2. Effect of Steric Hindrance on the Acylation Reaction of Pentacyclic Triterpenoids: Taking Glycyrrhetinic Acid and Oleanolic Acid as Examples
Chinese Pharmaceutical Journal 2018;53(22):1896-1900
OBJECTIVE To investigate the effect of steric hindrance on the acylation reaction of pentacyclic triterpenoids, taking glycyrrhetinic acid and oleanolic acid as examples. METHODS The parent nucleus of glycyrrhetinic acid and oleanolic acid were selected, for which the steric hindrance is different at the two carboxyl sites: one is non-angle, another is angle. The parent nucleus were combined with cyclopentylamine, cyclohexylamine, piperidine, and pyrrolidine, respectively. The four simple nitrogen-containing heterocycles are distinct from the four-membered or five-membered ring and the nitrogen atom is inside or outside the ring. By means of comparing and analyzing the acylation reaction results, the reasons influencing acylation reactions were explored. RESULTS Glycyrrhetinic acid had acylation reaction with four simple nitrogen-containing heterocycles normally, whereas oleanolic acid only formed a kind of intermediate active ester, “oleanolic acid-HOBt”, and no final target object could be obtained. CONCLUSION The effect of nitrogen-containing heterocycles on the acylation reaction is almost negligible, while the steric hindrance of different parent nucleuses has significant influence on the acylation reaction. This study is of reference significance for the acylation reaction of triterpenoid carboxyl groups.

The enzyme activity of ?- Acetolactate Decaroboxylases (ALDC) from different microbes was studied, the results demonstrated that it was quite different among them. There were diversities of their enzyme reaction velocities. It was clear that the enzyme activity was affected by the pH of the enzyme reaction system, for example, the optimum pH of ALDC from Lactococcus lactis was 6. 6, while for Aerobacter Aerogenes it was 5. 8. Addition leucine, valine and isoleucine into enzyme reaction system obviously affected the enzyme activity of ALDC from different microbes.

Objective To investigate the optimal concentration and site for ozoneadministration to relieve neuropathic pain in rats. Methods Twenty-four adult male rats with sciaticnerve injury (SNI) were randomized equally into 4 groups, and received ozone injections for analgesia atdifferent sites, namely the operative site, the sciatic nerve, the L5 spinal nerve or the operative site inaddition to the L5 spinal nerve. Another 4 groups of rats with SNI (n=6) had ozone injections at theconcentrations of 40, 50, 60, or 70 μg/mL, all administered at both the operative site and the L5 spinalnerve. The analgesic effects of the ozone injections were evaluated using paw withdrawal threshold test inall the rats. Results In rats with ozone injection at both the operative field and the L5 spinal nerve, themechanical pain threshold was significantly increased in comparison with that in rats with injections atother sites (P<0.05), but different doses of ozone simultaneously injected at the two sites did not producesignificant differences in the pain threshold (P>0.05). Conclusion Ozone produces obvious analgesiceffects in rats with SNI-induced neuropathic pain when injected at both the operative site and thespinal nerve, and ozone within the concentration range from 40 to 70 μg/mL has similar analgesic effect.

OBJECTIVETo investigate the effects of advanced glycosylation end products (AGEs) modified bovine serum albumin (AGE-BSA) on the expression of reactive oxygen species (ROS) and monocyte chemoattractant protein-1 (MCP-1) in human renal mesangial cells (HRMCs).
METHODSHRMCs were cultured in vitro with medium containing different doses of AGE-BSA or BSA (50,100, 200, 400 mg/L) for 48 hours, or with AGE-BSA (200 mg/L) for different times (12, 24, 48, 72 h). Immunocytochemistry assay was used to estimate the protein level of RAGE. The ROS in cells were measured by flow cytometry and the mRNA expression of MCP-1 were analyzed by semi-quantiative reverse transcription-polymerase chain reaction (RT-PCR) after treatment with AGE-BSA or BSA.
RESULTSThe protein level of RAGE was upregulated in the HRMCs with AGE-BSA. The expression of ROS and MCP-1 significantly enhanced by incubation of AGE-BSA in a time- and dose-dependent manner. The effects of AGE-BSA-induced up-regulation of ROS and MCP-1 level was significantly blocked by neutralizing antibodies to RAGE, while the expression of ROS and MCP-1 stood nearly unchanged after cultured with huamn IgG.
CONCLUSIONThe expression of ROS and MCP-1 in HRMCs is induced by AGE-BSA through RAGE, which may have potential effects in the pathgenic mechanism of diabetic nephropathy.

After 28 foreign species of AM fungi were inoculated in sterilized soil, the effects of the AM mycorrhizal colonization and the medicine quality of Paris polyphylla var. yunnanensis were observed by combination of inoculation test in pot at room temperature and instrumental analysis. The results showed that, compared with control group (CK), the inoculation of foreign AM fungi in the soil influenced the spore density, mycorrhizal infection rate, and colonization intensity of AM fungi in root system of P. polyphylla var. yunnanensis. The inoculation of foreign AM fungi enhanced the mycorrhiza viability of P. polyphylla var. yunnanensis by increasing the activity of succinic dehydrogenase (SDH) and alkaline phosphatase (ALP) in intraradical hyphae. The content of single steroid saponin in rhizome of P. polyphylla var. yunnanensis showed variation after P. polyphylla var. yunnanensis was inoculated by different foreign species of AM fungi, which was beneficial for increasing the medicine quality; however, the kinds of steroid saponin showed no difference. In a degree, there was a selectivity of symbiosis between P. polyphylla var. yunnanensis and foreign AM fungi. And we found that the Claroideoglomus claroideum and Racocetra coralloidea were best foreign AM fungi species for cultivating P. polyphylla var. yunnanensis under field condition.

Objective To study the effects of N-acetyl-seryl-aspartyl-lysyl-proline(AcSDKP) on proliferation and synthesis and secretion of extracellular matrix(ECM) in rat active hepatic stellate cells(HSCs). Methods Primary cultures of HSCs when actived were subjected to AcSDKP(0.01, 0.1, 1, 10 and 100 nmol/L, respectively) treatment, and control group was established. The expression of collagen Ⅰ(ColⅠ) and collagen Ⅲ(Col Ⅲ) mRNA were analyzed with RT-PCR. The proliferation of HSCs was detected by MTT. Hyaluronic acid (HA) and laminin(LN) in the supernatant were determined by enzyme-linked immunosorbent assay. Results Compared with control group, the proliferation of active HSCs was significantly inhibited by 1 and 10 nmol/L AcSDKP. 1, 10 nmol/L AcSDKP and 0.1 to 100 nmol/L AcSDKP significantly inhibited the expression of HSCs ColⅠ mRNA and Col Ⅲ mRNA, respectively. Expression of HA and LN in the supernatant were significantly inhibited by 0.1, 1, 10 nmol/L and 0.1, 1 nmol/L AcSDKP, respectively. ConclusionAcSDKP can inhibit synthesis and secretion of ECM in active HSCs in a dose-dependent manner, with a maximum inhibition effect at 1 nmol/L AcSDKP. The mechanism may involve the inhibition of the proliferation of HSCs,which leads to the decrease of HSCs that synthesize and secrete ECM.
