone identified 35% of the cancers (4/11).Among 22 cases with elevated NMP 22,1 case was dignosized as bladder cancer during 1 year visit. Conclusions Urine NMP 22 is a new useful marker in early diagnosis of bladder cancer.
9.Effect of physical stimulus on periosteal chondroplasia

Objective To study the impact of physical stimulus on periosteal chondroplasia with an in- tention of reconstructing cartilage tissue that has a biomechanical function and can be created either entirely in vivo or in vitro and subsequently implanted into the patient.Methods A series of biomechanical test methods were designed in order to investigate the mechanical properties of engineered cartilage under a variety of mechanical stress stimulations.The periosteal explants were harvested from proximal medial tibias of New Zealand rabbits and secured onto poly-?-caprolactone(PCL)scaffolds.The composites(periosteum/PCL)were suspended in the spinner flask bioreactor and the spinner flask was located on the stirring machine.The stem cells were stimulated by fluid shear stress under magnetic bar stirring.Results In gross observation,the thickness and length of neo-cartilage with shear force stimulation were significantly greater than those without shear force.The direction of elongation was parallel with fluid flow direction,regardless of composite orientation.Secondary,two different zones based on various cell morphology and distribution were clearly observed.The thickness of the superficial zone was greater in samples exposed to high shear force compared with low shear force.The superficial zone protein and lubricant were found in the engineered cartilage under shear stress stimulation.Conclusion The results reveal that shear stress can not only influence proliferation and differentiation of stem cells,but also change chondrocyte morphology and increase extracellular matrix production.
10.Practice of curriculum integration teaching reform for Traditional Chinese Orthopedics

To improve the quality of teaching, teaching reform and exploration on integration of Ttradition Chinese Orthopedics curriculum has been carried out by orthopedics teaching and research section in our school. Putting Traditional Chinese Orthopedics as the framework of curriculum knowledge, the im-portant and difficult points of Human Anatomy and Clinical Medical Science were combined, which builds the overall teaching mode of normal-disease-cases and get some results. The evaluation of teaching effect shows thatthe integration of Traditional Chinese Orthopedicscourse will enable students to lay a solid the-oretical foundation and improve their ability.
