Objective To explore the in vitro isolation of ?2m-/Thy-1+ bone marrow-derived liver stem cells(BDLSCs) which bear double features of stem and liver cells from bone marrow stem cells(BMSCs)as so to provide suitable donor cells for the treatment of liver diseases by cellular transplant. Methods ?2m-/Thy-1+ BDLSCs were isolated by magnetic bead cell sorting(MACS) method from cholestatic rats in vitro,and cell purity was detected using flow cytometry.Liver associated phenotype markers were characterized by RT-PCR and immunofluorescence staining. Results BDLSCs isolated by MACS were purified and viable,and possessed hepatocyte-like features at gene and protein levels. Conclusion ?2m-/Thy-1+ BDLSCs are special subsets of BMSCs which may have promising potentials in the stem cell-based treatment of liver diseases.
3.Summary and analysis of the intercomparison results of national individual monitoring for external exposure during 2015-2019

Objective:To enhance the capabilities of individual monitoring technology services for external exposure.Methods:The intercomparison results of the national individual monitoring for external exposure during 2015-2019 were presented, together with a summary and analysis provided of the main existing problems.Results:By 2019, 382 of individual monitoring technology service uints, from 30 provinces, autonomous regions or municipalities, participated in the individual monitoring intercomparisons results, involving disease control and prevention centers, occupational prevention and control institutions, research institutes, universities, nuclear industry, medical institutions, and companies. Except for slightly low in 2017, the pass rate in the other four years was above 90%. The excellence rate increased with the years.Conclusions:The capabilities of individual monitoring service units can meet the concerned requirements and provide standardized monitoring report for the period of 2015 to 2019, but with some being unqualified. These service units should carefully analyze and identify the reasons for the failure, standardize the quality control of laboratory, and improve the level of measurement and the ability of data analysis.

6.Effects of AcSDKP on proliferation and secretion of extracellular matrix in rat active hepatic stellate cells

Objective To study the effects of N-acetyl-seryl-aspartyl-lysyl-proline(AcSDKP) on proliferation and synthesis and secretion of extracellular matrix(ECM) in rat active hepatic stellate cells(HSCs). Methods Primary cultures of HSCs when actived were subjected to AcSDKP(0.01, 0.1, 1, 10 and 100 nmol/L, respectively) treatment, and control group was established. The expression of collagen Ⅰ(ColⅠ) and collagen Ⅲ(Col Ⅲ) mRNA were analyzed with RT-PCR. The proliferation of HSCs was detected by MTT. Hyaluronic acid (HA) and laminin(LN) in the supernatant were determined by enzyme-linked immunosorbent assay. Results Compared with control group, the proliferation of active HSCs was significantly inhibited by 1 and 10 nmol/L AcSDKP. 1, 10 nmol/L AcSDKP and 0.1 to 100 nmol/L AcSDKP significantly inhibited the expression of HSCs ColⅠ mRNA and Col Ⅲ mRNA, respectively. Expression of HA and LN in the supernatant were significantly inhibited by 0.1, 1, 10 nmol/L and 0.1, 1 nmol/L AcSDKP, respectively. ConclusionAcSDKP can inhibit synthesis and secretion of ECM in active HSCs in a dose-dependent manner, with a maximum inhibition effect at 1 nmol/L AcSDKP. The mechanism may involve the inhibition of the proliferation of HSCs,which leads to the decrease of HSCs that synthesize and secrete ECM.
8.Expression and identification of the functional domains of dengue virus type 1 envelope protein in 293T cells

Objective To construct a recombinant expression vector for expression of the function-al domains of dengue virus serotype 1 ( DENV1 ) envelope ( E ) protein in native soluble form. Methods The genes encoding the functional domains of DENV1-E protein (1-394 aa) were amplified with PCR and then cloned into the Psectag2B-Fc eukaryotic expression vector.The 293T cells were transfected with the recombinant vector by cationic lipid-based delivery.The cell clones expressing the fusion DENV1-E-Fc protein were screened out with 2 mg/ml of Zeocin.Immunofluorescence assay ( IFA) was performed to analyze the antigenicity and integrity of the fusion protein.The fusion proteins were purified from cell lysate with Protein-G and further identified by Western blot assay.Results The soluble form of fusion protein with a molecular weight of about 90×103 was obtained at a yield of about 25 μg per 1×107 cells.The results of IFA indicated that the fusion protein kept its integrity with right conformational epitopes.The fusion protein was successfully expressed with the advantage of good specificity as indicated by IFA and Western blot assay. Conclusion The recombinant fusion protein in soluble form was successfully expressed in eukaryotic ex-pression system, which paved the way for further investigation on the function of DENV1 E protein and its protective epitopes.
9.Effects of mycorrhizal colonization and medicine quality of Paris polyphylla var. yunnanensis inoculated by different foreign AM fungi species.

After 28 foreign species of AM fungi were inoculated in sterilized soil, the effects of the AM mycorrhizal colonization and the medicine quality of Paris polyphylla var. yunnanensis were observed by combination of inoculation test in pot at room temperature and instrumental analysis. The results showed that, compared with control group (CK), the inoculation of foreign AM fungi in the soil influenced the spore density, mycorrhizal infection rate, and colonization intensity of AM fungi in root system of P. polyphylla var. yunnanensis. The inoculation of foreign AM fungi enhanced the mycorrhiza viability of P. polyphylla var. yunnanensis by increasing the activity of succinic dehydrogenase (SDH) and alkaline phosphatase (ALP) in intraradical hyphae. The content of single steroid saponin in rhizome of P. polyphylla var. yunnanensis showed variation after P. polyphylla var. yunnanensis was inoculated by different foreign species of AM fungi, which was beneficial for increasing the medicine quality; however, the kinds of steroid saponin showed no difference. In a degree, there was a selectivity of symbiosis between P. polyphylla var. yunnanensis and foreign AM fungi. And we found that the Claroideoglomus claroideum and Racocetra coralloidea were best foreign AM fungi species for cultivating P. polyphylla var. yunnanensis under field condition.

10.Meta analysis of etiology and clinical features of recurrent acute pancreatitis in China in recent decade

Objective To summarize the etiology and clinical features of recurrent acute pancreatitis (RAP) in the recent l0 years in China.Methods Pubmed,Medline,EMbase,CNKI,Wan fang and VIP database were searched for relevant articles published from January 2001 to December 2016 using Pancreatitis,Recurrence and Etiology as term index words.Meta analysis was conducted by RevManS.3 software.Results A total of 16 studies involving 3 980 patients (RAP n =1 231,AP n =2 749) were included.The age and sex were not correlated with AP recurrence.Biliary diseases,alcohol,inappropriate diets were associated with AP recurrence,but these were not the factors influencing RAP.Hyperlipidemia was responsible for the occurrence of pancreatitis in 17.00% of RAP and 10.20% of AP,with statistically significant difference (P =0.002).Hyperlipidemia was an important risk factor influencing AP relapse.The percentage of SAP in AP and RAP patients was 16.83% and 24.13%,respectively,and the complication rate was 11.43% and 15.13%,and the percentage of jaundice was 28.20% and 32.53%.Those in RAP were higher than those in AP,and the differences were statistically significant (all P < 0.05).Conclusions Hyperlipidemia was the predominant risk factor for RAP in China,and the patients with RAP tend to progress into SAP,and have jaundice and complications.
