1.Expression of NANOG gene in cervical cancer and its significance

Objective To investigate the expression of NANOG gene in cervical cancer and its effect on the development of the cancer. Methods By using immunohistochemistry, NANOG expression levels were demonstrated in normal cervical tissues, cervical cancer in situ and cervical cancer tissues. Tumor sphere cells from cervical cancer tissue and their differentiated cells were injected into nude mice to observe the tumor growth. Results When compared with that of the normal cervical tissue, significantly higher NANOG protein expression level was found in cervical cancer and cervical cancer in situ (P<0.05). Tumor spheres were formed from the non-serum culture of cervical cancer tissue and the spheres could differentiate into epithelial-like cells. The sphere cells of cervical cancer caused tumor formation in nude mice. Conclusion The results suggest that cancer stem cells are present in cervical cancer and NANOG may play a crucial role in the development of cervical cancer.

4.Hot spots and co-authorship in foreign studies on pancreatic cancer radio chemotherapy in recent 5 years

Objective To collect the hot spots, development trends, high yield authors and their affiliated institutions by analyzing the papers on pancreatic cancer chemotherapy in recent 5 years. Methods The papers on pancreatic cancer chemotherapy were downloaded from PubMed. Their high frequency subject headings extracted by BICOMB were analyzed by bibliometric double clustering analysis and strategic coordinate analysis. The high yield authors were analyzed by co-authorship group analysis. Results The current studies on pancreatic cancer chemotherapy in foreign countries were focused on the pathological features of pancreatic duct adenocarcinoma before and after chemotherapy, drug therapy of pancreatic cancer according to its genetics, metabolism and pharmacology of antitumor drugs, surgical treatment combined with radiotherapy and chemotherapy for pancreatic cancer. The high yield authors could be divided into two large groups from Japan and USA and a number of small groups. Conclusion New breakthrough points should be found from the matured topics, the topics with a further developmental space should be greatly concerned, and cooperation should be strengthened between study groups or teams.
5.Absorbable bio-glass injection in osteoporosis mice:a support for the osteoporotic vertebral body and mechanism of osteogenic induction

BACKGROUND:Absorbable bio-glass injection composed by bio-glass and calcium phosphate bone cement can be tightly combined with bone tissue by a strong chemical bond to improve the stability of the bone-implant interface.
OBJECTIVE:To investigate the effect of absorbable bio-glass injection to support the vertebral body of osteoporosis mice and its mechanism of osteogenic induction.
METHODS:Osteoporosis models were prepared in 30 female Sprague-Dawley rats undergoing bilateral ovariectomy. After modeling, model rats were randomly divided into three groups, and given polymethylmethacrylate, injectable calcium phosphate bone cement and absorbable bio-glass injection into L3-5 vertebral bone defects, respectively. L3-5 segments were removed at 12 weeks after implantation to detect the biomechanical and degradation properties, levels of calcium and phosphate, alkaline phosphatase activity, bone mineral density, levels of bone morphogenetic protein 2 and transforming growth factor β, as wel as histological observation.
RESULTS AND CONCLUSION: In the absorbable bio-glass injection group, the degradation properties, compressive strength, surface hydroxyapatite deposition amount and bone mineral density were significantly higher than those in the other two groups(P< 0.05); trabecular bone relative volume, thickness and number were significant higher than those in the other two groups (P < 0.05); serum levels of calcium, alkaline phosphatase, bone morphogenetic protein 2 and transforming growth factor β were significantly higher than those in the other two groups(P < 0.05); but the level of serum phosphate was lower than that in the other two groups(P < 0.05). These results show that the absorbable bio-glass injection can enhance the support for the osteoporotic vertebral body and induce osteogenesis, probably by increasing bone morphogenetic protein 2 and transforming growth factor β levels.
6.Effect of β-adrenergic receptor stimulation on vascular smooth muscle cell apoptosis

ObjectiveTo investigate the effect of β-adrenergic receptor(β2AR) stimulation on vascular smooth muscle cell apoptosis in physiological state and receptor overexpression model.Methodsβ2AR overexpression model was established by transgenic techniques. Hoechst 33342 staining as well as flow cytometer(FCM) detected were chosen to measure the incidence of vascular smooth muscle cell apoptosis.ResultsVascular smooth muscle cell exhibited significantly fewer viable cell rate when stimulated with β2AR agonist isoproteronol for 48 hours compared with control (P<0.01),while no difference at the time point of 24 hours. Much fewer viable rate detected by FCM and high apoptotic rate by Hoechst staining were observed when VSMCs overexpressing β2AR were stimulated with isoproteronol for 24 hours (P<0.01).ConclusionStimulation of physiological and overexpressing β2AR could induce apoptosis of vascular smooth muscle cell.
8.Updated molecular genetic mechanisms of globozoospermia.

Globozoospermia is a rare and serious teratozoospermia, which is one of the important causes contributing to human male infertility. The assisted reproductive technique remains the only means for such patients to produce offspring. However, the pathogenesis of globozoospermia is not yet clear. In recent years, related studies have shown that some genes are connected with the onset of globozoospermia. This paper outlines the progress in the studies of pathogenicity genes, aiming to contribute to the molecular diagnosis and mechanism investigation of the disease.

9.Anti-SARS-CoV-2 activity of small molecule inhibitors of cathepsin L

The coronavirus disease 2019 (COVID-19) is an acute infectious disease caused by the new severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection, which has led to serious worldwide economic burden. Due to the continuous emergence of variants, vaccines and monoclonal antibodies are only partial effective against infections caused by distinct strains of SARS-CoV-2. Therefore, it is still of great importance to call for the development of broad-spectrum and effective small molecule drugs to combat both current and future outbreaks triggered by SARS-CoV-2. Cathepsin L (CatL) cleaves the spike glycoprotein (S) of SARS-CoV-2, playing an indispensable role in enhancing virus entry into host cells. Therefore CatL is one of the ideal targets for the development of pan-coronavirus inhibitor-based drugs. In this study, a CatL enzyme inhibitor screening model was established based on fluorescein labeled substrate. Two CatL inhibitors IMB 6290 and IMB 8014 with low cytotoxicity were obtained through high-throughput screening, the half inhibition concentrations (IC50) of which were 11.53 ± 0.68 and 1.56 ± 1.10 μmol·L-1, respectively. SDS-PAGE and cell-cell fusion experiments confirmed that the compounds inhibited the hydrolysis of S protein by CatL in a concentration-dependent manner. Surface plasmon resonance (SPR) detection showed that both compounds exhibited moderate binding affinity with CatL. Molecular docking revealed the binding mode between the compound and the CatL active pocket. The pseudovirus experiment further confirmed the inhibitory effects of IMB 8014 on the S protein mediated entry process.
