Polymerase chain reaction (PCR) is one of the common techniques in molecular biology, which can amplify nucleic acids through the cycle of denaturation, annealing and extension. Based on the principle of common PCR, rapid PCR is to realize the amplification of nucleic acids in less time without affecting the specificity, sensitivity and fidelity of the reaction. A lot of research work in this field has been going on in recent years. This article will make a review of the development of rapid PCR with emphases on the improvement of DNA polymerase, the choice of additives and the improvement of thermocyclers.

Objective To study metabolism of polyunsantured fatty acid (PUFA) and to investigate the relationship between different PUFA compositions and △6 desaturase (D6D) activity in colorectal carcinoma (CRC).Methods Fresh frozen malignant CRC tissues were obtained from 61 patients and blood samples were detected in 38 patients.The PUFA composition in these samples was determined by gas-liquid chromatography on a capillary column.The fatty acid product-to-precursor ratios in blood and tissue as estimates of desaturase activity (D6D activity index),and its relationship with the ratios between different PUFA compositions were investigated.Results Metabolism of ω-6 PUFA dominated in PUFA metabolism path.The ratio of ω-6/ω-3 PUFA was 4.89 in blood samples and 10.15 in tissues.The ratio of LA/ALA was 101.92 in blood samples and 86.16 in tissues.The ratio of ω-6/ω-3 LC-PUFA was 2.86 in blood samples and 4.51 in tissues.The D6D activity index in ω-6 PUFA metabolic processes both in tissues (t =1 1.609,P =0.00) and blood samples (t =-9.151,P =0.00) were higher than that in ω-3 PUFA.Conclusion In colorectal carcinoma,the ω-6 PUFA metabolic not only plays an important role in the metabolism of PUFA,but also is more active than the ω-3 PUFA metabolic,and in some way links to the development of tumorigenesis in CRC.

Objective To evaluate the security and biocompatibility of a new kind of absorbable high-strength PDLLA-CPP composite, which is used for internal fracture fixation. Methods According to the national standard of the procedure of appraise in food security and toxicology, micronucleus test of bone marrow cell, spermatic aberration test, chromosomal aberration test, and teratogenic test were measured with IP maximal tolerance dose of maceration extract from PDLLA-CPP composites. Meanwhile, chondrocyte culture with PDLLA-CPP composite was used to evaluate the effect of PDLLA-CPP composite on cartilage cell proliferation. Results Micronucleus test of bone marrow cell, spermatic aberration test, chromosomal aberration test, and teratogenic test were not affected by IP maximal tolerance dose of PDLLA-CPP maceration extract. PDLLA-CPP composite also had no cytotoxicity on cartilage cell proliferation. Conclusion PDLLA-CPP is a new and safe medical material with good biocompatibility and absorbability.

Objective To investigate the diagnostic value and the efficacy of magnetic resonance tomographic angiography(MRTA) in identifying the relationship between nerves and blood vessels of the patients with idiopathic trigeminal neuralgia.Methods Clinical and MR data of 280 ITN patients were analyzed retrospectively,and then were compared with operation results by using blind con-trol trials.Results Of the 280 ITN patients,there were a total of 267 patients with neurovascular contact,dubious contact or com-pression in affected sides (positive rate:95.3%),and 66 patients in normal sides (false positive rate:23.6%).And of 120 surgical patients,1 18 were found to have neurovascular contact or compression during the operation.There was significant statistical differ-ence between the neurovascular contact or compression in affected sides and that in normal sides (P <0.05).Using operation results as the gold standard,the specificity,sensitivity and the diagnostic accordance rate of MRTA in diagnosing neurovascular contact or compression were 100%,96.6% and 96.7% respectively.Conclusion MRTA can reliably display the relationship between nerves and blood vessels with higher diagnostic sensitivity and accuracy,thus providing the objective basis for the etiological diagnosis of and surgical treatment for ITN.

Objective:To investigate the range of normal value of total nasal cavity volume(NV)and total na-sopharyngeal cavity volume (NPV)of healthy adults. Method: Six hundred and fifty-nine healthy adults andeighty-two adults of chronic rhinitis were measured with acoustic rhinometry. Result:The range of NV was29. 922～37. 481 cm3 and NPV was 29. 369～44. 159 cm3. Comparing healthy adults with adults of chronic rhini-tis,there was a significant difference in NV. Conclusion:Acoustic rhinometry suited for objective assessment ofthe nasal airways in adults. It was demonstrated that these data could provide available information for the studyof nasal physiology and pathophysiology,as well as for the diagnosis and judgement of therapeutic effectiveness ofnasal diseases.

BACKGROUND: With the development of internal fixation materials, simple operation with internal fixation has become a tendency. Ni-Ti shape memory alloy embracing fixator has such advantages as slight injury, easy operation, security, reliable fixation, and better histocompatibility. The present study was to explore curative effect and postoperative results of Ti-Ni shape memory alloy embracing fixator in patients with multiple fractured ribs and flail chest. METHODS: The curative effect and long-term follow-up results were observed after internal fixation with a shape memory alloy embracing fixator in patients with multiple fractured ribs and flail chest from January 2006 to December 2009. RESULTS: All patients were cured with an average hospital stay of 10.31±3.14 days. Post-operative pain was less severe than preoperative pain (P=0.02).The rate of postoperative complications such as atelectasis, pulmonary infection, etc was 17.65％. There were fewer long-term complications and less influence on daily work and life. CONCLUSIONS: It is practical to perform an operation for fracture of multiple ribs using a Ti-Ni shape memory alloy embracing fixator. The fixator, which is less traumatic, simple, safe, and reliable, has a good-histocompatibility and fewer postoperative complications.
