1.Clinical evaluation of the preventing effect of 4 anti-caries preparations on enamel decalcification in orthodontic treatment with fixed appliance

Objective:To evaluate the preventing effects of 4 anti-caries preparations on enamel demineralization in fixed appliance orthodontic treatment.Methods:200 patients treated with fixed appliance were included and randomly divided into 4 groups (n =50),the teeth were treated with fluoride-containing toothpaste,fluoride varnish,fluoride free toothpaste and casein phosphopeptide-amorphous calcium phoaphate(CPP-ACP) respectively.The enamel decalcification incidence was calculated after orthodontic treatment.Results:The incidence of enamel decalcification calculated by tooth number was 16.3% in fluoride-containing toothpaste group,9.3% in fluoride varnish group,21.9% in fluoride free toothpaste and 8.5% in CPP-ACP group(among groups,P <0.05;between CPP-ACP and fluoride varnish,P > 0.05).Conclusion:Fluoride containing preparation can prevent the enamel demineralization during the fixed appliance orthodontic treatment,CPP-ACP and fluoride varnish are more effective.
2.Effect of human cementum extract prepared by guanidine on the attachment of gingival fibroblasts and osteoblasts

Objective: To study the bioactivity of human cementum extract . Methods: Cementum was harvested from freshly extracted heatlhy teeth and cementum extract was prepared by guanidine. Human gingival fibroblast and MC3T3 E1 osteoblast were cultured. The cells were incubated in DMEM with cementum extract concentration 0, 2.5, 5, 10 or 20 ?g/ml for an hour respectively, the cell attachment rate was measured by cell counting. The attachment rate of the cells in different incubation time(30 min,60 min, 90 min, 120 min) in DMEM containing 10 ?g/ml cementum extract was also assayed.Results: The extract increased the cell attachment rate in a concentration and time dependent effect. The extract at the concentration of 10 ?g/ml and with exposuse time of 90 min gave the most effective increase of cell attachment. Conclusion: Human cementum extract prepared by guanidine contains some bioactive proteins that promote the attachment of gingival fibroblasts and osteoblasts.
3.Simultaneous Determination of 1-Chloroanthracene and9-Bromoanthracene by Using Matrix Isopotential SynchronousFluorescence Spectrometry Combined with Derivative Technique

Matrix isopotential synchronous fluorescence spectrometric (MISF) method combined with derivative technique has been proposed to determine two spectrally-badly-overlapped anthracene derivatives, 1-chloroanthracene and 9-bromoanthracene,simultaneously. An optimal scanning route of MISF spectra was chosen. By using the novel method, 1-chloroanthracene and 9-bromoanthracene have been determined directly and simultaneously without tedious pre-separation in its mixture.The recoveries of 1-chloroanthracene and 9-bromoanthracene for the analysis are 83.5%~94% and 90.0%~94.0% respectively and their limits of detection are 0.69μg/L and 10μg/L respectively.

Livin is a new and important member of the inhibitor of apoptosis protein(IAPs)family.Livin is high expressed in most human tumor cells, involved in apoptosis inhibition and closely related with tumor development and progression.Studies found that through the down-regulation or inhibition of Livin expression and the induction specific antibodies of Livin could inhibi and kill tumor cells, which provided a new method for tumors'diagnosis, cell and gene targeted therapy.
6.The application of facial liposuction and fat grafting in the remodeling of facial contour.

OBJECTIVETo investigate the application of facial liposuction and fat grafting in the remodeling of facial contour.
METHODSFrom Nov. 2008 to Mar. 2014, 49 cases received facial liposuction and fat grafting to improve facial contours. Subcutaneous facial liposuction with tumescent technique and chin fat grafting were performed in all the cases, buccal fat pad excision of fat in 7 cases, the masseter injection of botulinum toxin type A in 9 cases, temporal fat grafting in 25 cases, forehead fat grafting in 15 cases.
RESULTSMarked improvement was achieved in all the patients with stable results during the follow-up period of 6 - 24 months. Complications, such as asymmetric, unsmooth and sagging were retreated with acceptance results.
CONCLUSIONCombination application of liposuction and fat grafting can effectively and easily improve the facial contour with low risk.

The introduction of highly active antiretroviral therapy (HAART) has greatly changed the pattern and natural history of ocular diseases of HIV-infected patients, resulting from the immune recovery and reduction of opportunistic infections. However, ophthalmic complica-tion continues to be concern in AIDS even in the HAART era, especially in developing areas, where absolute majority of HIV-positive patients live. Lack of test facilities and experience, poor conditions of hygiene, different microbiological environment, absence of effective treatment etc., characterize the ophthalmic manifestation of HIV-infected patients in developing countries from that in developed regions and thus pose a great challenge to the ophthalmic treatment in developing area. Not only varied from region to region, ocular complications are distinctive between adults and children. At the same time, the side effects due to the application of HAART pose their own risks of ocular complication and should, therefore, be given more research attention.

The tissue-specific and MeJA-induced transcriptional levels of BcUGT3 and BcUGT6 in Bupleurum chinense were analyzed in the present study. The transcriptional levels of BcUGT3 in root, leaf, flower and fruit were similar and they all were higher than those in stem. The transcriptional level of BcUGT6 was the highest in leaf and the lowest in flower among in all tested tissues. With non-treated adventitious roots as control, BcUGT6's transcriptional levels were elevated to nearly 2 folds for 2 h, 8 h, 24 h, 2 d and 4 d in MeJA-treated adventitious roots of B. chinense. It showed that the transcriptional level of BcUGT6 was slightly affected by MeJA. While, BcUGT3's transcriptional levels were gradually elevated, and till 4 d after MeJA treatment, the expression level was about 7 folds than that of non-treated control. Using pET-28a (+), the expressions of two genes was investigated. Induced by IPTG, the target proteins were expressed in E. coli and then purified. All the results obtained in the present study will be helpful for follow-up bio-function analysis of BcUGT3 and BcUGT6.

10.Genome-wide analysis of DNA 5-hydroxymethylcytosine in uremia patients using hMeDIP-chip

Objective To investigate the role of the 5-hydroxymethylcytosine (5-hmC) DNA modification in the onset of systemic lupus erythemosus (SLE),we compared tihe levels 5-hmC between SLE patients and normal controls.Methods With informed consent,whole blood was obtained from patients,and genomic DNA was extracted.Using hMeDIP-seq analysis and validation by quantitative real-time quantitative polymerase chain reaction (RT-PCR),we identified the differentially hydroxymethylated regions that were associated with SLE.Results There were 1 701 genes with significantly different 5-hmC levels at the promoter region in the SLE patients compared with the normal controls.The CpG islands of 3 826 genes showed significant difference at 5-hmC levels in SLE patients compared with the normal controls.Out of the differentially hydroxymethylated genes,three were selected for validation,including TREX1,CDKN1A,and CDKN1B.The hydroxymethylation levels of these three genes were confirmed by quantitative RT-PCR.Conclusion Our studies indicate that there are significant alterations of 5-hmC in SLE patients;these differentially hydroxymethylated genes may contribute to the pathogenesis of SLE.Such novel findings show the significance of 5-hmC as a potential biomarker or promising target for epigenetic-based SLE therapies.
