Objective To understand the gene characteristics of envelope region of human T lymphotropic virus type Ⅰ(HTLV Ⅰ)from Shantou region of China. Methods The gene of proviral envelope region of HTLV Ⅰ Shantou strain (named HTLV Ⅰ WHP)was amplified by PCR,using 4 designed primers, and then was sequenced, meanwhile it was compared with that of various HTLV Ⅰ representative strains from all around the world, and subsequently it was analyzed to construct its phylogenetic tree. Results The nucleotide sequence of complete envelope region of HTLV Ⅰ WHP provirus we have obtained was 1466 bp.There no deletion and insertion in the deducted amino acid sequence. The results of phylogenetic tree analysis showed that the HTLV Ⅰ WHP strain was most closed to some of the strains from Japan and Caribbean, and like the latter, belonged to the subtypea of cosmopolitan group. Conclusion The HTLV Ⅰ strains either from the coastal region of Southeast China or from Japan,Caribbean and so on have the common origin of evolution.

Objective cyclin D1 gene plays a significant role in regulating cell cycle progression.Suppression of cyclin D1 protcin expression can effect on cellular proliferation, distribution of cell cycle and apoptosis. This study was to determine whether this effect also existed in chronic leukemia ceil line K562 by inhibiting the expression of cyclin D1 protein through RNA interference in vetro. Methods Plasmid vectors expressing small hairpin RNA (shRNA) targeting at cyclin D1 gene were constructed and transfected into K562 cells by chitosan, cyclin D1 protein was examined by using Western blot analysis.Inhibition of cellular proliferation was evaluated hy soft agar colony formation assay. The cell cycle and apoptosis were determined by flow cytometry. Results Expression of cyclin D1 protein was markedly down-regulated and capability of colony formation was suppressed after transfection with pshRNA-419 and pshRNA-575 at 48h.Down-regulation of cyclin D1 protein could effect on distribution of cell cycle arrested at G_0/G_1 phase and markedly induce apoptosis of K562 cells. But there had no above biological effects ob- served after transfection with blank vector and control vector of m-pshRNA-790.Conclusion Down-regulation of cyclin D1 expression can inhibit growth of K562 cells, and effect on distribution of cell cycle arrested at G_0/G_1 phase. The primary results suggest that cyclin D1 gene might serve as an effective target for the treatment of leukemia.

Chinese Journal of Endocrinology and Metabolism 2001;0(05):-
Transforming growth factor-?_1 (TGF-?_) mRNA or protein expression in renal cortex of diabetic rats was assessed by real-time quantitative RT-PCR with SYBR Green, immunohistochemistry or Western blot. After melatonin treatment, the expressions of TGF-?_1 mRNA and protein were decreased, suggesting that beneficial effect of melatonin may result from its antioxidative property and inhibiting TGF-?_1 expressions.

Objective To apply autoimplantation technique in the treatment of verruca plana, and to investigate the mechanisms of it. Methods The changes of T cell subsets and IL-2 level in the peripheral blood of the patients with verruca plana, in conjunction with histopathological and immunohistochemical features of verruca plana tissue at reactive stage were studied by APAAP and radioimmunoassay, respectively. The tissue specimens were studied by H.E stain and immunohistochemical techniques. Results Significantly decreased CD4+ cell numbers, CD4+/CD8+ ratio and IL-2 level were found before treatment, compared with those in healthy controls (P 0.05). Lymphocytic infiltration, epidermal edema and dissolution were shown in the histopathological study. There were CD45RO+, CD6+, CD4+ and CD8+ cells in the lymphocytic infiltration, but no CD20+ and Mac387- cells. Langerhans cells with positive S-100 protein were observed in basal layer of the epidermis and reaction area. Conclusions T cell-mediated immune responses could be improved in autoimplantation, the warts are rejected mainly by of CD8+ subset cytotoxicity.

AIM:To study the effect and the molecular mechanism of CDX 2 over-expression on the prolifera-tion, growth and cell cycle of human gastric cancer cell line SGC-7901.METHODS:The SGC-7901 cells in LV-CDX2-GFP group were transfected with the recombinant lentivirus vector LV-CDX2-GFP, the cells in LV-GFP group were trans-fected with the negative control lentiviral vector for the negative control , and the cells in blank control group were without any treatment. The cell proliferation was detected by CCK-8 assay. The cell cycle distribution was analyzed by flow cytome-try.The expression of CDX2, Bax, Bcl-2, cyclin D1 and survivin was determined by semi-quantitative RT-PCR and Wes-tern blotting .RESULTS:Compared with LV-GFP group and blank control group , the proliferation activity of the SGC-7901 cells was significantly lower (P<0.05), the G0/G1 phase proportion increased (P<0.05), the mRNA and protein levels of Bcl-2, cyclin D1 and survivin were reduced (P<0.05), and the mRNA and protein levels of Bax were up-regula-ted (P<0.05) in LV-CDX2-GFP group. No statistically significant difference of the above indexes was observed (P>0.05) between LV-GFP group and blank control group .CONCLUSION:Over-expression of CDX2 mediated by lentivirus inhibits the proliferation and growth of human gastric cancer SGC-7901 cells and arrestes the cell cycle at G 0/G1 phase, which may be related to down-regulation of Bcl-2, cyclin D1 and survivin and up-regulation of Bax .

Acta Pharmaceutica Sinica 2015;50(6):738-745
Poly(ADP-ribose)polymerase-1 (PARP-1) plays a significant role in the DNA repair process by catalyzing the transfer of ADP-ribose from NAD+ to its receptors. It is a promising anticancer drug target and many PARP-1 inhibitors have been developed and used in the clinical trial. In this work, a series of 3-(2-oxo-2-substituted acetamido)benzamides have been synthesized and their inhibitory activities against PARP-1 were evaluated. Of all the tested compounds, six compounds displayed inhibitory activities with IC50 values ranging from 0.23 to 5.78 µmol.L-1 . The binding pose of compound 5a was predicted using molecular docking to facilitate further structural modification.

To study the function of expelling water retention with drastic purgative of different polarities of Kansui Radix stir-baked with vinegar on the cancerous ascites model rats, the furosemide was taken as positive control drug, and the cancerous ascites model rats were respectively orally administered with different polarities of Kansui Radix stir-baked with vinegar for 7 d. The amount of urine and ascites, the level of urinary sodium, potassium, chloride ion and pH, and the content of PRL1, AII, ALD in serum were investigated. Compared with model groups, ethyl acetate extract group showed a decreasing trend in ascites; the amount of urine of showed a significant increase (P < 0.05); the level of urinary sodium, potassium, chloride ion (P < 0.05, P < 0.01), pH (P < 0.05), and the content of PRL1, AII, ALD in serum all showed a significant decrease (P < 0.01). The effects of petroleum ether extract and n-butanol extract were weaker than that of ethyl acetate extract. The water exact was the weakest. The results showed that ethyl acetate extract is the active part of Kansui Radix stir-baked with vinegar on the function of expelling water retention with drastic purgative on the cancerous ascites model rats, alleviating the water-electrolyte disorder and body fluid acid-base imbalance, regulating the renin angiotensin aldosterone system.
