1.Comparison of intraocular pressure measured by Icare rebound tonometer and Goldmann applanation tonometer

Background As a new tonometer,it is necessary to assess the clinical value of Icare rebound tonometer.Objective This study was to compare the intraocular pressure(IOP)values measured by Icare with that measured by GAT,and discuss the clinical value of leare rebound tonometer. Methods IOP measurement was performed on 152 eyes of 78 subjects with suspicious glaucoma,glaucoma,refractive error and normal examinnee by Icare and GAT respectively.The Icare IOP was measured firstly and then the GAT IOP was carried out with the 3-or 5-minute interval.The IOP values were compared between Ieare and GAT.This study was approved by Ethic Committee of Wuhan General Hospital of Chinese PLA.Written informed consent was obtained from each subject prior to this study. Results The mean IOP values of Icare and GAT were(19.16±5.03)mmHg and(18.41±4.52)mmHg respectively.The differences between Icare IOP and CAT IOP were less than or equal to 1 mmHg in 96 of 105 eyes(63.2%).The positive correlation was found between the Icare IOP and GAT IOP(r=0.940,P<0.01).The Ieare IOP was lower than that of GAT when IOPIcare<16 mmHg,however,the IOP of Icare were higher when IOPIcare≥6 mmHg;the IOP of Icare were higher than that of GAT in the total CCT range.The correlation coefficients of IOP of Icare or CAT with CCT were 0.341(P<0.01)and 0.333(P<0.01),respectively. Conclusion Compared with GAT,Icare is more feasible in clinic because it is practicable and reliable.
2.Clinical Significance and Influence on Glucocorticoid Receptor Resulted from Tripterygium Wilfordii Hook F for Children with Nephrotic Type of Henoch-Schonlein Purpura Nephritis

Objective To explore the mechanism of tripterygium wilfordii hook f(TWHF)for Henoch-Schonlein purpura nephritis(HSPN,nephrotic type)in children.Methods All the children with HSPN(nephrotic type)were divided into 3 groups casually.Method of radioactive pairs conjugating analysis was used to determine the numbers of glucocorticoid(GC)receptors(GCRs).Results were statistically analysed respectively.Results GCRs tested in all the patients with HSPN(nephrotic type)before treatment were statistically lower than those in control group of normal children;GCRs in all the patients tested after 1 week treatment with GC were lower than before treatment.Three weeks later,GCRs in patients treated with TWHF increased much higher than them without TWHF.Following up the recurring times of all the patients 3 months to 5 years,they were much more in patients treated without TWHF treatment than in patients treated with it.Conclusions TWHF treatment on HSPN(nephrotic type)is obviously effective.It can not only decrease the dosage of GC,but decrease the recurring times.One of the mechanism may be TWHF can resist the down-regulation GC to GCR,and enhance the effect of GC.
4.Construction of Hexose Transporter-like HXT1 Deletion Mutant in Pichia pastoris

Glucose was transported by the large number of hexose transporters in yeast cells. There were 18 hexose transporter genes had been identified in Saccharomyces cerevisiae. However,as an excellent expression system,there was no information of these genes had been reported in Pichia pastoris. Based on high homologous recombination efficiency in yeast,we chose G418 resistance for screening,200 bp were cloned from the up and down sequences of HXT1 ORF respectively,then ligated to the 5′ and 3′ end of G418 resis-tance gene for recombination. After electroporation of GS115 spheroplast and screened through different G418 concentration plates,finally we obtained one HXT1 gene deletion mutant named GS115?HXT1. The growth rate and glucose consumption of this mutant were both lower than the wide type.
5.Effects of water extraction of Salvia miltiorrhiza in sperm quality of mice

Objective To research effects of water extraction of Salvia miltiorrhiza in sperm quality of mice. Methods Forty adult male Kunming mice were randomly divided into 5 groups, including low concentration group, medium concentration group, high concentration group, control group and positive control group in eight mice in each group. The low, medium and high concentration groups were given by gavage with 1 500, 3 000 and 6 000 mg/kg water extraction of salvia miltiorrhiza once a day respectively, while the control group was given gavage with 10 mL/kg normal saline daily, as well as the positive control group was injected with 100 mg/kg cyclophosphamide once; All mice were sacrificed after 30 days. We measured the weight gain, viscera coefficient and sperm quality in each mouse. Results There were no statistical difference in the weight of every week, increase of the weight, testis and epididymis coefficients among the control group, the low, medium and high concentration groups (P > 0.05), while the weight and weight increase were significantly higher than that in the positive control group (P < 0.05). Sperm concentration, viability, morphology, motility, PR, VSL, VAP of low, medium and high concentrations group were significantly improved than the control group (P < 0.05), and the sperm quality in the four groups were higher than that in the positive control group (P < 0.05). Conclusion Water extraction of salvia miltiorrhiza can obviously improve the sperm viability, morphology, and motility of mice.

7.Relationship between social support and occupational stress.

OBJECTIVETo explore the relationship between social support and occupational stress.
METHODSA cross-sectional survey involving 654 career people from different industries was conducted. Social support, the occupational stressors, personalities, and occupational strain were measured by using Occupational Stress Instrument.
RESULTSThe people from different occupations got different level of social support. Social support scores were significantly positively related to interpersonal relation, responsibility for people, responsibility for thing, advancement and participation, task identity, training adequacy, challenge, autonomy, job satisfaction, mental health, affective ambulance, positive emotion, self-esteem and coping strategy (P<0.05 or P<0.01), negatively to careers, role conflict, role ambiguity, job future ambiguity, negative emotion, depression symptoms, daily life stress and work locus of control scores (P<0.05 or P<0.01). The scores of interpersonal relation, responsibility for people, responsibility for thing, advancement and participation, task identity, training adequacy, challenge, autonomy, job satisfaction, mental health, affective ambulance, positive emotion, self-esteem and coping strategy with sufficient social support were significantly higher than those of deficient social support (P<0.05 or P<0.01). The scores of role conflict, role ambiguity, job future ambiguity, negative emotion, depression symptoms, daily life stress and work locus of control were higher than those of the latter (P<0.05 or P<0.01). Stepwise regression analysis showed that social support was the predictor of depression (R(2)>0.05).
CONCLUSIONSocial support affects occupational stress largely and plays an important role in maintaining mental health of career people. The people from different occupations get different level of social support.

8.Efficacy and safety of non-indwelling urinary catheter during perioperative period of cesarean section:a Meta-analysis

Objective To systematically assess the efficacy and safety of non-indwelling urinary catheter during perioperative period of cesarean section.Methods The databases including Medline(PubMed),EMbase,Cochrane Library,CNKI and WanFang Data were retrieved by computer.The randomized controlled trials (RCTs) on the efficacy and safety of non-indwelling urinary catheter during perioperative period of cesarean section were comprehensively collected.Two reviewers independently screened literatures,extracted the data and assessed the quality.The Meta-analysis was conducted by using RevMan 5.3 software.Results A total of 5 RCTs involving 1 090 patients were included.The meta-analysis results showed that compared with the indwelling urinary catheter group,although the non-indwelling urinary catheter group increased the incidence rate of urinary retention[RR=11.67,95%CI(2.22,61.24),P<0.01],but significantly decreased the incidence rate of urinary tract infection(UTI) [RR=0.10,95%CI(0.04,0.26),P< 0.01] and incidence rate of initial urination discomfort [RR=0.17,95%CI (0.04,0.74),P=0.02],significantly shortened the initial time of off-bed activities [SMD=-3.68 ,95%CI(-5.25,-2.12),P<0.01]and hospitalization time [SMD=-1.03,95%CI(-1.67,-0.38),P<0.01].There were no significant differences in the operation time[SMD=-0.13,95%CI(-0.32,0.07),P=0.20] and incidence rate of postpartum hemorrhage (PPH)[RR=1.50,95%CI (0.43,5.26),P=0.53],The incidence rates of bladder injury in the two groups were similar.Conclusion According to the evidence of current clinical researches,selecting non-indwelling urinary catheter in elective cesarean section is effective and safe,prompting that indwelling urinary catheter during the perioperative period of cesarean section is selective rather than routine indwelling.

10.Prognostic value of CD4+CD25+ Tregs as a valuable biomarker for patients with sepsis in ICU

BACKGROUND: Sepsis is a common complication of infections, burns, traumas, surgeries, poisonings, and post-cardiopulmonary resuscitation. The present study aimed to investigate prognostic value of CD4+CD25+ regulatory T cells (Treg) in peripheral blood of patients with sepsis. METHODS: Periphery blood from 28 patients diagnosed with sepsis was collected on day 1 and 7 after hospitalization in the ICU of Shanghai Changzheng Hospital between December 2013 to April 2014. The blood was used for analyses of Treg ratio using flow cytometry and for analyses of blood routine test, C-reactive protein (CRP), bilirubin, procalcitonin (PCT), and coagulation. APACHE II and sequential organ failure assessment (SOFA) scores were also investigated. The results were compared between two outcome groups of survival or death to evaluate prognostic value for sepsis. RESULTS: The patients had an average age of 60.36±15.03 years, APACHE II score 16.68±7.00, and SOFA score 7.18±3.78. Among the 28 patients, 12 had severe trauma (42.9%), 10 had septic shock (35.7%), and 9 (32.2%) died. The median ratio of Tregs was 2.10% (0.80%, 3.10%) in the survival group vs. 1.80% (1.15%, 3.65%) in the death group (Z=–0.148, P=0.883) on day 1; however it was significantly changed to 0.90% (0.30%, 2.80%) vs. 5.70% (2.60%, 8.30%) (Z=–2.905, P=0.004). CONCLUSION: With better prospects for clinical application, dynamic monitoring of Tregs ratio in peripheral blood has potential value in predicting prognosis of sepsis.
