2.Simultaneous determination of seven constituents in “ Zhongchai No.2 ” by HPLC

AIM To establish an HPLC method for the simultaneous content determination of saikosaponin a,saikosaponin c,saikosaponin d,rutin,quercetin,isorhmnetin and kaempferol in the aerial and underground parts of “Zhongchai No.2” (Bupleurum chinense DC.from Sichuan Province).METHODS The analysis of “Zhongchai No.2” methanol extract was performed on a 30 ℃ thermostatic Hypersil GOLD column (250 mm × 4.6mm,5.0 μm),with the mobile phase comprising of acetonitrile-0.1% phosphoric acid flowing at 0.8 mL/min in a gradient elution manner,and the detection wavelength was set at 210 nm.RESULTS Seven constituents showed good linear relationships within their own ranges (r≥0.999 1),whose average recoveries were 96.30%-102.7% with the RSDs of 0.95%-3.8%.There were a lot of flavonoids in the aerial parts of samples,while a large number of saponins existed in the underground parts,but almost no flavonoids were found.CONCLUSION The aerial parts (stems and leaves) of “Zhongchai No.2” also have medicinal value,to which we should pay attention.
3.Pneumorrhagia and pneumothorax occurring after CT-guided cutting needle biopsy for pulmonary solid nodules: a multivariate analysis

Objective To investigate the risk factors of pneumorrhagia and pneumothorax occurring after CT-guided cutting needle biopsy for pulmonary solid nodules.Methods The clinical data and imaging materials of 320 patients with pulmonary solid nodule (≤3 cm),who received 16 G semi-automatic cutting needle biopsy,were retrospectively analyzed.Both univariate and multivariate logistic regression analysis methods were used to evaluate the risk factors of pneumorrhagia and pneumothorax.Results The incidence of needle track hemorrhage was 33.1% and the incidence of pneumothorax was 18.1%,the diagnostic accuracy for benign and malignant nodules was 99.6%.Needle track length was an independent risk factor for pneumorrhagia;every increase of 3 cm in needle track length,the risk of pneumorrhagia would increase 3.881 times,besides,the risk of pneumorrhagia had a parallel relationship with the time used for puncturing (P=0.061) as well as with the number of pleural puncturing (P=0.062).The age,lesion's location and needle-pleural angle were independent risk factors for pneumothorax.Each increase of 10 years in age,the risk of pneumothorax would increase 2.102 times.The pneumothorax risk of pulmonary lesion located at upper lung lobe was strikingly lower than that of pulmonary lesion located at lower lung lobe.Each increase of 20 degrees in needle-pleural angle,the risk of pneumothorax would increase 2.413 times.Emphysema was excluded from the equation because it had only a minute difference (P=0.086).Based on the pneumorrhagia and pneumothorax probability values,ROC curves of pneumorrhagia and pneumothorax were drawn,and AUC values of pneumorrhagia and pneumothorax were 0.753 and 0.725 respectively.Conclusion The occurrences of pneumorrhagia and pneumothorax after CT-guided cutting needle biopsy for pulmonary solid nodules are affected by many factors.Careful preoperative evaluation and skilled intraoperative manipulation can effectively reduce the incidence of pneumorrhagia and pneumothorax.
4.The effect of miRNA-7 on chemoresistance in esophageal cancer cell TE-1

Objective To explore the impacts of over-expression of microRNA-7 (miRNA-7) on the sensitivity of cis-platin in esophageal carcinoma cell line TE-1, and the possible mechanism thereof. Methods Lipofectmin 2000 method was used to transient transfect with miRNA-7 mimic into esophageal cancer cell line TE-1, which was taken as transfection group, mimic negative control was taken as transfection conrtol group. The expressions of miRNA-7 and epidermal growth factor receptor (EGFR) mRNA were detected by RT-PCR in the above two groups and normal control group. The total EGFR and EGFR in cytoplasmic and nucleus were detected with Western blot assay in transfection group and transfection control group. CCK-8 was used to detect IC50 of cisplatin in transfection group and transfection control group. The expression of EGFR was observed with immunofluorescence confocal microscope in two groups. Results The miRNA-7 expression was signifi-cantly increased in transfection group than that of transfection conrtol group and control group. The expression of EGFR mRNA was significantly reduced in transfection group (P<0.001). The total EGFR was significantly decreased in transfec-tion group than that of transfection conrtol group. The level of nuclear EGFR was significantly increased ( P<0.01),and cyto-plasm EGFR expression was significantly decreased in transfection group than that of transfection control group ( P<0.05). CCK-8 results showed that after the over expression of miRNA-7 in TE-1, the IC50 of cisplatin (48 h) increased in transfec-tion group than that of control group (P<0.01). Immunofluorescence results showed that EGFG in nuclear was higher in transfection group than that of transfection control group but its expressions reduced in cell membrane and cytoplasm. Con-clusion The over-expressed miRNA-7 in esophageal cancer cells TE-1 can reduce cisplatin sensitivity by the increased EGFR in nuclear translocation.
5.Craniofacial characteristics of the Class II division 1 malocclusion patients with mouth-breating: a posteroanterior.

OBJECTIVETo evaluate the craniofacial characteristics of the Class II malocclusion patients with mouth-breating by posteroanterior cephalometry.
METHODSTo measure craniofacial width of the 12 Class II malocclusion patients with mouth-breathing, and to compared these measures with corresponding measures in a group of normal children.
RESULTSThe width of the maxillary base bone (J-J) was less than that in normal children significantly (P < 0.01). The mouth-breathing children's upper and lower arch width (at first molar and cuspid) were comparatively narrower, and lateronasal width (Lap-Lap) was narrower too.
CONCLUSIONMouth breathing may lead to craniofacial morphological abnormal development in craniofacial transverse structures.

6.Death caused by anaphylactic shock: a forensic pathological analysis of 142 cases.

OBJECTIVE:
To explore the forensic pathological features of death caused by anaphylactic shock.
METHODS:
One hundred and forty-two death cases of anaphylactic shock were retrospectively analyzed. The IgE level in the serum of anaphylactic shock cases were statistically compared with that of 62 non-anaphylactic shock cases.
RESULTS:
Most cases (77.46%) of anaphylactic shock death occurred in the medical institutes, with intravenous drug administration accounting for 53.53% of anaphylactic shock death. β-Lactam antibiotics, glucocorticoid and herbal medications were responsible for a significant proportion of such cases. Although characteristic histopathological changes were absent in vast majority of these anaphylactic shock cases, the differences of IgE levels in the serum between anaphylactic shock group and non-anaphylactic shock group were statistically significant (P<0.05).
CONCLUSION
Combined information including clinical data, autopsy results, IgE level, and other specific test results should be evaluated together in the forensic pathological diagnosis of anaphylactic shock.

8.The influence of infrasound on pathological and ultrastructural changes in the renal tissue of rats

Objective To investigate renal tissue changes in rats exposed to 16 Hz/130 dB infrasound.Methods Twenty male Sprague-Dawley adult rats were randomly divided into a control group,a pseudo-infrasound group,and two infrasound groups-A and B.Groups A and B were exposed to 16 Hz/130 dB infrasound for 2 h or4 h daily respectively over 7 days.Pathological and uhrastruetural changes in their renal tissues were observed with a light microscope and an electron microscope.Results Under the light microscope,Bowman's capsules expanded,epithelial cells were shed,and a little secretion was found in the renal tubules of infrasound group A.Slight degeneration and necrosis of the renal tubules and glomerular exudation could be observed in group B.Under the electron microscope,a large number of lysosomes displayed hyperplasia,there was interstitial edema,and leukocyte pavementing wag found in group B.Other changes such as swelling of podocytes' processes,fusion of foot processes,and vacuolization in the mitochondria could be observed in both infrasound groups.Conclusion Lengthy exposure to 16 Hz/130 dB infrasound can cause severe pathological and uhrastruetural changes in renal tissue,at least in rats.
9.Effects of paclitaxel loaded-drug micelles on cell proliferation and apoptosis of human lung cancer A549 cells.

This study was conducted to investigate the paclitaxel loaded by hydrazone bonds in poly(ethylene glycol)-poly(caprolactone) micelles (mPEG-PCL-PTX) on proliferation and apoptosis of human lung cancer A549 cells and its possible mechanisms of anti-tumor activity. The cell proliferation was measured with MTT assay. Flow cytometry were used to analyze the cell cycle. The cell apoptosis was analyzed using Hoechst/P staining. The expression levels of apoptotic genes expression in the mitochondrial apoptosis pathway were detected by RT-PCR and Western blotting, respectively. The mPEG-PCL-PTX could inhibit the proliferation of A549 cells and promote the apoptosis. The Bax, caspase-3 protein expression were increased while Bcl-2 protein expression was decreased in A549 cells. Results showed that the polymer containing hydrazone bond is non-toxic in vitro, the mPEG-PCL-PTX micelles can inhibit the proliferation and induce the apoptosis of A549 cells. 10.Factors Related with Neurological Function in Children after Hemispherectomy for Intractable Epilepsy

Objective To explore the factors related with neurological function in children after hemispherectomy for intractable epilep-sy. Methods Thirty-three children suffered hemispherectomy from May, 2014 to June, 2015 were analyzed. Their preoperative data were col-lected. A structured questionnaire was used to evaluate the neurological function. The relationship between preoperative parameters and post-operative functional outcomes was analyzed. Results Bilateral lesions in MRI (P<0.001) and age (P<0.001) were related with functional out-come. Conclusion The prognosis of hemispherectomy for children with intractable epilepsy is related to the contralateral lesion and age.
