1. Expression of HIF-lalpha and VEGF in papillary renal cell cancer and clear cell renal cell cancer and the correlation between each other
Tumor 2007;27(6):469-472
Objective: To study the expression of hypoxia inducible factor (HIF)-1 alpha and vascular endothelial growth factor (VEGF) in papillary renal cell cancer (PRCC) and clear cell renal cell cancer (CC-RCC) and the correlation between each other. Methods: The expressions of HIF-1 alpha and VEGF in PRCC (n=13), CC-RCC tissues (n=20) and normal renal tissues (n=10) were detected by immunohistochemistry. The difference and correlation between each group were compared and analyzed. Results: The positive rates of HIF-1 alpha in PRCC, CC-RCC, and normal renal tissues were 38.46%, 85%, and 0%, respectively. There was significant difference between the three groups (P < 0.05). The positive rate of VEGF in PRCC, CC-RCC and normal renal tissues were 76.92%, 80%, and 10%, respectively. There was no significant difference between PRCC and CC-RCC. But the difference between renal cancer tissues and normal renal tissues was significant (P < 0.05). Expression of HIF-1alpha and VEGF had no correlation in PRCC tissues but had positive correlation in CC-RCC tissues. Conclusion: HIF-1 alpha is weakly expressed and VEGF is over-expressed in PRCC tissues. The expressions of HIF-1alpha and VEGF have no correlation. It suggests that the pattern of tumor angiogenesis in PRCC was different with CC-RCC.

In this study, we use pot experiment to evaluate the effect of plant growth regulator on plant morphology and biomass allocation of Salvia miltiorrhiza. Different concentrations of uniconazole were supplied to S. miltioohiza by means of foliar spray. Height, breadth and stem diameter were measured dynamically, the biomass of leaf, stem, flower and fruit, root biomass and biomass ratio were also examined at the harvest time. Owing to the treatment, plant morphology showed significant changes, the height had been greatly reduced and the breadth decreased largely. Meanwhile, the biomass allocation changed too. The biomass ratio of leaf and stem had been notably reduced while the biomass ratio of root had been increased remarkably. It appears that foliar application of uniconazole during vigorous growth period in S. miltioohiza has dramatic effect on dwarfing plant and improving resistant to lodging. This measure could also be applied to condensed cultivation of S. miltioohiza to increase production.

6.Efficacy and safety of tadalafil for erectile dysfunction: an updated review.
National Journal of Andrology 2009;15(10):956-959
As a long-acting phosphodiesterase type 5 (PDE5) inhibitor, tadalafil is administered orally as the first line therapy for erectile dysfunction (ED). Its efficacy and safety have been confirmed by many clinical studies in the treatment of ED in general patients, elderly patients and those with diabetes mellitus or spinal cord injury or after prostate cancer surgery. With its prolonged action of 36 hours, tadalafil can not only increase the self-esteem of ED men but also improve the quality of life of both the patients and their partners.

This study was conducted to investigate the paclitaxel loaded by hydrazone bonds in poly(ethylene glycol)-poly(caprolactone) micelles (mPEG-PCL-PTX) on proliferation and apoptosis of human lung cancer A549 cells and its possible mechanisms of anti-tumor activity. The cell proliferation was measured with MTT assay. Flow cytometry were used to analyze the cell cycle. The cell apoptosis was analyzed using Hoechst/P staining. The expression levels of apoptotic genes expression in the mitochondrial apoptosis pathway were detected by RT-PCR and Western blotting, respectively. The mPEG-PCL-PTX could inhibit the proliferation of A549 cells and promote the apoptosis. The Bax, caspase-3 protein expression were increased while Bcl-2 protein expression was decreased in A549 cells. Results showed that the polymer containing hydrazone bond is non-toxic in vitro, the mPEG-PCL-PTX micelles can inhibit the proliferation and induce the apoptosis of A549 cells. To investigate the effect of GST-KGDX (glutathione S-transferase-Lys-Gly-Asp-X) fusion protein, GP IIb/IIIa receptor antagonist, on platelet function in vitro. The KGDX (Lys-Gly-Asp-X) gene was assembled from 2 synthetic oligonucleotides, 36 bp in length, using BamH I and Xho I restriction enzyme sites at the end of the gene for cloning into the expression vector pGEX4T-1. Expression of fusion protein was directed by the tac promoter. The Escherichia coli DH5a contained the plasmid pGEX-4T-1-KGDX was expressed by 37 degrees C heat induction. The fusion protein of KGDX with glutathione S-transferase (GST-KGDX) was purified in one step from the bacterial lysate by glutathione-agarose beads for affinity chromatography. GST-KGDX was found to be soluble and abundant, the yield of 35 mg/L of cultures was obtained. The GST-KGDX was expressed in E. coli to a level of 48.02% of total cellular protein. GST-KGDX inhibited ADP-induced human platelet aggregation stronger than GST (P < 0.05 or < 0.01). In flow cytometry assay for fibrinogen binding, both GST and GST-KGDX inhibited platelet aggregation by binding with high affinity to GPIIb/IIIa. Mean fluorescence intensity of GST-KGDX fusion protein was significantly higher than that of GST. It is concluded that the GST-KGDX fusion protein can be produced by E. coli and used as an antiplatelet agent.
