1.Research of drug screening model based on recombinant hPPAR?-LBD as a target protein for lipid metabolism

Objective:To establish a method of antitumor drug screening model based on recombinant hPPAR?-LBD as a target protein for lipid metabolism. Methods:pReceiver-B01-PPAR?- LBD expression plasmid was constructed and transferred into Escherichia coli BL2(1DE3)competent cells. Growth conditionswere optimized to induce soluble expression of hPPAR?-LBD recombinant protein. Purification was carried out by Nickel affinity chromatography. Ligand binding activity of the recombinant protein was determined by novel size exclusion chromatography and high performance liquid chromatography(SEC-HPLC)method with rosiglitazone as reference ligand and GW9662 as antagonist. Results:Under optimal conditions (16℃,0.6 mmol/L IPTG and induction time 20 h),the soluble recombinant protein was expressed successfully. After one-step purification with Nickel affinity chromatography,41 mg of recombinant protein with more than 95% purity could be obtained from per liter Luria-Bertani(LB)medium. The Kd and percentage ofspecific bindingofrosiglitazone and tetrazanbigen tohPPAR?-LBDare 625 nmol/L,65% and 1 000 nmol/L,60%,respectively.Conclusion:In this study,a fast,stable and simple method could be used successfully to screen antitumor drug.

Objective To study whether the mechanism of mifepristone in treating adenomyosis is suppressing matrix metalloproteinase-9 and tissue inhibitors of metalloproteinase-1(MMP-9/TIMP-1). Methods Thirty-five patients in the mifepristone treated group(19 cases of adenomyosis) and the control group(16 cases of adenomyosis,non-drug treated) underwent hysterectomy.Endometrium was looked as eutopic endometrium and adenomyosis as ectopic endometrium.Expression of MMP-9 and TIMP-1 in eutopic and ectopic endometrium were measured by immunohistochemical techniques. Aim To explore the characteristic of NO-cGMP signal pathway in the regulation of thoracic aortic relaxation in thyroxine-induced hypertensive rats.Methods Hyperthyroidism was induced by administering Lthyroxine(T4,0.5 mg?kg~-1,sc)daily for 16 days.Sham-treated euthyroid control rats received only vehicle saline for 16 days.SNAP,an NO donor,was used to define the differential relaxation in the thoracic aorta from euthyroid and hyperthyroid rats.To determine the mechanisms involved changes in NO-cGMP signal pathway in the regulation of aortic relaxation from hyperthyroid rats,BAY 41-2272(BAY)was used to activate soluble guanylate cyclase(sGC),ODQ was used to inhibit sGC,and 8-Br-cGMP was used to acti vate protein kinase G(PKG),respectively.Results Thyroid hormone excess for 16 days showed characteristic changes in body weight,heart rate and systolic blood pressure in rats.The body weight was significantly decreased,while heart rate,pulse pressure and systolic blood pressure were increased in T4-treated rats.SNAP caused relaxation in the aorta in both euthyroid and hyperthyroid rats.However,SNAP-induced aortic relaxation was significantly attennuated in hyperthyroid rats than in euthyroid rats.In the presence of ODQ,SNAP-induced aortic relaxation was blocked in both euthyroid and hyperthyroid rats.BAY and 8-BrcGMP-induced aortic relaxation was significantly attennuated in hyperthyroid rats than in euthyroid rats.Conclusion These data suggest that the attenuated effect of NO-cGMP signal pathway is involved in the regulation of aortic relaxation in the pathophysiology of hyperthyroidism,which may be related to the downregulation of sGC and PKG functions.

7.Analysis of Doppler flow spectra of internal jugular vein in healthy adults

Objective To analyze the flow spectra of in ternal jugular vein (IJV) in healthy adults by pulsed Doppler ultrasonography. Methods IJV flow velocities in 120 healthy volunteers were recorded during cardiac and respiratory cycles respectively with the transducer placed in cervical region view. Results In healthy subjects, IJV waveforms were composed of one reverse wave A and two forward waves S and D during cardiac cycle. The flow velocities of wave S [( 27.17 ? 19.88 )cm/s vs ( 19.04 ? 14.26 )cm/s] and wave D [( 17.43 ? 12.78 )cm/s vs ( 12.70 ? 10.57 )cm/s] were quicker during inspiration than those during expiration (P 0.05 ]. 8.Study on regular patterns of superior vena cava Doppler flow spectra in patients with superior vena cava syndrome

Objective To consecutively investigate the changes of the superior vena cava flow Doppler patterns in patients with superior vena cava syndrome (SVCS) and predict SVCS development.Methods Twenty-two definative cases with SVCS due to tumor were enrolled in this study.At different stages of therapy the superior vena cava was detected by ultrasonography.The superior vena cava Doppler spectra during cardiac cycle and respiratory cycle were analyzed respectively.Results In 22 patients with SVCS the superior vena cava Doppler flow patterns presented a developing tendency,from turbulent,continuous and high velocity pattern to normalization.The S and D waves peak velocities were ( 154.78 ?52.15 ) cm/s,( 159.38 ? 46.56 ) cm/s respectively before therapy,while after therapy ( 58.78 ? 33.43 ) cm/s,( 34.96 ? 17.56 ) cm/s respectively.The S wave and D wave respective differences in respiratory cycle were ( 2.14 ? 2.08 ) cm/s,(2.73?2.68)cm/s before therapy,while those were ( 7.68 ? 6.22 ) cm/s,( 6.32 ? 4.98 ) cm/s after therapy.Conclusions The regular changes of the superior vena cava Doppler flow patterns in tumor-induced SVCS development course were embodied in dynamic diversity.These changes could suggest SVCS development and evaluate the therapy effect on SVCS.
9.Ultrasonographic observations on proliferative activity and invasiveness of rabbit renal VX_2 tumor

Objective To explore the correlation between the ultrasonographic parameters describing the tumor and proliferative activity, invasiveness of the rabbit renal VX 2 tumor. Methods Animal models were established in 56 rabbits,in which left kidneys were transplanted with VX 2 tumor. The second harmonic imaging and video-density quantitative determination were applied to observe the size, echo-intensity and edge echo-pattern of the rabbit renal VX 2 tumor. The findings were compared with those of histopathologic section and DNA analysis calculated with the flow cytometer. Results The number of tumor cell with higher proliferative index(PI) and ploidy cell rate(PCR) increased with the tumor size. The tumors of hyperecho and more blood flow with obvious necrosis tissue, fibrotic lesion and blood sinusoid got higher PI and PCR than the tumors of hypoecho and less blood flow, while there was no statistic significant correlation between the echoic patterns of the tumor edge and DNA analysis.Conclusions The ultrasonographic parameters describing the tumor, such as the size, echo-intensity and blood flow, can be used to evaluate the proliferative activity and invasiveness of the renal tumor.

The growth and invasion of ectopic endometrium in endometriosis depends on hormone which will work in combination with its receptors.This review introduces the construction,function,gene,subtypes of estrogen receptor and progesterone receptor and their expressions in eutopic and ectopic endometrium,and explores their significance in the genesis,development,treatment and prognosis of endometriosis.
