2.Effect of Guishen Pill on expression levels of Oct-4, MVH, and Egr-1 in mice with diminished ovarian reserve.

OBJECTIVETo study the effect of Guishen Pill (GSP) on expression levels of Oct-4, MVH, and Egr-1 in mice with diminished ovarian reserve (DOR).
METHODSTotally 40 female C57BL/6J mice were randomly divided into 4 groups, the normal control group, the model group, the GSP group, and the dehydroepiandrosterone (DHEA) group, 10 in each group. Pregnant mare serum gonadotropin (PMSG), human chorionic gonadotropin (HCG), and prostaglandin F2α (PGF2α) were sequentially administrated to produce superovulation. The DOR model was established by exposing to ozone inhalation. Mice in the GSP group were intragastrically administered with GSP at 0.3 mL. Those in the DHEA group were intragastrically administered with DHEA at 0.3 mL. Equal volume of normal saline was intragastrically administered to mice in the normal control group and the model group. All mice wer treated for 21 days. Serum levels of estrogen (E2), progestogen (P), and anti-Müllerian hormone (AMH) were measured by ELISA. Changes of Oct-4, anti-AMH, and early growth response gene-1 (Egr-1) mRNA in ovaries were dtected by Real-time PCR.
RESULTSCompared with the model group, serum levels of E2, P, and AMH, as well as contents of estrogen receptor (ER), progestogen receptor (PR), MVH, and Oct-4 mRNA significantly increased in the GSP group and the DHEA group (P < 0.05).
CONCLUSIONGSP could improve expression levels of Oct-4, MVH, and Egr-1 mRNA in DOR mice and their ovarian function.

The aim of this study was to investigate the effect of Bu-Shen-An-Tai recipe (BSATR) and its two components (Bushen recipe, and Huoxue recipe) on endometrial morphology during peri-implantation in superovulated mice. Mice were randomly divided into five groups, including the normal (N), model (M), Bushen (BS), Huoxue (HX) and Bu-Shen-An-Tai (BH) groups. The uteri were collected on day 4 of pregnancy, and the endometrium thickness, microvessel density (MVD) and number of pinopodes observed. Compared with the M group, the endometrial thickness in the BS, HX and BH groups was significantly increased and there was a significant difference in endometrial thickness between the BS and the BH groups. The mean MVD was significantly lower in the M group than in the N group, and there was a significant increase in MVD in the BS, HX and BH groups as compared with the M group. Compared with the M group, the pinopode scores in the endometrium were significantly increased in the HX and BH groups; and the BS group had significantly higher pinipode scores than the HX and BH groups. In conclusion, the results of the present study demonstrated that the recipes (Bushen, Huoxue and BSATR) could improve the endometrial environment by regulating the endometrial thickness, MVD and the number of pinopodes at the window of implantation. Moreover, the Huoxue recipe and the BSATR were more efficient than the Bushen recipe, with the BSATR tending to have the most beneficial effects.

5.Cloning and prokaryotic expression analysis of HDS from Salvia miltiorrhiza bge.f.alba.

According to the designed specific primers of gene fragment based on the Salvia miltiorrhiza transcriptome data, with the method of reverse transcription polymerase chain reaction (RT-PCR), this study cloned full-length cDNA sequence of 1-hydroxy-2-methyl-2-(E)-butenyl-4-diphosphate synthase gene from Salvia miltiorrhiza bge.f.alba, this sequence is named as SmHDS and its GenBank registration number is KJ746807. SmHDS, 2 529 bp long, contains an ORF of 2 229 bp, encodes 742 amino acids, including 5' UTR 170 bp and 3' UTR 130 bp. Using bioinformatics software, having made a homology analysis of the obtained sequence, we can have a conclusion that SmHDS have a close genetic relationship with HDS of Salvia miltiorrhiza. Analysis result of prokaryotic expression revealed that in Escherichia coli, SmHDS expressed target proteins which in size are comparable with the protein predicted. Meanwhile, the 4 factors which can influence the protein expression were optimized, the 4 factors are inducing temperature, inducing time, IPTG concentrations and density of inducing host bacterium (A600). The optimal expression conditions of SmHDS were 30 degrees C until the A600 is 0.6, and add IPTG to a final concentration of 0.2 mmol x L(-1), and the induction time of 20 h. It provides theoretical basis for the further study of the function of 1-hydroxy-2-methyl-2-(E)-butenyl-4-diphosphate synthase in the biosynthesis of tanshinone compounds.

7.Effects of acupuncture on different acupoints in combination with rehabilitation on hemiplegic muscle spasticity in hemiplegia patients.

OBJECTIVETo observe the clinical effect of hemiplegic muscle spasticity treated with acupuncture on different acupoints in combination with rehabilitation.
METHODSNinety cases of post-stroke muscle spasticity were randomized into a corresponding and central axis acupuncture group (group A), a conventional acupuncture group (group B) and a rehabilitation group (group C), 30 cases in each one. In group A, the acupoints on the Governor Vessel were mainly selected, as well as those on Hand-Shaoyang meridian and Foot-Taiyang meridian. In group B, the conventional needling and rehabilitation training were applied in combination, of which, the acupoints were mainly from Hand and Foot-Yangming meridians, associated with the acupoints of Shaoyang meridian. In group C, only rehabilitation training was applied. The assessments according to modified Ashworth scale and CSS score were performed before treatment, after 2 weeks and 4 weeks treatment respectively.
RESULTSThe level of modified Ashworth scale of the flexor of elbow and wrist was reduced apparently after treatment in group A as compared with that before treatment (P < 0.01). The level of modified Ashworth scale of the flexor of elbow was reduced apparently after treatment in group C as compared with that before treatment (P < 0.05). There was significant difference on the level change in modified Ashworth scale for the flexor of elbow between group A and group C after 4 weeks treatment (P < 0.05). CSS score decreased significantly in group A as compared with that before treatment (P < 0.01), the improvement after treatment in group A was apparent as compared with the other two groups (P < 0.01).
CONCLUSIONThe corresponding and central axis acupuncture can improve muscle tone of hemiplegic limb, which is superior to the effect achieved by the conventional acupuncture in combination with rehabilitation training and the simple rehabilitation training.

8.Evidence synthesis of effects of physical exercise on fall prevention in community-dwelling elderly people

Objective To evaluate and summarize the best available evidence of effects of physical exercise on fall prevention in community-dwelling elderly people.Methods We searched Cochrane Library,JBI Library,Registered Nurses' Association of Ontario (RNAO),National Guideline Clearinghouse (NGC),PubMed,EMbase,Scottish Intercollegiate Guideline (SIGN),British Medical Journal Best Practice,British Medical Journal Best Practice,CBM,Wanfang Database and CNKI,to collect literatures including guideline,evidence summary(ES),best practice information sheet (BPIS),recommend practice (RP),system review(SR) and original research.Three authors independently evaluated quality of literatures and extracted data.Results Ten articles were enrolled,including four guidelines,two SRs,one RP,one ES,and two randomized controlled trials.Best evidence included:exercises should aim to provide moderate or high balance function training.Tai Chi and yoga were effective in fall prevention.Multifactorial assessment,group-based or home-based exercises were able to prevent falls.Conclusion Community should provide physical exercise recommendations for the elderly and satisfy the corresponding conditions.Available best evidence is recommended to use according to individual situation of the elderly.
9.Thermal stability of neuraminidase in influenza vaccine

Abstract:Objective To analyze the stabilities of neuraminidase(NA)in influenza vaccine at different temperatures and
provide a reference for further complete understanding of overall shelf life of vaccines. Methods Monovalent bulks of
influenza H1N1,H3N2 and B vaccines were stored at 4(low temperature),25(room temperature)and 37 ℃(changed
temperature)for 0. 5,2,7,24 and 48 h separately,using that at 100 ℃(extreme temperature)for 1 h as control,and
determined for NA activity by enzyme⁃linked lectin method. Results The NA activities of influenza H1N1 vaccines stored at
25 and 37 ℃ decreased significantly with the increasing of time. No significant decreases were observed in H3N2 and B
vaccines even after storage at two non⁃storage temperatures for 48 h. However,all the NA activities of three vaccines
decreased at 100 ℃. Conclusion Both H3N2 and B vaccines showed high stability at abnormal storage temperatures not
more than 37 ℃,while H1N1 vaccine was relatively sensitive to the temperature for storage.
10.Inhibitory effect of KAI1 gene on breast cancer cell growth in vitro.

OBJECTIVETo explore the inhibitory effect of KAI1 gene on breast cancer cell growth in vitro.
METHODSHighly metastatic human breast cancer cell line MDA-MB-231 was transfected with pCMV-KAI1 or mock transfected plasmid pCMV with lipofectamine. Western blot was used to determine the expression of target protein of KAI1. The proliferative ability of cells was tested by MTT assay and colony-forming test. The cell cycle pattern was assayed by flow cytometry. The metastatic ability was investigated by cell adhesion and invasion assays.
RESULTSA stable cell clone transfected with KAI1 gene was obtained and over-expression of KAI1 protein was observed. There was a significant decline in cell proliferative ability of pCMV-KAI1 transfected MDA-MB-231 cells in comparison with the mock-transfected ones and non-transfected ones, revealed by MTT assay and colony-forming test (P < 0.05). The ability of adherence and invasion of pCMV-KAI1 transfected cells was significantly reduced in comparison with the other two groups (P < 0.05). Also, flow cytometry analysis revealed that in KAI1 transfected cell group the number of cells in G0/G1 phase increased markedly from 36.78% +/- 0.61% to 64.00% +/- 7.56%, while the number of cells in G2/M phase decreased from 17.88% +/- 0.76% to 7.63% +/- 0.60%, comparing with the non-transfected ones.
CONCLUSIONKAI1 gene suppresses the invasive ability of human breast cancer cells in vitro and may inhibit the proliferative ability by changing the cell cycle pattern.
