OBJECTIVETo observe the liver oxidant damage for diabetic model in mice.
METHODSMale kunming mice were feed with high fat dietary for a week and then were randomly divided into two groups by weight, with 10 mice in each group. One group was induced by small dose streptozotocin (STZ) and obtained STZ-induced diabetic mice, and the other group was regarded as the control. Both of the two groups were feed with high fat dietary. After 6 weeks, the activities of enzymes, such as superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GSH-PX) and nitric oxide synthase (NOS) were measured. Glutathione (GSH), maleic dialdehyde (MDA), nitric oxide (NO) levels in the liver and the liver viscera quotient were also measured. Liver histological manifestations were observed.
RESULTSIn diabetes group, there was a significant decrease in body weight, and the activities of GSH, CAT, and NOS decreased significantly (t value were 5.370, 10.639, 5.235, 3.089, respectively, P < 0.01). While, the liver viscera quotient, the levels of MDA, GSH-PX and NO increased remarkably (t value were -6.246, -2.728, -2.660, -4.924, respectively, P < 0.01 or P < 0.05). The significant difference was not observed in SOD between the two groups (t value was -0.405, P > 0.05). The liver histological damages were observed in diabetes group, light microscope observation showed hepatocytes swelling, ballooned changing and fatty droplets clustering.
CONCLUSIONThe oxidant damage might exist in the liver diabetic model in mice.

Objective To study the application value of dynamic monitoring of blood pressure in the prevention and treatment of elderly hypertension. Methods 519 hypertensive patients from December 2017 to December 2018 were monitored with ambulatory blood pressure monitoring, and were divided into the elderly group (≥60 years old, 264 cases) and the control group (<60 years old, 255 cases). The results of ambulatory blood pressure monitoring in two groups were analyzed, which inclued the circadian rhythm of blood pressure, blood pressure, pulse pressure, coefficient of variation of blood pressure, blood pressure load value, average heart rate and morning blood pressure surge. Results The incidence of abnormal circadian rhythm of ambulatory blood pressure in the elderly group was 76.5%. Compared with the control group, there were differences in the indexes of diastolic blood pressure (DBP), diastolic blood pressure load value (DBPLV), pulse pressure (PP), 24 h average heart rate (24 hAHR), systolic blood pressure coefficient of variation (SBPCV), 24 h diastolic blood pressure coefficient of variation (24 h DBPCV) and morning diastolic blood pressure surge (MDBPS) between the two groups(all P<0.05). There were differences in 24 h systolic blood pressure (24hSBP), night systolic blood pressure (nSBP), night diastolic blood pressure (nDBP), night pulse pressure (nPP), day systolic blood pressure load value (dSBPLV), ninght systolic blood pressure load value (nSBPLV), 24 h SBPCV, 24 h dDBPCV and other indicators among different blood pressure types in the elderly group (all P<0.05). Conclusion Ambulatory blood pressure monitoring indicators have important guiding value for the prevention and treatment of elderly hypertension.

Objective To propose a personalized design of anatomic ankle prosthesis that can avoid and reduce the high failure rate and risk of ankle prosthesis in clinic. Methods The 3D finite element non-linear model of normal human ankle system was established and verified. The anatomic ankle prosthesis was then designed to simulate total ankle replacement, and the 3D finite element model with both the prosthesis and ankle system was established. The biomechanical characteristics of this prosthesis were calculated and analyzed after gait loads were applied. Results For the normal ankle system, the maximum plantar contact stress was 214.6 kPa and the maximum Von Mises stress of foot bone was 8.96 MPa. The reliability of the normal ankle system model was verified by comparing the simulated results with those reported by literature. After the prosthesis implantation, the simulated maximum Von Mises stresses of talus prosthesis, tibial UHMWPE liner, tibial prosthesis were 23.88, 19.24 and 73.01 MPa, respectively. The stress of the ankle prosthesis increased drastically compared with that of normal ankle system. Conclusions The comparison results by finite element analysis examine the feasibility of the personalized ankle prosthesis, and provide references for optimization of prosthesis design and its clinical application.
